Brazilin as a new sunless tanning agent

To develop an active material for skin darkening, we examined the effect of 300 plants on tyrosinase activity and found only Caesalpinia sappan has an ability to increase tyrosinase activity highly and melanin contents in B-16 melanoma cells. A compound increasing tyrosinase activity and melanin production was isolated from Caesalpinia sappan Lignum. Brazilin was identified as a new active agent. Brazilin increases the tyrosinase activity and malanin production of B-16 melanoma cells. In conclusion, it seems that brazilin can be used as a new sunless tanning agent.
Inhibitory effects of Prunus persica flower extracts on UV-induced skin damage

For an attempt to develop safe materials protecting UV-induced skin damage, plant extracts were evaluated for their antioxidative and free radical scavenging activities. From the results of these screening procedures, the ethanol extract of the flowers of Prunus persica was selected for further study. It was found that Prunus persica (50-200 μg/ml) inhibited UVB-induced DNA damage measured by tail moment in the Single Cell Gel Electrophoresis(COMET assay) and inhibited UV-induced lipid peroxidation, especially against UVB-induced peroxidation at higher than 10 μg/ml. Also P. persica(100~1,000μg /㎖) inhibited the amount of <sup>14</sup>C-arachidonic acid metabolites release from UVB- irradiated keratinocytes and it possessed the protective activity against UV-induced cytotoxicity of keratinocytes. All these results indicate that the flowers of P. persica extract may be beneficial for protection UV-induced skin damage when topically applied.

open button Abstract In the aged skin especially in the face and eyelid, deep and slight wrinkles are one of the remarkable phenomena of aging and the cause of wrinkle is various. Among the cause of wrinkles an oxidative stress plays an important roles in wrinkle formation process. It caused the lipid peroxidation of cell membrane, the increase of the MMPs(MatrixMetalloProteinase) gene expression and cellular DNA damage. These ROS induced materials may cause the degradation of collagen matrix system in the dermis and cause the formation of skin wrinkle. So, it is very important for protecting skin wrinkle formation to regulate ROS activity. In this study, we developed one active ingredient having multi functional activities such as activation of collagen synthesis, inhibition of MMPs activity, lipid peroxidation and free radical scavenging activity and inhibition of free radical induced DNA damage in vitro. Pericarpium castaneae extracts showed collagen synthesis increase in Normal Human Fibroblast and the inhibition of elastase activity (IC<sub>50</sub>/ of Elastase: 43.9㎍/㎖). It showed also anti-oxidative activity (IC<sub>50</sub>/ : 48㎍/㎖) and free radical scavenging activity(IC<sub>50</sub>/: 7.6㎍/㎖). Conclusively, Pericarpium castaneae extracts may be used as an ingredient for new anti-wrinkle cosmetics.
