<P> <I>Background.</I> Pueraria <I>lobata</I> is used in traditional Asian medicine to treat cardiovascular diseases, diarrhea, diabetes mellitus, and diabetic complications such as diabetic retinopathy. Oxidative stress in retinal pigment epithelial cells is implicated in the pathogenesis of retinopathy and age-related macular degeneration (AMD). Here, we evaluated whether the <I>P. lobata</I> extract can prevent cell death and decrease membrane permeability in oxidative stress-induced human retinal pigment epithelial cells. <I>Methods.</I> The effects of <I>P. lobata</I> extract on hydrogen peroxide- (H<SUB>2</SUB>O<SUB>2-</SUB>) induced oxidative stress were investigated using 2′,7′-dichlorofluorescin diacetate, western blotting, and immunohistochemistry in human retinal pigment epithelial cells. The effects of puerarin, daidzein, and daidzin isolated from <I>P. lobata</I> extract were also studied by determining cell death, reactive oxygen species (ROS) generation, and p38 mitogen-activated protein kinase (MAPK) and c-Jun N-terminal kinase (JNK) phosphorylation. <I>Results.</I> Our results showed that the <I>P. lobata</I> extract inhibited ROS generation, suppressed the disruption of zonula occludens-1 (ZO-1), and reduced membrane permeability in H<SUB>2</SUB>O<SUB>2-</SUB>induced human retinal pigment epithelial cells. Additionally, the <I>P. lobata</I> extract prevented the inhibition of p38 MAPK and JNK phosphorylation. <I>Conclusion.</I> Our findings suggest that the <I>P. lobata</I> extract has the potential to prevent AMD development by inhibiting the mechanism underlying oxidative stress-mediated ocular disorders. </P>
ISAG-recommended Microsatellite Marker Analysis Among Five Korean Native Chicken Lines

The objective of this study was to determine genetic variation of five Korean native chicken lines using 30 microsatellite(MS) markers, which were previously recommended by ISAG(International Society for Animal Genetics). The initial study indicated that two microsatellite markers, MCW0284 and LEI0192, were not amplified in these lines and excluded for further analysis. Twenty eight microsatellite markers were investigated in 83 birds from five Korean native chicken lines. The identified mean number of alleles was 4.57. Also, the expected, observed heterozygosity(He, Ho) and polymorphism information content(PIC) values were estimated in these markers and they ranged from 0.31~0.868, 0.145~0.699, and 0.268~0.847, respectively. The results were used for the discrimination of five chicken lines using genetic distance values and also neighbor-joining phylogenetic tree was constructed. Based on the He and PIC values, eighteen markers are enough for the discrimination of these Korean native chicken lines for the expected probability of identity values among genotypes of random individuals(PI), random half sibs(PIhalf-sibs) and random sibs(PIsibs). Taken together, these results will help the decision of conservation strategies and establishment of traceability system in this native chicken breed. Also, the use of ISAG-recommended microsatellite markers may indicate that the global comparison with other chicken breeds is possible.

Objectives: This study was designed to examine the anti-cancer activity by innate immunomodulating and anti-inflammatory effects of liriopis tuber water extract (LPE). Methods: Cell cytotoxicity was tested with 4T1 mouse mammary carcinoma cells, spleen cells, macrophage, and RAW264.7 cells. To investigate innate immunomodulating effects of LPE on macrophage, we measured tumor necrosis factor-alpha (TNF-α), interleukin-12 (IL-12), and interleukin-10 (IL-10). To investigate innate immunomodulating effects of LPE on RAW264.7 cell, we measured TNF-α, interleukin-6 (IL-6). In addition, TNF-α and nitric oxide (NO) induced by lipopolysaccharide (LPS) were measured after treating with LPE to observe innate immunomodulating effect of LPE on RAW264.7 cell. Also, mitogen-activated protein kinase (MAPK) and nuclear factor κB (NF-κB) were examined by western blot analysis. Results: In an in vitro cytotoxicity analysis, LPE affected tumor cell growth above specific concentration. As compared with the control group, the production of TNF-α, IL-12, and IL-10 were increased in macrophage. As compared with the control group, the production of TNF-α and IL-6 were increased in RAW 264.7 cell. The expression of TNF-α and NO induced by LPS after treating LPE was decreased. In addition, treatment of RAW 264.7 cell with LPE increased the phosphorylation levels of p-extracellular signal-regulated kinase (p-ERK), p-Jun N-terminal kinase (p-JNK), and p-p38. Conclusions: LPE might have impact on the anti-cancer effect by activation of innate immune system and inflammation control.

