        미세주입을 이용한 난자로의 분리된 미토콘드리아 전달

        고구마 현탁배양세포의 EST library에 높은 빈도로 존재하는 metallothionein (MT) 유전자를 선별하였다(IbMT3). MT 유전자는 세포와 조직의 스트레스 조절과 연관되어 있다고 알려져 있다. 본 연구에서 IbMT3 cDNA의 전장을 확보하여 염기서열 분석을 한 결과, IbMT3 유전자는 구조적으로 유형 3에 속하는 MT 단백질을 암호화하고 있었다. 식물에서 유형 3에 속하는 MT 단백질의 기능은 명확히 알려지지 않다. Northern blot 분석 결과, IbMT3 유전자는 고구마 식물체 잎보다 현탁배양 세포에서 매우 강하게 발현되었다. 일반적으로 세포배양은 세포에 산화 스트레스 상태를 부과하는 것으로 알려져 있다. 이에, 고구마 식물체에 산화 스트레스를 처리하여 IbMT3 유전자의 발현이 어떻게 조절되는지 조사하였다. 제초제인 methyl viologen (MV)을 6, 12, 24시간 동안 처리했을 때, IbMT3 유전자의 발현은 6시간 후에 아주 강하게 유도되었고 그 이후에는 감소함을 알 수 있었다. 저온 스트레스(15℃)를 24, 48시간 동안 처리했을 때, IbMT3 유전자는 처리시간이 경과함에 따라 발현이 더 많이 유도되었다. 이로써, IbMT3 유전자는 환경 및 산화 스트레스에 반응하여 발현이 조절되는 유전자임을 알 수 있었다. IbMT3 isoform은 고구마 식물체 내에서 항산화제로써 작용할 가능성이 있을 뿐 아니라, 스트레스 하에서의 세포 적응 메커니즘에 중요한 기능을 할 것으로 사료된다. Mitochondria play a central role in energy generation by using electron transport coupled with oxidative phosphorylation. They also participate in other important cellular functions including metabolism, apoptosis, signaling, and reactive oxygen species production. Therefore, mitochondrial dysfunction is known to contribute to a variety of human diseases. Furthermore, there are various inherited diseases of energy metabolism due to mitochondrial DNA (mtDNA) mutations. Unfortunately, therapeutic options for these inherited mtDNA diseases are extremely limited. In this regard, mitochondrial replacement techniques are taking on increased importance in developing a clinical approach to inherited mtDNA diseases. In this study, green fluorescence protein (GFP)-tagged mitochondria were isolated by differential centrifugation from a mammalian cell line. Using microinjection technique, the isolated GFP-tagged mitochondria were then transferred to bovine oocytes that were triggered for early development. During the early developmental period from bovine oocytes to blastocysts, the transferred mitochondria were observed using fluorescent microscopy. The microinjected mitochondria were dispersed rapidly into the cytoplasm of oocytes and were passed down to subsequent cells of 2-cell, 4-cell, 8-cell, morula, and blastocyst stages. Together, these results demonstrate a successful in vitro transfer of isolated mitochondria to oocytes and provide a model for mitochondrial replacement implicated in inherited mtDNA diseases and animal cloning.

        Synthesis of nanoparticle CT contrast agents: <i>in vitro</i> and <i>in vivo</i> studies

        <P>Water-soluble and biocompatible D-glucuronic acid coated Na<SUB>2</SUB>WO<SUB>4</SUB> and BaCO<SUB>3</SUB> nanoparticles were synthesized for the first time to be used as x-ray computed tomography (CT) contrast agents. Their average particle diameters were 3.2 ± 0.1 and 2.8 ± 0.1 nm for D-glucuronic acid coated Na<SUB>2</SUB>WO<SUB>4</SUB> and BaCO<SUB>3</SUB> nanoparticles, respectively. All the nanoparticles exhibited a strong x-ray attenuation. <I>In vivo</I> CT images were obtained after intravenous injection of an aqueous sample suspension of D-glucuronic acid coated Na<SUB>2</SUB>WO<SUB>4</SUB> nanoparticles, and positive contrast enhancements in the kidney were clearly shown. These findings indicate that the nanoparticles reported in this study may be promising CT contrast agents.</P>

