        Effect of Gypsum Fibrosum on Interleukin-4 Secretion of Mice Splenocytes

        석고는 황산염류 광물로 주로 함수황산칼슘(CaSO_4 · 2H_2O)으로 되어 있으며, 解肌淸熱, 除煩止渴 등의 효능이 있다. Interleukin(IL)-4는 T helper 2 세포에서 생산되며 사람의 면역작용과 관련이 있어 과량 분비되면 면역계에 과민반응을 일으키는 것으로 알려져 있다. 목적 : 석고가 면역계에 작용하는 기전 중 마우스 비장세포에서 IL-4 분비저하에 대한 영향을 밝히기 위해 본 실험이 구상되었다. 방법 : 마우스에서 비장세포를 적출하여 배양 한 다음, concanabalin(Con) A를 단독 처치 혹은 석고 포화용액을 희석 비율대로 전처치하고 Con A를 처리한 후 분비된 IL-4의 양을 측정하였다. 결과 : 실험결과 Con A를 처리한 대조군에서 IL-4의 분비량은 44.2 ± 2.3 pg/ml이었고, 증류수에 포화시킨 석고 용액을 10000배, 1000배, 100배로 희석하여 각각 전처리 한 경우에 IL-4의 양이 각각 26.8 ± 6.7 pg/ml , 12.9 ± 6.8 pg/ml, 2.8 ± 5.7 pg/ml이었고, 이 중 1000배와 100배로 희석하여 처치한 군에서 통계적 유의성을 나타내었다. 결론 : 석고는 용량 의존적으로 마우스 비장 세포의 IL-4 분비를 감소시켰다. 특히 석고 포화 용액을 1000배와 100배로 희석하여 처치한 경우 모두에서 유의한 감소를 나타내었다. 본 연구결과는 석고의 면역 조절작용에 대한 실험적 근거를 제시한다.

        Effect of Cibotii Rhizoma on the Growth Hormone Secretion of Adolescent Male Rats

        뇌하수체전엽에서 분비된 성장호르몬의 작용으로 성장이 촉진된다는 것이 지금까지의 정설이고, 최근 그 외 부갑상선 호르몬, 성호르몬, 국소호르몬 등이 관련되어 있다는 연구보고가 있다. 목적 : 성장촉진 효능이 있는 구척추출물의 작용기전 중 성장호르몬 분비에 대한 영향을 밝히기 위해 본 실험이 구상되었다. 방법 : 성장기 흰쥐에게 구척추출물을 투여하고 성장촉진 효과를 나타낸 농도에서, 투여시간에 따른 혈중 성장호르몬의 변화를 측정하였다. 결과 : 구척 투여전 20분에서 투여후 60분까지 총 80분간의 혈중 성장호르몬 변화를 측정한 결과, 투여후 5분에서 혈중 성장호르몬이 약간 증가하는 경향을 나타내었다. 5분후의 혈중 성장호르몬의 양을 구척추출물 10 mg/kg과 100 mg/kg의 농도별로 투여한 후 측정하였으나 성장호르몬의 증가를 관찰할 수 없었다. 결론 : 구척은 유의한 성장촉진작용을 나타내지만, 혈중 성장호르몬의 양을 증가시키지는 않았다. 따라서 구척의 성장촉진 효능에 대한 작용기전에 성장호르몬 분비촉진기전은 없는 것으로 사료된다.

        Anti-inflammatory Effects of Co-treatment of Jingyoganghwaltang and Cheongsimhwan on Croton Oil Induced Hemorrhoid Model in Rats

