        Optimal Isosceles Trapezoid Cross Section of Laterally Loaded Piles based on Friction Soil Arching

        The paper presents a novel analytical solution to determine an optimal isosceles trapezoid cross-section of stabilizing piles subjected to the lateral load. In view of the pile-soil interaction mechanical analysis on the friction soil arching between the adjacent stabilizing piles, a general mechanical model for analyzing the behavior of the laterally loaded stabilizing piles with isosceles trapezoid cross-section is put forward. The reasonable net pile spacing expression for all kinds of isosceles trapezoid section piles is established via the analytical deduction, and it indicates that there is a positive relationship between the reasonable net pile spacing and gradient of pile sidewall. In consideration of the strength failure possibility in the apex of friction soil arching, the complete expressions of both the gradient of pile sidewall and corresponding reasonable net pile spacing of stabilizing piles can be established. The case study of Jinle landslide verifies that the reasonable pile spacing obtained by the presented model is slightly greater than that of the conventional design scheme, which shows that the stabilizing piles with optimal isosceles trapezoid cross-section can results in reducing the investment of the whole stabilizing piles.

        Transcriptomic Features of Echinococcus granulosus Protoscolex during the Encystation Process

        Cystic echinococcosis (CE) is a zoonotic infection caused by Echinococcus granulosus larvae. It seriously af- fects the development of animal husbandry and endangers human health. Due to a poor understanding of the cystic fluid formation pathway, there is currently a lack of innovative methods for the prevention and treatment of CE. In this study, the protoscoleces (PSCs) in the encystation process were analyzed by high-throughput RNA sequencing. A total of 32,401 transcripts and 14,903 cDNAs revealed numbers of new genes and transcripts, stage-specific genes, and differ- ently expressed genes. Genes encoding proteins involved in signaling pathways, such as putative G-protein coupled re- ceptor, tyrosine kinases, and serine/threonine protein kinase, were predominantly up-regulated during the encystation process. Antioxidant enzymes included cytochrome c oxidase, thioredoxin glutathione, and glutathione peroxidase were a high expression level. Intriguingly, KEGG enrichment suggested that differentially up-regulated genes involved in the va- sopressin-regulated water reabsorption metabolic pathway may play important roles in the transport of proteins, carbo- hydrates, and other substances. These results provide valuable information on the mechanism of cystic fluid production during the encystation process, and provide a basis for further studies on the molecular mechanisms of growth and de- velopment of PSCs.

        Low Potential Amperometric Determination of Ascorbic Acid at a Single-Wall Carbon Nanotubes-Dihexadecyl Hydrogen Phosphate Composite Film Modified Electrode

        A sensitive and selective electrochemical method was developed for the amperometric determination of ascorbic acid (AA) at a glassy carbon electrode (GCE) modified with single-wall carbon nanotubes-dihexadecyl hydrogen phosphate (SWNT-DHP) composite film. The SWNT-DHP composite film modified GCE was characterized with SEM. The SWNT-DHP composite film modified GCE exhibited excellent electrocatalytic behaviors toward the oxidation of AA. Compared with the bare GCE, the oxidation current of AA increased greatly and the oxidation peak potential of AA shifted negatively to about –0.018 V (vs. SCE) at the SWNT-DHP composite film modified GCE. The experimental parameters, which influence the oxidation current of AA, were optimized. Under the optimal conditions, the amperometric measurements were performed at a applied potential of 0.015 V and a linear response of AA was obtained in the range from 4 × 107 to 1 × 104 mol L1 and with a limit of detect (LOD) of 1.5 × 107 mol L1. The interferences study showed that the SWNT-DHP composite film modified GCE exhibited good sensitivity and excellent selectivity in the presence of high concentration uric acid and dopamine. The proposed procedure was successfully applied to detect AA in human urine samples with satisfactory results.

