      • Stability Analysis and Evaluation of ESR for Hysteresis Controlled Buck Converter

        Jinlong WANG,Jinbin ZHAO,Jianfeng DAI,Keqing QU,Tiankai YE 전력전자학회 2015 ICPE(ISPE)논문집 Vol.2015 No.6

Buck converter using hysteresis controller has been widely used for power supply in industry applications owing to its instantaneous load transient. However the equivalent series resistance (ESR) of output capacitor has a significant effect for the stability performance. In this paper, the critical value of ESR is analyzed and calculated in the hysteresis controlled buck converter. Finally, simulation results indicate that the larger output capacitor ESR is necessary to make sure the converter operate normally, otherwise, the converter will operate abnormally.

Three-dimensional dynamics of vortex-induced vibration of a pipe with internal flow in the subcritical and supercritical regimes

        Jinlong Duan,Ke Chen,Yunxiang You,Renfeng Wang,Jinlong Li 대한조선학회 2018 International Journal of Naval Architecture and Oc Vol.10 No.6

The Three-dimensional (3-D) dynamical behaviors of a fluid-conveying pipe subjected to vortex-induced vibration are investigated with different internal flow velocity v. The values of the internal flow velocity are considered in both subcritical and supercritical regimes. During the study, the 3-D nonlinear equations are discretized by the Galerkin method and solved by a fourth-order Runge-Kutta method. The results indicate that for a constant internal flow velocity v in the subcritical regime, the peak Cross-flow (CF) amplitude increases firstly and then decrease accompanied by amplitude jumps with the increase of the external reduced velocity. While two response bands are observed in the In-line (IL) direction. For the dynamics in the lock-in condition, 3-D periodic, quasi-periodic and chaotic vibrations are observed. A variety of CF and IL responses can be detected for different modes with the increase of v. For the cases studied in the supercritical regime, the dynamics shows a great diversity with that in the subcritical regime. Various dynamical responses, which include 3-D periodic, quasi-periodic as well as chaotic motions, are found while both CF and IL responses are coupled while v is beyond the critical value. Besides, the responses corresponding to different couples of m1 and m2 are obviously distinct from each other.

Novel Mutations in CYP51B from Penicillium digitatum Involved in Prochloraz Resistance

        Jinlong Wang,Jinhui Yu,Jing Liu,Yongze Yuan,Na Li,Muqing He,Ting Qi,Geng Hui,Li Xiong,Deli Liu 한국미생물학회 2014 The journal of microbiology Vol.52 No.9

Green mold caused by Penicillium digitatum is one of themost serious postharvest diseases of citrus fruit, and it isubiquitous in all citrus growing regions in the world. Sterol14α-demethylase (CYP51) is one of the key enzymes of sterolbiosynthesis in the biological kingdom and a prime targetof antifungal drugs. Mutations in CYP51s have been foundto be correlated with resistance to azole fungicides in manyfungal species. To investigate the mechanism of resistance toprochloraz (PRC) in P. digitatum, the PRC sensitivity wasdetermined in vitro in this study to assess the sensitivity of78 P. digitatum isolates collected in Hubei province. Theresults showed that 25 isolates were prochloraz-resistant(PRC-R), including six high-resistant (HR) strains, twelvemedium-resistant (MR) and seven low-resistant (LR) strains. A sequence analysis showed no consistent point mutationsof PdCYP51A in the PRC-R strains, but four substitutionsof CYP51B were found, Q309H in LR strains, Y136H andQ309H in HR strains, and G459S and F506I in MR strains, which corresponded to the four sensitivity levels. Based onthe sequence alignment analysis and homology modelingfollowed by the molecular docking of the PdCYP51B protein, the potential correlation between the mutations and PRC resistance is proposed.

Astragaloside IV Prevents Obesity-Associated Hypertension by Improving Pro-Inflammatory Reaction and Leptin Resistance

        Jiang, Ping,Ma, Dufang,Wang, Xue,Wang, Yongcheng,Bi, Yuxin,Yang, Jinlong,Wang, Xuebing,Li, Xiao Korean Society for Molecular and Cellular Biology 2018 Molecules and cells Vol.41 No.3

