Satellite Bulk Tropospheric Temperatures as a Metric for Climate Sensitivity

We identify and remove the main natural perturbations (e.g. volcanic activity, ENSOs) from the global mean lower tropospheric temperatures (TLT) over January 1979 - June 2017 to estimate the underlying, potentially human-forced trend. The unaltered value is +0.155 K dec−1 while the adjusted trend is +0.096 K dec−1, related primarily to the removal of volcanic cooling in the early part of the record. This is essentially the same value we determined in 1994 (+0.09 K dec−1, Christy and McNider, 1994) using only 15 years of data. If the warming rate of +0.096 K dec−1 represents the net TLT response to increasing greenhouse radiative forcings, this implies that the TLT tropospheric transient climate response (ΔTLT at the time CO2 doubles) is +1.10 ± 0.26 K which is about half of the average of the IPCC AR5 climate models of 2.31 ± 0.20 K. Assuming that the net remaining unknown internal and external natural forcing over this period is near zero, the mismatch since 1979 between observations and CMIP-5 model values suggests that excessive sensitivity to enhanced radiative forcing in the models can be appreciable. The tropical region is mainly responsible for this discrepancy suggesting processes that are the likely sources of the extra sensitivity are (a) the parameterized hydrology of the deep atmosphere, (b) the parameterized heat-partitioning at the oceanatmosphere interface and/or (c) unknown natural variations.

Objective Determine differences between faculty, residents, and nurses regarding night shift preparation, performance, recovery, and perception of emotional and physical health effects. Methods Survey study performed at an urban university medical center emergency department with an accredited residency program in emergency medicine. Results Forty-seven faculty, 37 residents, and 90 nurses completed the survey. There was no difference in use of physical sleep aids between groups, except nurses utilized blackout curtains more (69%) than residents (60%) and faculty (45%). Bedroom temperature preference was similar. The routine use of pharmacologic sleep aids differed: nurses and residents (both 38%) compared to faculty (13%). Residents routinely used melatonin more (79%) than did faculty (33%) and nurses (38%). Faculty preferred not to eat (45%), whereas residents (24%) preferred a full meal. The majority (>72%) in all groups drank coffee before their night shift and reported feeling tired despite their routine, with 4:00 a.m. as median nadir. Faculty reported a higher rate (41%) of falling asleep while driving compared to residents (14%) and nurses (32%), but the accident rate (3% to 6%) did not differ significantly. All had similar opinions regarding night shift-associated health effects. However, faculty reported lower level of satisfaction working night shifts, whereas nurses agreed less than the other groups regarding increased risk of drug and alcohol dependence. Conclusion Faculty, residents, and nurses shared many characteristics. Faculty tended to not use pharmacologic sleep aids, not eat before their shift, fall asleep at a higher rate while driving home, and enjoy night shift work less.

Cardiovascular and central nervous system (CNS) toxicity, including tachydysrhythmia, agitation, and seizures, may arise from cocaine or bupropion use. We report acute toxicity from the concomitant use of cocaine and bupropion in a 25-year-old female. She arrived agitated and uncooperative, with a history of possible antecedent cocaine use. Her electrocardiogram demonstrated tachycardia at 130 beats/min, with a corrected QT interval of 579 ms. Two doses of 5 mg intravenous metoprolol were administered, which resolved the agitation, tachydysrhythmia, and corrected QT interval prolongation. Her comprehensive toxicology screen returned positive for both cocaine and bupropion. We believe clinicians should be aware of the potential for synergistic cardiovascular and CNS toxicity from concomitant cocaine and bupropion use. Metoprolol may represent an effective initial treatment. Unlike benzodiazepines, metoprolol directly counters the pharmacologic effects of stimulants without respiratory depression, sedation, or paradoxical agitation. A lipophilic beta-blocker, metoprolol has good penetration of the CNS and can counter stimulant-induced agitation.

