        Comparison of the Three Molecular Diagnostic Assays for Molecular Identification of Mycobacterium tuberculosis and Nontuberculous Mycobacteria Species in Sputum Samples

        Mycobacterium tuberculosis (MTB) continues to be one of the main causative agents of tuberculosis (TB); moreover, the incidence of nontuberculous mycobacteria (NTM) infections has been rising gradually in both immunocompromised and immunocompetent patients. Precise and rapid detection and identification of MTB and NTM in respiratory specimens are thus important for MTB infection control. Molecular diagnostic methods based on the nucleic acid amplification test (NAAT) are known to be rapid, sensitive, and specific compared to the conventional acid-fast bacilli (AFB) smear and mycobacterial culture methods. In the present study, the clinical performances of three commercial molecular diagnostic assays, namely TB/NTM PCR (Biocore), MolecuTech Real MTB-ID<SUP>®</SUP> (YD Diagnostics), and REBA Myco-ID<SUP>®</SUP> (YD Diagnostics), were evaluated with a total of 92 respiratory specimens (22 AFB smear positives and 67 AFB smear negatives). The sensitivity and specificity of TB/NTM PCR were 100% and 75.81%, respectively. The corresponding values of MolecuTech Real MTB-ID<SUP>®</SUP> and REBA Myco-ID<SUP>®</SUP> were 56.52% and 90.32%, and 56.52% and 82.26%, respectively. TB/NTM PCR showed the highest sensitivity; however, the concordant rate was 10% compared with sequence analysis. Although MolecuTech Real MTB-ID<SUP>®</SUP><SUP></SUP> showed lower sensitivity, its specificity was the highest among the three methods. REBA Myco-ID<SUP>®</SUP> allowed accurate classification of NTM species; therefore, it was the most specific diagnostic method. Of the three PCR-based methods, MolecuTech Real MTB-ID<SUP>®</SUP> showed the best performance. This method is expected to enable rapid and accurate identification of MTB and NTM.

        Probe-based qPCR Assay for Rapid Detection of Predominant Candida glabrata Sequence Type in Korea

        Recent years have seen an increase in the incidence of candidiasis caused by non-albicans Candida (NAC) species. In fact, C. glabrata is now second only to C. albicans as the most common cause of invasive candidiasis. Therefore, the rapid genotyping specifically for C. glabrata is required for early diagnosis and treatment of candidiasis. A number of genotyping assays have been developed to differentiate C. glabrata sequence types (STs), but they have several limitations. In the previous study, multi-locus sequence typing (MLST) has performed with a total of 101 C. glabrata clinical isolates to analyze the prevalent C. glabrata STs in Korea. A total of 11 different C. glabrata STs were identified and, among them, ST-138 was the most commonly classified. Thus, a novel probe-based quantitative PCR (qPCR) assay was developed and evaluated for rapid and accurate identification of the predominant C. glabrata ST-138 in Korea. Two primer pairs and hybridization probe sets were designed for the amplification of internal transcribed spacer 1 (ITS1) region and TRP1 gene. Analytical sensitivity of the probe-based qPCR assay was 100 ng to 10 pg and 100 ng to 100 pg (per 1 μL), which target ITS1 region and TRP1 gene, respectively. This assay did not react with any other Candida species and bacteria except C. glabrata. Of the 101 clinical isolates, 99 cases (98%) were concordant with MLST results. This novel probebased qPCR assay proved to be rapid, sensitive, highly specific, reproducible, and cost-effective than other genotyping assay for C. glabrata ST-138 identification.

      • 시장경제질서에서 정부실패의 원인과 정부개입의 폐해

        본 연구의 목적은 시장경제질서에서 정부의 시장 개입이 어떤 결과를 가져올 것인지를 비판적인 시각에서 이론적으로 검토하는 것이다. 특히 정부실패의 원인과 정부개입이 가져올 폐해와 부작용을 집중적으로 조명하였다. 정부 개입이 야기하는 시장의 왜곡, 질서충돌에 따른 계층 간의 갈등과 자원 낭비, 부정부패와 비리의 개연성, 분배문제의 악화, 그리고 책임소재의 부재 등은 비록 시장이 만족스럽지 못할 경우가 있다 하더라도 정부개입은 최소화해야 함을 말해준다. 정부의 개입은 가능한 모든 소비자가 동일하게 소비할 수 있는 순수한 공공재의 공급에 한정해야 한다. 예를 들어 국방, 치안, 소방, 추상적이고 일반적이며 자명한 법제도의 구비 등이 그것들이다. 이것은 초기 고전적 자유주의자들과 오스트리아 학파를 중심으로 하는 구(舊) 자유주의자들의 생각과 그 괘를 같이한다. The main purpose of this paper is to illuminate theoretically and in a critical view what the interventionist of the economy and coercive system leads to. This paper tries to show why the big government cannot but fail and what bad effects it makes. The government intervention in a market economy system rests on the concept of the market failure. But this paper argues that on the contrary to what might be believed it cannot be the reason of government intervention and furthermore the concept is lack of clarity and based even on the wrong logics. As soon as government intervenes in a market, conflicts are created, because each person or group may participate in a scramble to be a net gainer rather than a net loser. The conflict appears also on the scene. Every economic order has mechanisms and institutions to allocate scarce resources. However the way of resource allocation is totally different between by the market and by the government. Clashes arise when the rules, each of which has a different mechanism or function in resource allocation, exist in a economy. It results in market distortion or less market coordination, resource waste, deterioration of the distribution problem, and frictions among social classes. Another reason why an intervention coercive system makes more serious problems is in the fact that we cannot charge the government failure and the politicians and officials commit corruptions and illegalities with the high probability in comparison with the institution of market. The results of the study suggest that public services should sharply decline and we should retrospect the economic thoughts of the early classical and the Austrian school economists.