        N/A Background: Peptic ulcer is a heterogenous disease which has genetic or acquired factors in the development of the disease. Serum pepsinogen I (PG I) was reported as a marker of the genetic factors. Rut it is less reported about the relations between serum PG I and acquired factors in the development of the disease. The aim of this study was to evaluate serum PG I levels as the genetic or acquired factors in the development of duodenal ulcer. Methods: Fasting serum PG I levels were tested in 85 patients with duodenal ulcer, 64 their family members, and 52 normal controls. Results: 1) Serum PG I levels in duodenal ulcer patients (104±44ng/ml) and their family members (74±29ng/ml) were significantly higher than that of the control subjects (57±24ng/ml)(p<0.05). 2) The PG I levels of the family in their family members (78±30ng/ml) who had duodenal ulcer patient with elevated PGI levels in their family were somewhat higher than that of the family members (67±19ng/ml) who had the duodenal ulcer patients with normal PG I levels, but the difference did not acquire statistical significance (p<0.05). 3) The mean serum PG I level of smoker (111±47ng/ml) was higher than that of non-smoker (84±35ng/ml). 4) The mean serum PG I levels did not change in duodenal ulcer patients even after the treatment with H2-receptor blocker. Conclusion: These results suggested that serum PG I levels may be useful as a genetic marker in duodenal ulcer disease, but might be affected by acquired risk factor like smoking as well.

        Various ligand-coated ultrasmall gadolinium-oxide nanoparticles: Water proton relaxivity and in-vivo T1 MR image

        <P>Surface coating of nanoparticles with ligands is essential in magnetic resonance imaging (MRI) because of solubility in water and biocompatibility. In this study, five organic molecules were used for surface coating of ultrasmall gadolinium-oxide (Gd2O3) nanoparticles (d (avg) = 2.0 nm). All of the samples showed large longitudinal (r(1)) and transverse (r(2)) water proton relaxivities with r(2)/r(1) ratios that were close to one, corresponding to ideal conditions for T-1 MRI contrast agents. Finally, in-vivo T-1 MR images were acquired to prove the effectiveness of the surface-coated ultrasmall Gd2O3 nanoparticles as a T-1 MRI contrast agent.</P>

        통상적 99mTc - DTPA 신장스캔을 이용한 GFR측정

        N/A Many previously described nuclear medicine procedures to assess glomerular filtration rate (GFR) required numerous blood samples obtained over a period of several hours to determine plasma concentrations of the injected radiopharmaceuticals. And other indirect methods of determining renal clearance have some problems due to individual variations in volume of distribution of the radionuclides used. Rescently reported Jackson's method have the great advantages that is a direct measurement method requiring less than 40 min of imaging time and single blood sampling. And it correctly accounts for individual variations in volume of distribution of the radiopharmaceuticals and can be done with routine renal scintigraphy. We measured Tc- 99m-DTPA renal clearance with Jackson's method during the routine Tc-99m-DTPA renal scintigraphy in 63 patients admitted to department of internal medicine in SNUH. In 23 cases among 63 patients creatinine clearence was accounted simultaneously. The range of ClDPDA was from 19.9 ml/min to 170 mi/min and the correlation of ClDPDA and creatinine clearance was discribed by Y=16.2570+0.7852X(X=C1DPDA, Y=creatinine clearance). And the correlation coefficient r was 0.88. We concluded that Tc-99m-DTPA renal clearance measurement with Jackson's method was clinically useful to account GFR that can be done with routine Tc-99m-DTPA renal scintigraphy simultaneously.

