Estrogen exposure causes the progressive growth of SK-Hep1-derived tumor in ovariectomized mice

Liver cancer, one of the leading death causes, has different incidence and mortality rates in men and women. The influencing factor is considered to estrogen. However, the role of estrogen in liver cancer remains controversial. In this study, we investigated the effects of estrogen on tumor progression. Total RNA sequencing was analyzed in SK-Hep1-derived tumor tissues, and 15 genes were expressed only in female mice. Among the differentially expressed genes, matrix metalloprotease 7 (MMP7), germ cell associated 1 (GSG1), and chromosome 6 open reading frame 15 (C6orf15) were associated with significantly different overall survival rates based on their expression level in liver cancer patients. Interestingly, exogenous estrogen aggravated SK-Hep1-derived tumor growth in ovariectomized (OVX) mice. When OVX mice were treated with exogenous estrogen, SK-Hep1-derived tumor tissues exhibited high MMP7 expression levels and low GSG1 and C6orf15 expression levels. These expression patterns were consistent with those of liver cancer patients with low overall survival rates. These results suggest that these genes are expected to be prognostic biomarkers of liver cancer. In conclusion, our results suggest that continuous estrogen exposure may promote tumor growth in OVX mice.
Sex‑dependent effects of estrogen pellets in human liver cancer xenograft models

Liver cancer shows noticeable differences in the incidence rate and mortality between genders. To investigate the estrogen effect on tumor progression in liver cancer, we developed a xenograft model using estrogen pellets. SK-Hep1 cells (human male liver carcinoma) were inoculated into male or female nude mice. Subsequently, estrogen pellets were subcutaneously implanted into these xenograft models. Interestingly, the marked adverse effect of estrogen pellets (0.5 mg/21 days) were observed in the male-derived xenograft model, with increased ulcerative dermatitis in male mice than in female mice. Additionally, necrosis was observed in male mice with SK-Hep1-derived tumors. However, the estrogen pellet (0.5 mg/60 days) did not exhibit these adverse effects. Tumor growth in female mice was significantly suppressed by estrogen (0.5 mg/60 days). Tumor growth was also suppressed in male mice implanted with estrogen (0.5 mg/60 days), but the suppression was not significant. We found that estrogen-induced skin damage was more severe in male mice than female mice. The tumor suppression of estrogen was effective in female mice compared to male mice bearing liver cancer. The results suggest that the sex difference affects estrogen activity and thus should be considered in the preclinical assessment.

이 연구는 결혼과 자녀, 가족에 대한 청년들의 인식의 변화를 실증자료를 통해 검토하고, 이에 따른 결혼 및 가족에 대한 성역할 태도와 인식 변화의 의미를 분석하였다. 이를 위해 통계청의 사회조사와 2018년 한국보건사회연구원이 발표한 전국출산력 및 가족보건복지 실태조사 결과를 바탕으로 내용분석을 실시하였다. 연구결과, 청년층에서 자발적 비혼률이 높아지고 있었으며, 결혼에 대한 긍정적 인식의 감소하였다. 또한 가사 및 신혼집 마련에 대해 평등분담, 일 우선주의는 감소하고, 가족 우선주의는 증가하였다. 연령별 가족관계만족도의 차이를 통해 가족관계의 질에 대한 세대간 차이를 확인하였다. 향후, 세대간 그리고 세대 내의 차이를 보다 명확히 진단하여, 남성 돌봄 역량 강화를 위한 정책을 제공하고, 나아가 청년들의 다양한 가족구성 실험을 지원할 필요가 있음을 제언하였다. This study examines the changes of young people’s perception about marriage, child-bearing, and family structure by analyzing empirical data, and highlights the meaning of the their changed attitude toward gender role in terms of marriage and family. A content analysis has also been conducted based on the social survey by Statistics Korea and The 2018 National Survey on Fertility and Family Health and Welfare published by KIHASA. This paper shows an increase in the rate of young people intentionally choosing not to marry, reflecting a decrease in positive perceptions of marriage. Furthermore, as the Household labor as well as housing cost were equally shared, work priority decreased, and family priority increased. The inter-generational age gaps and their impacts on family relationship were identified through their different degrees of satisfaction in family relationship. Finally, the study suggests that the differences between and within generations should be diagnosed more clearly in order to provide policies that strengthen male caring capacity and support young people’s various family constitution.