        본 논문은 신고전파 경제학의 틀을 부정하는 질서경제학의 학문적 당위성을 지식경제학의 흐름속에서 소개한다. 그러면서 본 논문은 신고전파 경제학의 틀 속에서 지식 문제를 다루고 있는 신제도 경제학과 질서자유주의의 발전 방향의 당위성도 설명하면서 질서경제학이 이들 이론들과의 조화가능성을 열어 놓고 있다. 지식경제학은 인간 이성의 한계를 받아들이고 열린 세계를 전제로 하면서 지식 문제를 분석의 핵심으로 다룬다. 지식경제학은 복잡계 경제학과 질서경제학으로 전개되고 있다. 질서경제학은 닫힌 세계를 부정하고, 인간의 자발적 상호조정 및 진화를 받아들이며 질서자유주의 이론을 융합하여 발전한 학문이다. 그런데 질서경제학 형성의 주된 역할을 한 질서자유주의가 이론으로 발전하기 위해서 신고전파 경제이론을 받아들일 것을 암묵적으로 요구한다. 신제도 경제학은 신고전파 경제이론을 부정하지 않으면서 제도 분석을 통한 인간의 인식 한계의 문제를 다루고 있다. 이런 점에서, 질서경제학이 신고전파 경제이론과 접목 가능할 수 있는 여지를 보여준다. The strengths of the neoclassical economics are identified as a perfect formalizing with a theoretical consistency under assumptions of a perfect mankind. But the order-economics as a field of knowledge economics starts from the rejection of the assumptions for the neoclassical economics. It tries to draw a real picture of a mankind suffering in a limitation of cognition and his open world. This paper is to explore why the order-economics should be treated as an important analysis-tool for describing and explaining economic problems and to seek the possibility of introducing the choice model of neoclassical economics to the order-economics.

        관용 표현을 이루는 접속 관형절 구성 : 인과 관계를 나타내는 의미를 중심으로

        In general, the adnominal clause modify the head nouns which have core meaning and are free form in a sentence. However, in some ‘V+adnominal clause endings+N+P’ constructions core meanings of nouns become weakened or changed. And then these ‘V+adnominal clause endings+N+P’ constructions function as the adverbial endings in the contemporary Korean. We will call these specific constructions as ‘Construction of Conjunctive Adnominal Clause’. The purpose of this study analogize the Grammaticalization of the ‘Construction of Conjunctive Adnominal Clause’. For the purpose of this study we analyze Idioms based in these ‘V+adnominal clause endings+N+P’ constructions, in the sense that meanings and functions. In this study we will compare meanings that ‘-ㄴ 결과 (로), -ㄴ 관계로, -ㄴ 덕분에, -(으)ㄴ/-는 마당에, -ㄹ 마당에, -는 바람에, -(으)ㄴ/- 는 탓에, -(으)ㄴ/-는 판국에, -ㄹ 판국에’ with the adverbial endings. In this study we deal with meanings of nouns in above idioms which relate causal connective relationship in meanings. In this paper, we suggest the possibility that Construction of Conjunctive Adnominal Clause are grammatical units as the connective endings in that grammatical function.

        山東 龍山文化의 衰落과 離散

        There was a highly-civilized society in Shandong Yongshan Culture, where ceremonial systems were established. However the society perished from the culture rapidly. This research will investigate both the causes for the sudden decline of the culture and the dispersion process of the culture due to the decline. According to the research, there existed regional differences in the decline tendency. Particularly there was a strong decline tendency at the southeastern region whose altitude is lower than the northwestern region It has been known that such differences were due to the rapid temperature drop in climate around 2000 B.C. and the deluge in that period. It is assumed that the deluge struck a conclusive blow to the community of the culture whose altitude became lower. The survival environment was changing like this, and the society would have been declined rapidly. The circumstance for the decline and the dispersion was assumed to be most heavily influenced by the climate change at that time. The decline are to assume a number of other causes. However, it would have been the main cause of the decline of a society under the condition when both there was a drastic change in subsistence environment and the society was not able to handle the change. Consequently, it can be assumed that a group of the people in the culture attempted to migrate massively to other region. Further, it can be assumed that, during the migration, the elements of the culture was to be reappeared at the other region.