      • Role of protein kinase C delta in X-ray-induced apoptosis of keratinocyte

        <P>Abstract: </P><P>In this study, we investigated the process of X-ray-induced apoptosis of skin keratinocyte, and the functional role of protein kinase C delta (PKC&dgr;) and downstream signalling cascade. High-dose X-ray irradiation (10 Gy) led to the apoptosis of HaCaT keratinocyte, accompanied by PKC&dgr; cleavage. Treatment with PKC&dgr; inhibitor and adenoviral transduction of dominant-negative PKC&dgr; clearly inhibited the X-ray-induced apoptosis of keratinocyte. In addition, X-ray induced the phosphorylation of extracellular signal-regulated kinases 1/2 (ERK1/2) and inhibition by ERK1/2 inhibitor abrogated the X-ray-induced apoptosis. Interestingly, overexpression of dominant-negative PKC&dgr; markedly blocked the X-ray-induced phosphorylation of ERK1/2, suggesting that ERK1/2 is the functional downstream effector of PKC&dgr;. Next, we investigated the difference between UVB and X-ray response. UVB induced the apoptosis of keratinocyte in a PKC&dgr;-dependent manner, similar to X-ray response. However, UVB irradiation induced the phosphorylation of c-jun N-terminal kinases (JNK) and inhibition of JNK significantly protected the UVB-induced apoptosis. These results demonstrate that PKC&dgr; is a key regulator in X-ray-induced apoptosis of keratinocyte and suggest that there is subtle difference in downstream signalling cascade between UVB and X-ray response of keratinocyte.</P>

        <P><B>Abstract</B></P> <P>Ultrasmall Gd<SUB>2</SUB>O<SUB>3</SUB> nanoparticles can potentially be used as a positive magnetic resonance imaging (MRI) contrast agent owing to their high content of Gd(III), which exhibits the highest spin magnetic moment of all elements due to its seven unpaired 4<I>f</I> electrons. Herein, the above nanoparticles (average diameter = 2.0 nm) were coated with hydrophilic biocompatible polyacrylic acids (PAAs) of different molecular weights (<I>M</I> <SUB>w</SUB> = 1200, 5100, 15000 Da), and the PAA-size-dependent relaxometric properties of the thus obtained composites were investigated. In addition, the biocompatibility of these composites was assessed by <I>in-vitro</I> cell viability measurements.         위 림프종의 내시경적 소견

        Gastric lymphoma occupies about i-7% of gastric cancer and is the mast common type of extranodal lynuphama. To evaluate the ieridoegopic morphologic characteristics of gastric lymphoma, we analysed the endoecon finds in 5 patients with pathologically-proven gastric lymphoma. 1) The mean ague vvas 53.4(t 13.6) years and the ratio of male to female was 2 : 1. 2) The most frmqwent symptom was epagastric pain(89%), followed by weight loss(25%). indigestion, nausea, vomiting(13%), fever(7%); dysphaga(%) and abdominal distension(2%). 3) The ntcst common size of gastric Imphoma was body of stomach(3l%), followed by antrum(29%)sid entire stomach(16%). 4) Gastrascopy revealed-ulcerative lesion in 17 cases(3896), ulcerative and caass lesion in 14 case(31), erive lesion in 7 cases(15% inass lesion 4 case(9%) and mixed type in 3 case. 5) As a ,initial endoscopic impression gastric lymphoma was suggested in 5 cases(11%), advanced fastric cancer in 29 cases(65%), early gastric; cancer in 4 cases(9'), leiomyoma in 5 cases(tl)and benign gastric peptic ulcer in 2 cases(4). The grs endasaapic findings which suggested gastxic lymphoma initially were preserved distensibiliy of stosnach wall compared to the severity of disease involvement (3 cases), and submucosa tumor with ulcer those margin was heaved-up(2 cases). 6) Ptho[logic reports of biopsy specimens were malignant lymphoma in 35 cases(78%), adenaclrcioma in 3 cases, undifferentiated carcinoma in 2 cases, and chronic superficial gastritis in 5 ciases. 7) In conclusion, gastric lymphoma couldn't be differentiated from the other disease by their endoscopie gross morphology a1ane except a few cases and even endascopic biopsy were not able to detect malignant lyrnplloma cell in about 22% patients. So recognition of specific endoscopic patterns along with high index of suspicion and'multiple biapsy may lead to earlier diagnosis of gastric lymphoma.