        Objectives : Hemorrhoids are one of the most common diseases in humans. Jingyoganghwaltang (JG) and Cheongsimhwan (CS) have been used for treating hemorrhoids in Korean traditional clinical practice. The present study was designed to evaluate the traditional effects of JG and CS on the experimental hemorrhoid model in rats. Methods : Hemorrhoids are closely related to inflammation. Accordingly, we examined the nitric oxide (NO) production in macrophage cell line in order to evaluate the anti-inflammatory effect. The expression levels of inflammation related genes including IL-1 beta, IL-6, INOS, and TNF-alpha were examined via a real-time quantitative PCR. Croton oil-induced hemorrhagic animal model was used to test the in vivo efficacy against hemorrhoids. The rectal tissues were weighed and the inflammatory proteins were measured to confirm the anti-inflammatory effects. Results : JG and CS have a statistically significant effect on inhibition of NO production and on the reduction of inflammatory gene expression such as IL-1 beta, IL-6, INOS, and TNF-alpha. The synergistic effects of co-treatment of JG and CS were found out in the IL-6 gene expression. The in vivo study using croton oil-induced hemorrhoid model in rat was performed to check the co-treatment effects. As a result, the co-treatment reduced the inflammation of the rectal tissue and decrease the inflammation related protein productions including ICAM1, MMP2 and MMP9. Conclusions : These results suggest that JG and CS co-treatment demonstrated anti-inflammatory effects in croton oil-induced hemorrhoid model in rat.

        Anti-inflammatory Effect of Arbitrary Waveform Generator Treatment in Rats

        Inflammation is the complex biological response of injured tissues to harmful stimuli. A cascade of biochemical events propagates and matures the inflammatory response, involving the local vascular system, the immune system, and various cells within the injured tissue. The immune system is often involved with inflammatory disorders, demonstrated in both allergic reactions and some myopathies, with many immune system disorders resulting in abnormal inflammation. An Arbitrary Waveform Generator (AWG) is a piece of electronic test equipment used to generate electrical waveforms for the treatment of patients. The patients with gastritis and arthritis have been known to have a relatively favorable prognosis with AWG treatment. Accordingly, we examined the effects of AWG treatment in gastritis and arthritis animal model. The compound 48/80 was used to induce animal gastritis model. The tissue malone dialdehyde (MDA) and serum histamine levels, and the activity of superoxide dismutase (SOD) in stomach tissue were measured. The tissue MDA and serum histamine levels in AWG treated groups exhibited the decreased tendency compared with control group, whereas the tissue SOD activity was slightly increased. The Freund's complete adjuvant was used to induce animal arthritis model as well. The paw edema volume and the width of ankle joint were determined. The AWG treatment significantly decreased the paw edema volume after 5th day of treatment. Although further studies should be performed to confirm the effects of AWG treatment, present study suggest that AWG treatment might be used as a complementary treatment for the gastritis or arthritis treatment.

        Identification of Akebia and Aristolochia species by Pyrosequencing

        Objective : Akebia species and Aristolochia species are some of the most medicinally important genera in the Oriental medicine. This study was performed to determine if Akebia species and Aristolochia species could be identified by genetic analysis and to verify Pyrosequencing analyses, which was used to assess genetic variation. Method : The DNAs of Akebia quinata Decaisne and Aristolochia manshuriensis Kom. were extracted, and we have investigated the typing of single-base variations of Akebia species and Aristolochia species in DNA by using Pyrosequencing. Result : Akebia quinata Decaisne showed different pattern compared with Aristolochia manshuriensis Kom.. The peak of Akebia quinata Decaisne was very week in the second A nucleotide base but The peak of Aristolochia manshuriensis Kom. was very high. From these results we verified that our Akebia species and Aristolochia species-specific sequencing primers were well designed. Conclusion : Pyrosequencing analysis might be able to provide the identification of the Akebia species and Aristolochia species.

      • Decrease of the electroacupuncture-induced analgesic effects in nuclear factor-kappa B1 knockout mice

        To investgate the involvement of ruclear factor kappa B1 (NF-kB1; p50/p105) in electroacupuncture (EA)-induced analgesia, 2 and 100 Hz EA stimulations were applied at acupoint ST36 (Zusanil) in NF-kBl knockout mice. EA was performed for 30 min and tail-flick latencies (TFLs) were evaluated every 15 min for 1h. Wild-type mice displayed a 63.3% increase in TFLs compared to baseline after 2 Hz EA, whereas NF-kBl + /- mice exhibited a 41.8% increase and NF-kBl-/- mice showed only a 3.9% increase of TFLs. The TFLs of 100 Hz EA showed similar trends: a 72.6% increase of TFLs in wild-type, a 38.6% increase in NF-kBl +/- and a 9.3% increase in NF-kBl -/- mice. The present findings suggest that NF-kBl may play a crucial role in both low and high frequency EA-induced analgesic effects. ⓒ 2002 Elsevier Science lreland Ltd. All rights reserved.
