        BRCA1 Reversion Mutation Confers Resistance to Olaparib and Camrelizumab in a Patient with Breast Cancer Liver Metastasis

        Reversion mutations are associated with clinical resistance to poly(ADP-ribose) polymerase inhibitors (PARPi). Here, we describe the detection of a BRCA1 reversion mutation in a 39-year-old woman with metastatic breast cancer harboring a heterozygous germline BRCA1 exons 7–8 deletion who received PARPi olaparib combined with immune checkpoint inhibitor camrelizumab as third-line therapy. During progression from the olaparib and camrelizumab combination therapy, we identified via genomic sequencing a novel 7-base pair somatic deletion in BRCA1 (c.617_623delACAAATC). Sequence analyses indicated that this mutation realigned the reading frame of BRCA1, which potentially led to the reversal of its normal function and conferred resistance to PARPi.

        Multi-layers grid environment modeling for nuclear facilities: A virtual simulation-based exploration of dose assessment and dose optimization

        Dose optimization for Radioactive Occupational Personal (ROP) is an important subject in nuclear and radiation safety field. The geometric environment of a nuclear facility is complex and the work area is radioactive, so traditional navigation model and radioactive data field cannot form an effective environment model for dose assessment and dose optimization. The environment model directly affects dose assessment and indirectly affects dose optimization, this is an urgent problem needed to be solved. Therefore, this paper focuses on an environment model used for Dose Assessment and Dose Optimization (DA&DO). We designed a multi-layer radiation field coupling modeling method, and then explored the influence of the environment model to DA&DO by virtual simulation. Then, a simulation test is done, the multi-layer radiation field coupling model for nuclear facilities is demonstrated to be effective for dose assessment and dose optimization through the experiments and analysis.

        Construction of a 2D Co(II) Coordination Polymer with (4,4)-Connected Topology: Synthesis, Crystal Structure, and Surface Photo-electric Property

        A 2D grid-like (4, 4)-connected topology coordination polymer, [Co(BTA)2(H2O)2]n (1), where HBTA = 2- (1H-benzotriazol-1-yl)acetic acid, has been synthesized by hydrothermal method and characterized by single crystal X-ray diffraction, IR spectroscopy, elemental analysis and surface photovoltage spectroscopy (SPS). X-ray diffraction analyses indicated that 1 displays octahedral metal centers with secondary building units (SBUs) [Co(BTA)2(H2O)2] bridged by the BTA− ligands. In the crystal, the 2D supramolecular architecture is further supported by O–H···O, O–H···N, C–H···O hydrogen bonds and π···π stacking interactions. The SPS of polymer 1 indicates that there are positive response bands in the range of 300–600 nm showing photo-electric conversion properties. There are good relationships between SPS and UV–Vis spectra.

        Silencing of ABCG2 by MicroRNA-3163 Inhibits Multidrug Resistance in Retinoblastoma Cancer Stem Cells

        To investigate the function and regulation mechanism of ATP-binding cassette, subfamily G, member 2 (ABCG2) in retinoblastoma cancer stem cells (RCSCs), a long-term culture of RCSCs from WERI-Rb1 cell line was successfully established based on the high expression level of ABCG2 on the surface of RCSCs. To further explore the molecular mechanism of ABCG2 on RCSCs, a microRNA that specifically targets ABCG2 was predicted. Subsequently, miR-3163 was selected and confirmed as the ABCG2-regulating microRNA. Overexpression of miR-3163 led to a significant decrease in ABCG2 expression. Additionally, ABCG2 loss-of-function induced anti-proliferation and apoptosis-promoting functions in RCSCs, and multidrug resistance to cisplatin, carboplatin, vincristine, doxorubicin, and etoposide was greatly improved in these cells. Our data suggest that miR- 3163 has a significant impact on ABCG2 expression and can influence proliferation, apoptosis, and drug resistance in RCSCs. This work may provide new therapeutic targets for retinoblastoma.

      • The study of Harvest System for Organic Perishable Vegetable in Plugs

        Vegetable is the indispensable food for the daily life of mankind. Due to the demand for increasing the quality of vegetable in Taiwan, the protected culture must be developed for cultivation of vegetable. There are two common approaches of cultivation, including seeding and plug transplant, and both approaches require much labor. By applying the Organic Perishable Vegetable in Plugs planted, the labor problem in the past for plug seeding, sprout cultivation, transplantation and harvesting could be improved. Besides, the yield and quality could be increased. However, the short of labor is still a major problem for harvesting. This study would develop a set of harvesting machinery that is applicable for Organic Perishable Vegetable in Plugs. This could reduce the time and labor for harvesting in fields, and reach the objectives of saving of labor and increasing the productivity.

