Improvement of FK506 Production by Selection of 4-Aminobutyric Acid-tolerant Mutant and Optimization of its Fermentation Using Response Surface Methodology

        Tianyi Yang,Jiaojiao Li,Liangzhi Li,Haiyang Zhang,Jing Ma,Zhi Chen,Cuiying Hu,Xin Ju,Jiaolong Fu 한국응용생명화학회 2014 Applied Biological Chemistry (Appl Biol Chem) Vol.57 No.6

Tacrolimus (FK506), a 23-membered polyketidemacrolide with immunosuppressant activity, can be produced byStreptomyces tsukubaensis. We studied a variety of mutant strainsof S. tsukubaensis for the microbial production of FK506. Thebest strain (CZ-19) was obtained from the parent strain LLZ-1 bynitrosoguanidine mutation and 4-Aminobutyric acid (FK506precursor structure analogs) adaption. In the shake-flask experiments, titer of FK506 by CZ-19 was 532.44 mg/L, increased by 65.13%compared to that of the parent strain. Through single factorexperiments and response surface methodology, we furtheroptimized the medium for improved FK506 production by CZ-19in shake flask culture. The optimal medium for enhanced FK506production was as follows: 17.19 g/L corn starch, 21.78 g/Lglucose, 8.06 g/L peptone, and 18.98 mg/L 4-aminobutyric acid. The predicted FK506 titer was 906.49 mg/L, and the experimentaldata confirmed the validity of the model. The present studydemonstrates that S. tsukubaensis CZ-19 is a promising strain forindustrial production of FK506.

PEDOT-PSS-Coated FeFe(CN)6 Composite Cathode for Lithium-ion Batteries with the Improved Electrochemical Performances

        Lihuan Xu,Jiaojiao Ma,Pengju Guo,Chang Su 성균관대학교(자연과학캠퍼스) 성균나노과학기술원 2019 NANO Vol.14 No.9

Prussian-blue type FeFe(CN)6 nanocrystals with perfect nanocubic morphology were prepared by a facile hydrolytic precipitation method using Fe(CN)3- 6 as a single iron-source. The resulting FeFe(CN)6 nanocrystals have subsequently been encapsulated within a mixed electronically and ionically conducting polystyrene sulfonic acid (PSS)-doped poly(3,4-ethylenedioxythiophene) (PEDOT) (PEDOT-PSS) with ethylene glycol (EG) as the polar solvent to obtain a high electrical conducting organic–inorganic nanohybrid. The FeFe(CN)6/PEDOT-PSS nanohybrid offers discharge capacity of 106.3 mAh · g -1, which is improved compared to that of the naked FeFe(CN)6(94.4 mAh · g -1). Also, it demonstrated the improved capacity retention and rate capability, which makes it a promising way for high performance Li-ion batteries for energy storage application.

Effects of a Death Education Intervention for Older People with Chronic Disease and Family Caregivers: A Quasi-Experimental Study

        Weilin Chen,Hongmei Ma,Xiao Wang,Jiaojiao Chen 한국간호과학회 2020 Asian Nursing Research Vol.14 No.4

Purpose: To investigate the effectiveness of a structured death education program for older adults withchronic illness and their family caregivers. Methods: This study adopted two-group, nonrandomized quasi-experimental design. Patientecaregiverdyads in the intervention group (N ¼ 40 dyads) engaged in the death education program at the bedsideonce a week for 5 weeks, and were compared with participants (N ¼ 40 dyads) in the control group whoreceived usual health education. The program consisted of five sessions based on the Interaction Modelof Client Health Behavior. Death attitude, death competence, well-being, family function, and satisfactionwere measured at baseline (T0), immediately after the intervention (T1), and 1 month later (T2). Datacollection was conducted from July 30, 2019, to December 30, 2019. Results: The intention-to-treat analysis The intention-to-treat analysis of between groups at 1-monthfollow-up revealed that the intervention group had greater decreases in the fear of death (p=.002, 95% CI-2.53, -0.47; p<.001, 95% CI -3.61, -1.65) and death avoidance (p<.001, 95% CI -3.46, -1.84; p<.001, 95% CI-3.89, -2.43), had greater increases in the neutral acceptance (p=.032, 95% CI 0.05, 1.38; p<.001, 95% CI0.99, 2.56) and death competence (p<.001, 95% CI 4.10, 8.01; p<.001, 95% CI 7.80, 12.11) in patients andcaregivers, respectively. There were significant intergroup differences over time for patient well-being of(p<.001, 95% CI 3.06, 9.74) and satisfaction of (p<.001, 95% CI 2.01, 4.59). Results were consistent with theresults from the sensitivity analysis. Conclusion: This study demonstrated the feasibility and benefits of death education in hospitals andprovided an implementation plan for nursing professionals. Nurses should consider providing deatheducation for older adults with chronic diseases and their families to promote the development ofpalliative care and the quality of end-of-life.

The rationale and potential for using Lactobacillus in the management of periodontitis

        Wang Jiaqi,Liu Yingman,Wang Weiru,Ma Jiaojiao,Zhang Manman,Lu Xiaoying,Liu Jie,Kou Yurong 한국미생물학회 2022 The journal of microbiology Vol.60 No.4

Periodontitis refers to a wide range of the inflammatory conditions of supporting dental structures. For some patients with periodontitis, antibacterial agents are needed as an adjuvant to mechanical debridement treatments and oral hygiene maintenance. However, the widespread use of broad-spectrum antibiotics for the prophylaxis and treatment of periodontal infections results in the emergence of resistant pathogens. Therefore, probiotics have become markedly interesting to researchers as a potentially safe alternative to periodontal treatment and maintenance. Probiotics have been used in medicine for decades and extensively applied to the treatment of inflammatory diseases through the modulation of microbial synergy and other mechanisms. A growing amount of evidence has shown that using Lactobacillus strains for oral cavity maintenance could improve periodontal health. In this study, we reviewed studies showing proof of the inhibitory effects of Lactobacillus species on periodontal inflammation. We also explored the rationale and potential for using Lactobacillus species in the management of periodontitis.
