Preventive and Antioxidative Effects of Crude Drug Preparation(E-kong-san) on Cisplatin Induced Nephrotoxicity. Proceedings of International Symposium on East-West Medicine, Seoul. 201-211, 1999. -Nephroprotective effects of a crude drug-prearation (E-kong-san) were determined from cisplatin on rabbit kidney proximal tubule and human renal cortical cells by MTT assays and sustained glucose consumption on ciplatin-induced human renal cortical tissue. Levels of creatinine and blood ure nitrogen(BUN) in serum after administration of cisplatin(0.75mg/kg,i.p.) to E-kong-san(0.75g/kg/day,p.o.)-pretreated rats were markedly lower compared to those of cisplatin-treated rats. Moreover, the administration of E-kong-san significantly inhibited the loss of body weight of cisplatin injected rts. These findings suggest that E-kong-san on the cisplatin induced nephrotoxicity led us to investigate whether the effect of this water extract was a result of anti-oxidation. E-kong-san showed strong free radical scavenger activities on 1,1-dipheny1-2picrylhydrazil(DPPH) radical and xanthine/xanthine oxidase(XOD) generated superoxide anion radical(O2-). We further studied the effects of E-kong-san on lipid peroxidation in rat liver microsomes induced by enzymatic and nonenzymatic methods. E-kong-san exhibited significant inhibition on both ascorbic acid/Fe2+and ADP/NADPH/Fe3+ induced lipid peroxidation in rat liver microsomes. Based on these results, we suggest that-E-kong-san attenuate the cisplatin induced cytotoxicity and its mechanism can be eplained by antioxidant.

        Single Oral Dose Toxicity Evaluation of Leejung-tang, a Korean Traditional Herbal Formula, in Crl:CD (SD) rats

        Objective: Leejung-tang (Rechu-to in Japanese) is a traditional Korean herbal formula used for treatment of gastrointestinal disorders such as vomiting, stomach pain, chronic gastritis and gastrointestinal ulceration. The present study was carried out to investigate the potential acute toxicity of Leejung-tang water extract (LJT) by a single oral dose in Crl:CD (SD) rats in compliance with current guidelines. Methods: In the preliminary study, there were no adverse effects such as death, clinical signs, and body weight changes at dose levels of 500, 1000, and 2000 mg/kg/day body weight. Based on the results, a dose of 2000 mg/kg was selected as the toxicological limited dose. LJT was administered once by gavage to male and female rats at dose levels of 0 and 2000 mg/kg bodyweight. During the study period, mortalities, clinical findings, and body weight changes were observed for 14 days following the administration. On day 14 after the treatment, the animals were sacrificed by carbon dioxide overdose and complete gross postmortem examinations were performed. Results: In present study, no treatment-related deaths were observed. There were no adverse effects on clinical signs and body weight changes. In addition, there were no observed gross findings in all groups except for a kidney cyst in the 2000 mg/kg/day female group. Conclusion: The results indicated that LJT did not induce toxic effects at a dose level up to 2000 mg/kg in rats and its median lethal dose ($LD_{50}$) was considered to be over 2000 mg/kg/day body weight for both genders.

        다양한 골 이식재의 임상 효과에 대한 고찰

        Purpose: Various bone graft materials are being used for periodontal tissue regeneration. Th materials are being developed continuously for ideal clinical effects. Therefore, it is necessary to identify the clinical characteristics of each bone graft material through comparing the various bone graft materials statistically and in doing so, proposing a more efficient bone graft material. In this study, the following results were attained through comparing the clinical effects among the bone graft materials, using the statistical method based on the clinical studies published at the department of periodontology of Yonsei hospital. Materials and Method: 6 selected studies of department of Periodontology at Yonsei University Hospital were based on clinical study of bone grafting in intrabony defects. It was compared the clinical parameters among the 6 clinical studies, using the statistical META analysis. Result: When comparing the probing depth reduction, there was a relatively great amount of decease when using the xenograft, Anorganic Bovine Derived Hydroxapatite Bone Matrix/Cell Binding Peptide(ABM/P-15: PepGen $P-15^{(R)}$) and the autogenous bone and absorbable membrane, d, 1-alctide/glycolide copolymer(GC: $Biomesh^{(R)}$). The allogfrafts showed a relatively low decrease in the probing depth and clinical attachment change. It also showed a slight decrease in the bone probing depth. The allografts showed various results according to different bone graft materials. When comparing the ABM/P-15 and bovine bone $powder(BBP^{(R)})$, ABM/P-15 showed a relatively high clinical attachment level and the bovine bone powder showed a relatively high clinical attachment level. The probing depth change and gingival recession change showed a lower value than the mean value between the two bone graft materials. The synthetic bone showed a relatively high decrease in clinical attachment level and periodontal probing depth change. There was a relatively larger amount of gingival recession when using Bioactive Glass(BG) but a relatively low bone regeneration effect was seen. Conclusion: Good restorative results of the periodontal tissue can be attained by applying the various bone graft materials being used today after identifying the accurate clinical effects.