<P>Site-specific glycosylation (SSG) of glycoproteins remains a considerable challenge and limits further progress in the areas of proteomics and glycomics. Effective methods require new approaches in sample preparation, detection, and data analysis. While the field has advanced in sample preparation and detection, automated data analysis remains an important goal. A new bioinformatics approach implemented in software called GP Finder automatically distinguishes correct assignments from random matches and complements experimental techniques that are optimal for glycopeptides, including nonspecific proteolysis and high mass resolution liquid chromatography/tandem mass spectrometry (LC/MS/MS). SSG for multiple N- and O-glycosylation sites, including extensive glycan heterogeneity, was annotated for single proteins and protein mixtures with a 5% false-discovery rate, generating hundreds of nonrandom glycopeptide matches and demonstrating the proof-of-concept for a self-consistency scoring algorithm shown to be compliant with the target-decoy approach (TDA). Influence of continental outflow events on the aerosol composition at Cheju Island,South Korea

The chemical composition of aerosols measured at Cheju Island, Korea, over the 3-year period March 1992 to February 1995 are presented and discussed, with a particular emphasis on the Pacific Exploratory Mission in the Western Pacific (PEM-West B) time period. Cheju Island is under the influence of continental outflow conditions nearly 70% of the year, and as a result the aerosol loading of sea salt as well as continental aerosol components is high. The 3-year mean values derived from the daily tape filters are non-sea-salt (nss) SO₄²?=6.8㎍/㎥, NO₃?=1.2㎍/㎥, Cl??=1.9㎍/㎥, Na+=1.7㎍/㎥, Ca²?=0.5㎍/㎥, NH₄?=1.3㎍/㎥, Mg²?=0.3㎍/㎥, and K+=0.4㎍/㎥. Sea-salt components show peak values in winter, while calcium, nitrate, potassium, and to a lesser extent, sulfate and ammonium, show higher values in the spring, and all species exhibit a pronounced minimum in summer. Trajectory and principal component analysis show that elevated levels of primary aerosols (both sea salt and soil-derived) occur with strong wind conditions associated with winter and spring, and high concentrations of non-sea-salt components are most strongly associated with springtime continental outflow events. During the PEM-West B period, nitrate and calcium are found to be ∼60% higher than the annual mean, sea-salt components ∼30% higher, and sulfate ∼10% higher. The aerosol data are combined with gaseous SO₂ concentrations, precipitation chemistry data and companion aerosol measurements taken at the same site, to provide further insights into the aerosol composition at Cheju. At Cheju, sulfate and ammonium are found mostly in the fine fraction (∼80 to 90%), while calcium and nitrate reside in the coarse size fraction. sulfate and nitrate are not associated with the primary aerosols, but rather become associated with the aerosol during the long-range transport process.

<P>Gqalpha family members (Gqalpha, G11alpha, G14alpha, and G15/16alpha) stimulate phospholipase Cbeta (PLCbeta) and inositol lipid signaling but differ markedly in amino acid sequence and tissue distribution predicting unappreciated functional diversity. To examine functional differences, we compared the signaling properties of Gqalpha, G14alpha, and G15alpha and their cellular responses in vascular smooth muscle cells (VSMC). Constitutively active forms of Gqalpha, G14alpha, or G15alpha elicit markedly different responses when introduced to VSMC. Whereas each Galpha stimulated PLCbeta to similar extents when expressed at equal protein levels, Gqalpha and G14alpha but not G15alpha initiated profound cell death within 48 h. This response was the result of activation of apoptotic pathways, because Gqalpha and G14alpha, but not G15alpha, stimulated caspase-3 activation and did not alter phospho-Akt, a regulator of cell survival pathways. Gqalpha and G14alpha stimulate nuclear factor of activated T cell (NFAT) activation in VSMC, but Galpha-induced cell death seems independent of PKC, InsP(3)/Ca(2+), and NFAT, in that pharmacological inhibitors of these pathways did not block cell death. Gene expression analysis indicates that Gqalpha, G14alpha, and G15alpha each elicit markedly different profiles of altered gene sets in VSMC after 24 h. Whereas all three Galpha stimulated changes (> or =2-fold) in 50 shared mRNA, Gqalpha and G14alpha (but not G15alpha) stimulated changes in 221 shared mRNA, many of which are reported to be pro-apoptotic and/or involved with TNF-alpha signaling. We were surprised to find that each Galpha also stimulated changes in nonoverlapping Galpha-specific gene sets. These findings demonstrate that Gqalpha family members activate both overlapping and distinct signaling pathways and are more functionally diverse than previously thought.</P>