        Nonlinear Optimization for Geometric Parameters of Reinforced Concrete Coupled Structural Walls

        The coupling ratio (CR) has been widely used to quantitatively reflect the degree of coupling action between the component walls and link beams of a coupled wall. However, the CR index is very difficult to evaluate at early design stages. In order to develop alternative design parameters that can be easily utilized and ensure the dual lateral force resisting mechanism of coupled walls, nine prototype coupled walls were designed considering various aspect ratios of wall pier (αw) and span-to-depth ratios of coupling beams (αb). The incremental dynamic analyses on the seismic fragility was conducted to examine the influences of αw and αb on the fragility characteristics and the margin capacities corresponding to different performance limit states. The double-factorial quadratic response surface method was used to further reveal the interactive impact of αw and αb on the overall behavior of coupled walls. Then the combinations of αw and αb that can produce the optimal values of response variables were obtained. The curve-fitting technique was used to find the relationship between optimal αb and αw. It is indicated that αb and αw can effectively serve as the alternative parameters to CR and ensure optimal overall structural performance of RC coupled walls.

        Baicalein facilitates gastric cancer cell apoptosis by triggering endoplasmic reticulum stress via repression of the PI3K/AKT pathway

        Objective Gastric cancer (GC) remains a prevailing threat to life. Baicalein exhibits anti-cancer properties. This study estimated the mechanism of baicalein in GC cell apoptosis by mediating endoplasmic reticulum stress (ERS) through the PI3K/AKT pathway. Methods After treatment with different concentrations of baicalein, GC cell (HGC-27 and AGS) viability was detected by MTT assay. AGS cells more sensitive to baicalein treatment were selected as study subjects. The IC50 of baicalein on AGS cells was determined. Colony formation, cell cycle, and apoptosis were detected using crystal violet staining and flow cytometry. Levels of ERS-related and BTG3/PI3K/AKT pathway-related proteins were determined by Western blot. Intracellular Ca2+ level was measured using Fluo-3 AM fluorescence working solution. GC mouse models were established by subcutaneously injecting AGS cells into the right rib and were intragastrically administrated with baicalein. Tumor volume and weight were recorded. Expression of Ki67 in tumor tissues and positive expression of apoptotic cells were detected by immunohistochemistry and TUNEL staining. Results Baicalein inhibited cell proliferation and induced G0/G1 arrest and apoptosis by regulating the cell cycle, and triggered ERS in GC cells. Baicalein impeded the PI3K/AKT pathway by activating BTG3, thereby triggering ERS and inducing apoptosis. BTG3 inhibition reversed baicalein-induced apoptosis and ERS. Baicalein regulated GC cells in a concentration-dependent manner. Moreover, in xenograft mice, baicalein prevented tumor growth, decreased Ki67- positive cells, activated BTG3, and inhibited the PI3K/AKT pathway, thus activating ERS and increasing apoptotic cells. Conclusion Baicalein facilitates GC cell apoptosis by triggering ERS via repression of the PI3K/AKT pathway.

      • Scalar dark matter interpretation of the DAMPE data with U(1) gauge interactions

        <P>Recently, the Dark Matter Particle Explorer (DAMPE) experiment released the new measurement of the total cosmic e(+) e(-) flux between 25 GeV and 4.6 TeV, which indicates a spectral softening at around 0.9 TeV and a tentative peak at around 1.4 TeV. We utilize a scalar dark matter (DM) model to explain the DAMPE peak by XX -> Z'Z' -> l (l) over barl'(l) over bar 'l with an additional anomaly-free gauged U (l) family symmetry, in which X, Z'and l((')) denote, respectively, the scalar DM, the new gauge boson, and l((')) = e, mu, tau, with m(x) similar to m(z)' similar to 2 x 1.5 (TeV). We first illustrate that the minimal framework Gsm X U(l)(Y') with the above mass choices can explain the DAMPE excess, which, however, be excluded by LHC constraints from the Z' searches. Then, we study a nonminimal framework G(SM) X U(l)(Y') x U(i)(Y') in which U(l)(Y') mixes with U(l)(Y'). We show that such a framework can interpret the DAMPE data and at the same time survive all other constraints including the DM relic abundance, DM direct detection, and collider bounds. We also investigate the predicted e(+) e(-) spectrum in this framework and find that the mass splitting Delta m = m(x) - m(z') should be less than about 17 GeV to produce the peaklike structure.</P>
