        A probabilistic analysis of Miner’s law for different loading conditions

        In this paper, the normalized variable V=(log N-B)(log Δσ-C), as derived from the probabilistic S-N field of Castillo and Canteli, is taken as a reference for calculation of damage accumulation and probability of failure using the Miner number in scenarios of variable amplitude loading. Alternative damage measures, such as the classical Miner and logarithmic Miner, are also considered for comparison between theoretical lifetime prediction and experimental data. The suitability of this approach is confirmed for it provides safe lifetime prediction when applied to fatigue data obtained for riveted joints made of a puddle iron original from the Fão bridge, as well as for data from experimental programs published elsewhere carried out for different materials (aluminium and concrete specimens) under distinct variable loading histories.

        Strain energy-based fatigue life prediction under variable amplitude loadings

        With the aim to evaluate the fatigue damage accumulation and predict the residual life of engineering components under variable amplitude loadings, this paper proposed a new strain energy-based damage accumulation model by considering both effects of mean stress and load interaction on fatigue life in a low cycle fatigue (LCF) regime. Moreover, an integrated procedure is elaborated for facilitating its application based on S-N curve and loading conditions. Eight experimental datasets of aluminum alloys and steels are utilized for model validation and comparison. Through comparing experimental results with model predictions by the proposed, Miner’s rule, damaged stress model (DSM) and damaged energy model (DEM), results show that the proposed one provides more accurate predictions than others, which can be extended for further application under multi-level stress loadings.

      • Analytical investigation of the cyclic behaviour of I-shaped steel beam with reinforced web using bonded CFRP

        Recent experimental studies showed that deep steel I-shaped profiles classified as high ductility class sections in seismic design international codes exhibit low deformation capacity when subjected to cyclic loading. This paper presents an innovative retrofit solution to increase the rotation capacity of beams using bonded carbon fiber reinforced polymers (CFRP) patches validated with advanced finite element analysis. This investigation focuses on the flexural cyclic behaviour of I-shaped hot rolled steel deep section used as beams in moment-resisting frames (MRF) retrofitted with CFRP patches on the web. The main goal of this CFRP reinforcement is to increase the rotation capacity of the member without increasing the overstrength in order to avoid compromising the strong column-weak beam condition in MRF. A finite element model that simulates the cyclic plasticity behavior of the steel and the damage in the adhesive layer is developed. The damage is modelled using the cohesive zone modelling (CZM) technique that is able to capture the crack initiation and propagation. Details on the modelling techniques including the mesh sensitivity near the fracture zone are presented. The effectiveness of the retrofit solution depends strongly on the selection of the appropriate adhesive. Different adhesive types are investigated where the CZM parameters are calibrated from high fidelity fracture mechanics tests that are thoroughly validated in the literature. This includes a rigid adhesive commonly found in the construction industry and two tough adhesives used in the automotive industry. The results revealed that the CFRP patch can increase the rotation capacity of a steel member considerably when using tough adhesives.

        Effect of blood type on anti-α-Gal immunity and the incidence of infectious diseases

        The identification of factors affecting the susceptibility to infectious diseases is essential toward reducing their burden on the human population. The ABO blood type correlates with susceptibility to malaria and other infectious diseases. Due to the structural similarity between blood antigen B and Galα1-3Galβ1-(3)4GlcNAc-R (α-Gal), we hypothesized that self-tolerance to antigen B affects the immune response to α-Gal, which in turn affects the susceptibility to infectious diseases caused by pathogens carrying α-Gal on their surface. Here we found that the incidence of malaria and tuberculosis, caused by pathogens with α-Gal on their surface, positively correlates with the frequency of blood type B in endemic regions. However, the incidence of dengue fever, caused by a pathogen without α-Gal, was not related to the frequency of blood type B in these populations. Furthermore, the incidence of malaria and tuberculosis was negatively correlated with the anti-α-Gal antibody protective response. These results have implications for disease control and prevention.

        <P>Chagas disease, a neglected illness, affects nearly 12-14 million people in endemic areas of Latin America. Although the occurrence of acute cases sharply has declined due to Southern Cone Initiative efforts to control vector transmission, there still remain serious challenges, including the maintenance of sustainable public policies for Chagas disease control and the urgent need for better drugs to treat chagasic patients. Since the introduction of benznidazole and nifurtimox approximately 40 years ago, many natural and synthetic compounds have been assayed against Trypanosoma cruzi, yet only a few compounds have advanced to clinical trials. This reflects, at least in part, the lack of consensus regarding appropriate in vitro and in vivo screening protocols as well as the lack of biomarkers for treating parasitaemia. The development of more effective drugs requires (i) the identification and validation of parasite targets, (ii) compounds to be screened against the targets or the whole parasite and (iii) a panel of minimum standardised procedures to advance leading compounds to clinical trials. This third aim was the topic of the workshop entitled Experimental Models in Drug Screening and Development for Chagas Disease, held in Rio de Janeiro, Brazil, on the 25th and 26th of November 2008 by the Fiocruz Program for Research and Technological Development on Chagas Disease and Drugs for Neglected Diseases Initiative. During the meeting, the minimum steps, requirements and decision gates for the determination of the efficacy of novel drugs for T. cruzi control were evaluated by interdisciplinary experts and an in vitro and in vivo flowchart was designed to serve as a general and standardised protocol for screening potential drugs for the treatment of Chagas disease.</P>