        랫드에서 쌍화탕의 급성독성에 관한 연구

        Objectives: This study was conducted to evaluate the acute toxicity and safety of Ssanghwa-tang (Shuanhetang in Chinese, Sou-wa-to in Japanese) in Crl: CD Sprague-Dawley (SD) rat though the current regulatory guideline. Methods: In this study, 10 rats of each sex were randomly assigned to two groups of 5 rats each and were administrated singly by gavage at dose levels of 0 and 2000 mg/kg/day of ssanghwa-tang water extract (SHT). After single administration of SHT, mortalities, clinical signs, body weight changes, gross findings were observed for the 15-day period. Results: Acute toxicity tests revealed that a single oral administration of SHT at dose levels of 2000 mg/kg did not affect clinical signs, body weight, and gross findings, evaluating the safety of SHT. The SHT treatment did not result in any toxicologically significant changes in mortality, clinical signs, body weight changes. Conclusions: These results showed that the single oral administration of SHT did not cause any toxic effect at the dose levels of 2000 mg/kg/day in rats. In conclusion, the median lethal dose (LD50) of SHT was considered to be over 2000 mg/kg/day body for both sexes.

      • 이공산(異功散)의 in vitro에서 Cisplatin 유도 신장독성의 감소 효과

        In vitro model systems are increasingly used to investigate the mechanism of chemical-induced toxicity and for toxicological screening of new drug families. The effect of Ekongsan on the cytotoxicity induced by cisplatin on human and rabbit kidney cells was evaluated by tetrazolium-dye(MTT) assay. On the basis of Cl_50 value, cisplatin-induced cytotoxicity on human renal cortical cells was increased depend on concentration, whereas Ekongsan alone did not influence Simultaneous administration of cisplatin and Ekongsan, cytotoxicity of cisplain was reduced depend on the concentration of Ekongsan. With primary cultured proximal tubule cells results to reduce the cisplatin induced-cytotoxicity by Ekongsan was comparable to those with renal cortical cells. Based on these results, Ekongsan may has the potency to protect the nephrotoxicity induced by cisplatin.

      • Flat fading 채널에서 직교 복조 시스템의 성능 분석

        This paper presents an analysis of direct conversion receiver in frequency flat fading propagatoin environment. The channel is outdoor vehicular residential region and receiving system is vector demodulator with demodulation error owing to imbalance of gain and phase in I and Q channel. Simulations show that a receiving system with a compensated demodulation error is superior to a comnventional system in AWGN and flat fading environment.

        This paper presents an performance analysis of digital modulator and real transmitted data samples used in 5.8GHz mobile data communications. Transmitted data adopt BPSK modulation type for an easy hardware realization, a roll-off factor of RRC filter is 0.35, and data rate is 1Mbps. Experiments show that NCO is stable and modulated IF outputs is adequate to data format.

        In this paper, a receiving system specifications of a CDMA2000 mobile station are analyzed based on the CDMA2000 3GPP2 MS Minimum Performance. Lately the minimum performance for the CDMA2000 mobile station was recommended. But the specification was given with the value of FER under certain conditions. This value is the reference to decide the design parameter. therefore system parameters for the CDMA2000 mobile station such as noise figure, selectivity, dynamic range, and intermodulation are derived and simulated based on the 3GPP2.

      • PF 스케쥴링 알고리듬을 채택한 TxAA 전송 다이버시티 시스템의 채널 용량 해석

        In this paper, the performane of TxAA transmit diversity(TD) system combined with proportinal fairness(PF) scheduling algorithm is analyzed under independent frequency flat Rayleigh fading. The PDF of instantaneous channel gain of the each user and average channel capacity are induced in case of single transmit antenna and TD system, respectively. Numerical results shows that TxAA TD technique with closed-loop array gain have a cooperative relationship with multiuser diversity.