        Comparison of One-Tube Nested-PCR and PCR-Reverse Blot Hybridization Assays for Discrimination of Mycobacterium tuberculosis and Nontuberculous Mycobacterial Infection in FFPE tissues

        Currently, molecular diagnostic assays based on nucleic acid amplification tests have been shown to effectively detect mycobacterial infections in various types of specimen, however, variable sensitivity was shown in FFPE samples according to the kind of commercial kit used. The present study therefore used automated PCR-reverse blot hybridization assay (REBA) system, REBA Myco-ID HybREAD 480®, for the rapid identification of Mycobacterium species in various types of human tissue and compared the conventional one-tube nested-PCR assay for detecting Mycobacterium tuberculosis (MTB). In conventional nested-PCR tests, 25 samples (48%) were MTB positive and 27 samples (52%) were negative. In contrast, when conducted PCR-REBA assay, 11 samples (21%) were MTB positive, 20 samples (39%) were NTM positive, 8 samples (15%) were MTB-NTM double positive, and 13 samples (25%) were negative. To determine the accuracy and reliability of the two molecular diagnostic tests, the one-tube nested-PCR and PCR-REBA assays, were compared with histopathological diagnosis in discordant samples. When conducted nested-PCR assay, 10 samples (59%) were MTB positive and seven samples (41%) were negative. In contrast, when conducted PCR-REBA test, three samples (17%) were MTB positive, 10 samples (59%) were NTM positive and four samples (24%) were negative. In conclusion, the automated PCR-REBA system proved useful to identify Mycobacterium species more rapidly and with higher sensitivity and specificity than the conventional molecular assay, one-tube nested-PCR; it might therefore be the most suitable tool for identifying Mycobacterium species in various types of human tissue for precise and accurate diagnosis of mycobacterial infection.

        In this study, I conducted a quantitative survey of the compound verbs that appear in the Proverbs. Based on this, the basic verbs were compared to the compound verbs to identify the aspects of compound verbs used in the Bible. As a result, only about 10% of the compound verbs included in the basic verbs were found to have a very high level of difficulty in the Bible. In addition, compound verbs, which have a large number of patterns in the posterior elements, account for about 37% of the total compound verbs, so it can be confirmed that pattern-specific learning is efficient in learning complex verbs. If complex verbs classified by frequency and pattern obtained through this survey are used step by step according to the learner level, more efficient compound verb learning can be achieved.

        Hormonal regulation of beta-catenin during development of the avian oviduct and its expression in epithelial cell-derived ovarian carcinogenesis

        <P><B>Abstract</B></P> <P>Beta-catenin (CTNNB1) is a dual function molecule that acts as a key component of the cadherin complex and WNT signaling pathway. It has a crucial role in embryogenesis, tumorigenesis, angiogenesis and progression of metastasis. Recently, it has been suggested that the CTNNB1 complex is a major regulator of development of the mouse oviduct and uterus. However, little is known about the <I>CTNNB1</I> gene in chickens. Therefore, in this study, we focused on the <I>CTNNB1</I> gene in the chicken reproductive tract and hormonal control of its expression in the chicken oviduct. <I>CTNNB1</I> was localized specifically to the luminal and glandular epithelium of the four segments of chicken oviduct and DES (diethylstilbestrol, a synthetic non-steroidal estrogen) increased its expression primarily in LE of the magnum. In addition, CTNNB1 mRNA and protein were expressed abundantly in glandular epithelium of endometrioid-type ovarian carcinoma, but not in normal ovaries. Moreover, <I>CTNNB1</I> expression was post-transcriptionally regulated via its 3′-UTR by binding with target miRNAs including <I>miR-217</I>, <I>miR-1467</I>, <I>miR-1623</I> and <I>miR-1697</I>. Collectively, these results indicate that <I>CTNNB1</I> is a novel gene regulated by estrogen in epithelial cells of the chicken oviduct and that it is also abundantly expressed in epithelial cells of endometrioid-type ovarian carcinoma suggesting that it could be used as a marker for diagnosis of ovarian cancer in laying hens and women.</P> <P><B>Highlights</B></P> <P> <UL> <LI> We examine hormonal and post-transcriptional regulation of chicken CTNNB1. </LI> <LI> We evaluate <I>CTNNB1</I> gene expression in cancerous ovaries of laying hens. </LI> <LI> CTNNB1 is a novel estrogen-induced gene during chick oviduct development. </LI> <LI> CTNNB1 is a post-transcriptionally regulated gene by specific microRNAs. </LI> <LI> CTNNB1 is abundant in the endometrioid-type ovarian carcinomas of laying hens. </LI> </UL> </P>