Low-grade pro-inflammatory state and leptin resistance are important underlying mechanisms that contribute to obesity-associated hypertension. We tested the hypothesis that Astragaloside IV (As IV), known to counteract obesity and hypertension, could prevent obesity-associated hypertension by inhibiting pro-inflammatory reaction and leptin resistance. High-fat diet (HFD) induced obese rats were randomly assigned to three groups: the HFD control group (HF con group), As IV group, and the As IV + ${\alpha}$-bungaratoxin (${\alpha}-BGT$) group (As IV+${\alpha}-BGT$ group). As IV ($20mg{\cdot}Kg^{-1}{\cdot}d^{-1}$) was administrated to rats for 6 weeks via daily oral gavage. Body weight and blood pressure were continuously measured, and NE levels in the plasma and renal cortex was evaluated to reflect the sympathetic activity. The expressions of leptin receptor (LepRb) mRNA, phosphorylated signal transducer and activator of transcription-3 (p-STAT3), phosphorylated phosphatidylinositol 3-kinase (p-PI3K), suppressor of cytokine signaling 3 (SOCS3) mRNA, and protein-tyrosine phosphatase 1B (PTP1B) mRNA, pro-opiomelanocortin (POMC) mRNA and neuropeptide Y (NPY) mRNA were measured by Western blot or qRT-PCR to evaluate the hypothalamic leptin sensitivity. Additionally, we measured the protein or mRNA levels of ${\alpha}7nAChR$, inhibitor of nuclear factor ${\kappa}B$ kinase subunit ${\beta}/nuclear$ factor ${\kappa}B$ ($IKK{\beta}/NF-KB$) and pro-inflammatory cytokines ($IL-1{\beta}$ and $TNF-{\alpha}$) in hypothalamus and adipose tissue to reflect the anti-inflammatory effects of As IV through upregulating expression of ${\alpha}7nAChR$. We found that As IV prevented body weight gain and adipose accumulation, and also improved metabolic disorders in HFD rats. Furthermore, As IV decreased BP and HR, as well as NE levels in blood and renal tissue. In the hypothalamus, As IV alleviated leptin resistance as evidenced by the increased p-STAT3, LepRb mRNA and POMC mRNA, and decreased p-PI3K, SOCS3 mRNA, and PTP1B mRNA. The effects of As IV on leptin sensitivity were related in part to the up-regulated ${\alpha}7nAchR$ and suppressed $IKK{\beta}/NF-KB$ signaling and pro-inflammatory cytokines in the hypothalamus and adipose tissue, since co-administration of ${\alpha}7nAChR$ selective antagonist ${\alpha}-BGT$ could weaken the improved effect of As IV on central leptin resistance. Our study suggested that As IV could efficiently prevent obesityassociated hypertension through inhibiting inflammatory reaction and improving leptin resistance; furthermore, these effects of As IV was partly related to the increased ${\alpha}7nAchR$ expression.

Identification of Saccharum CaM gene family and function characterization of ScCaM1 during cold and oxidant exposure in Pichia pastoris

        Wang Hengbo,Feng Meichang,Zhong Xiaoqiang,Yu Qing,Que Youxiong,Xu Liping,Guo Jinlong 한국유전학회 2023 Genes & Genomics Vol.45 No.1

Background Calmodulin (CaM) plays an essential role in binding calcium ions and mediating the interpretation of Ca2+ signals in plants under various stresses. However, the evolutionary relationship of CaM family proteins in Saccharum has not been elucidated. Objective To deduce and explore the evolution and function of Saccharum CaM family. Methods A total of 104 typical CaMs were obtained from Saccharum spontaneum and other 18 plant species. The molecular characteristics and evolution of those CaM proteins were analyzed. A typical CaM gene, ScCaM1, was subsequently cloned from sugarcane (Saccharum spp. hybrid). Its expression patterns in different tissues and under various abiotic stresses were assessed by quantitative real-time PCR. Then the green fluorescent protein was used to determine the subcellular localization of ScCaM1. Finally, the function of ScCaM1 was evaluated via heterologous yeast expression systems. Results Three typical CaM members (SsCaM1, SsCaM2, and SsCaM3) were identified from the S. spontaneum genome database. CaMs were originated from the two last common ancestors before the origin of angiosperms. The number of CaM family members did not correlate to the genome size but correlated with allopolyploidization events. The ScCaM1 was more highly expressed in buds and roots than in other tissues. The expression patterns of ScCaM1 suggested that it was involved in responses to various abiotic stresses in sugarcane via different hormonal signaling pathways. Noteworthily, its expression levels appeared relatively stable during the cold exposure in the cold-tolerant variety but significantly suppressed in the cold-susceptible variety. Moreover, the recombinant yeast (Pichia pastoris) overexpressing ScCaM1 grew better than the wild-type yeast strain under cold and oxidative stresses. It was revealed that the ScCaM1 played a positive role in reactive oxygen species scavenging and conferred enhanced cold and oxidative stress tolerance to cells. Conclusion This study provided comprehensive information on the CaM gene family in Saccharum and would facilitate further investigation of their functional characterization.

Hyper-heuristic algorithm for traffic flow-based vehicle routing problem with simultaneous delivery and pickup

Wang Zhengwei,Liu Jinlong,Zhang Jingling 한국CDE학회 2023 Journal of computational design and engineering Vol.10 No.6

To address the realistic problem of seriously reducing distribution efficiency and increasing distribution cost caused by road traffic congestion, this paper constructs a time-dependent speed describing vehicle travel speed and road traffic flow by simulating the change of urban traffic flow, to establish a vehicle route problem model considering traffic flow with distribution cost and customer satisfaction as optimization objectives. To solve this problem, a hyper-heuristic algorithm based on tabu search is designed in this paper, in which the underlying search operator is selected more efficiently by a high-level heuristic strategy. In addition, the correctness of the model and the effectiveness of the algorithm are verified by conducting simulation experiments on several benchmark sets. Experiment results are shown as the travel speed of the vehicle increases, the average customer satisfaction in lc1-type instances increases to 0.94. And the impact of urban traffic changes on logistics costs and customer satisfaction is further analyzed.