This study, using the case of Sejong Cityexamines the meaning of the discussion on the archive construction of the Korean Local Culture Digital Dictionary, which is operated by the Central Research Institute of Korean Studies. Let us take a look at the entire compilation process of the Digital Sejong City Culture Dictionary, which began in 2018 and was released in 2020. The final result is a text of about 8,000 pages of manuscripts, a list of archive construction, and Sejong city local culture audiovisual data. Sejong City’s local culture audiovisual data consisted of about 1,000 photos, 100 diagrams and drawings, and 30 videos and sounds. In particular, it is of great significance to organize the history of the Sejong region as a cultural record and description. First, the material restores and reproduces the gradually disappearing local culture. Second, the foundation for regional cultural history research was laid through a digital archive built as a comprehensive management system for data. Third, the Digital Sejong City Cultural Exhibition has a circular knowledge information management system. The results of the project will also be of value as learning materials for consumers in other regions who require information about the new Sejong City, especially students living in Sejong City. In addition, so that the digital archive construction of the Korean Local Culture Digital Dictionary can be more valuable than it is now, I have suggested measures for improving some practical problems.

Background: Korean native chicken (KNC) is a well-known breed due to its superior meat taste. This breed, however, owing to a low growth rate, has a high market price. In order to overcome this disadvantage, the National Institute of Animal Science (NIAS) in Korea developed a commercial KNC breed, named Woorimatdag version 2 (WM2), an upgraded version of the Woorimatdag (WM1) breed and the WM2 was created by crossing the KNC with meat type breeds. This study aims to discriminate between WM2 and other chicken breeds using microsatellite (MS) markers. Methods: A total of 302 individuals from eight Korean chicken populations were examined. The genetic diversity and population structure analysis were investigated using Cervus, API-CALC, STRUCTURE, PowerMarker programs. Results: Based on heterozygosity and polymorphic information content (PIC) values, 30 MS markers were initially selected from 150 markers. The identified average number of alleles (Na), expected heterozygosity, and PIC values for the WM2 samples were 7.17, 0.741, and 0.682, respectively. Additionally, the paternity of individuals was assigned with a success rate of greater than 99% using 12 markers, the best minimum number of markers. The 12 selected markers contained heterozygosity and PIC values above 0.7 and probability of identity values around zero. Using these markers, the determined probability of identity (PI), PIhalf-sibs, and PIsibs values were 3.23E-33, 5.03E-22, and 8.61E-08, respectively. Conclusions: WM2 is well differentiated with respect to other chicken breeds based on estimated genetic distances. The results presented here will contribute to the identification of commercial WM2 chicken in the market.

Objective: The purpose of this study was to examine the effects of hip joint exercise using an elastic band on dynamic balance, agility, and flexibility in healthy subjects. Design: Randomized controlled trial. Methods: Thirty-five subjects (between 19 and 23 years) were randomly allocated to two groups: hip flexion exercise (HFE) group (n=17) and the hip abduction exercise (HAE) group (n=18). The HFE group participated in flexion exercise of the hip joint using an elastic band for 50 minutes a day, three days a week for four weeks, while the HAE group participated in abduction exercises of the hip joint using an elastic band for the same period. Dynamic balance was measured using the timed up and go (TUG) test, agility was measured with the standing long jump, and flexibility was measured using the Schober's test (5 cm, 10 cm). Results: The HFE group showed significant differences in the TUG test, standing long jump, and the Schober's test (10 cm) after training (p<0.05). The HAE group showed significant differences in the TUG test, standing long jump and the Schober's test (5 cm, 10 cm) after training (p<0.05). However, there was no significant difference between the HFE group and the HAE group. Conclusions: Flexion and abduction exercises of the hip joint using and elastic band increased dynamic balance, agility, and flexibility in healthy subjects. Additional research on hip joint exercises using an elastic band for improving dynamic balance, agility and flexibility are necessary.