        Acupuncture for ventilator-dependent patients at a hospital-based respiratory care center: A randomized controlled trial

        Background: In intensive care units, mechanical ventilation is an important therapy to help patients with dysp- nea. However, long-term ventilator dependence would consume huge medical resources and increase the risk of morbidity and mortality. The aim of the study was to examine the efficacy of the acupuncture combined with western medical care on ventilator parameters in ventilator-dependent patients. Methods: In this clinical trial, 80 ventilator-dependent patients aged 20 to 80 years old were randomly assigned to acupuncture group and control group in the respiratory care center (RCC) of Changhua Christian Hospital. Besides regular medical care and therapy, participants in the acupuncture group received acupuncture therapy at the same 17 acu-points for 20 minutes once a day, a total of 12 sessions. The ventilator parameters were recorded to evaluate the respiratory efficiency for all participants. The primary outcome was rapid shallow breathing index (RSBI), and secondary outcomes were respiratory rate (RR), tidal volume (TV) and ventilation per minute (MV). Results: Though there was no significant difference in the parameter between the acupuncture group and the control group, we found the trend of decreasing RSBI in the acupuncture group. In subgroup analyses, the mean of RSBI significantly decreased 16.02 (with the SD in 60.84) in acupuncture group, while it increased 17.84 (with the SD in 39.38) in control group (p = 0.036) after 12 sessions. Conclusion: Acupuncture treatment can improve breathing ability of patients with respirator dependence in respiratory care center.

        Study of the H2O/Al2O3 Interface and the Acting Mechanism of Water in the Working Electrolyte

        Using a working electrolyte containing mixed solvents of ethylene glycol and N,N-dimethylformamide, this paper presentsa study of the reactions on the H2O/Al2O3 interface with sum frequency vibrational spectroscopy and the effects of differentwater content on the performance of the working electrolyte and an aluminum electrolytic capacitor and summarizes the rulesof the variations in the performance parameters of the working electrolyte and aluminum electrolytic capacitor with respect tothe water content. The results demonstrate that, when the water content is increased from 2.5 to 15%, the conductivity of theworking electrolyte increased by 930 μS/cm, and the sparking voltage decreased by 27 V. Also, the increased water contentcauses lower oxidation efficiency and lower thermal stability. The leakage current of the aluminum electrolytic capacitorafter high-temperature storage increases with an increase in the water content, and the attenuation rate of capacitor’s thelow-temperature capacitance decreases with an increase in the water content.

        Roles of NMDA NR2B Subtype Receptor in Prefrontal Long-Term Potentiation and Contextual Fear Memory

        <P><B>Summary</B></P><P>Cortical plasticity is thought to be important for the establishment, consolidation, and retrieval of permanent memory. Hippocampal long-term potentiation (LTP), a cellular mechanism of learning and memory, requires the activation of glutamate N-methyl-D-aspartate (NMDA) receptors. In particular, it has been suggested that NR2A-containing NMDA receptors are involved in LTP induction, whereas NR2B-containing receptors are involved in LTD induction in the hippocampus. However, LTP in the prefrontal cortex is less well characterized than in the hippocampus. Here we report that the activation of the NR2B and NR2A subunits of the NMDA receptor is critical for the induction of cingulate LTP, regardless of the induction protocol. Furthermore, pharmacological or genetic blockade of the NR2B subunit in the cingulate cortex impaired the formation of early contextual fear memory. Our results demonstrate that the NR2B subunit of the NMDA receptor in the prefrontal cortex is critically involved in both LTP and contextual memory.</P>

        중국 C2C 소셜커머스 플랫폼에서 판매자와 브랜드 신뢰가 지속적 구매의도에 미치는 영향

        Purpose - This study analyzed the correlation and influence between social support, trust (seller, brand), and continuous purchase intention in C2C social commerce in China. Design/methodology/approach - To test the hypothesis, SPSS and Smart PLS 3.0 statistical packages were used based on the collected data. Findings - First, it was confirmed that social support (emotional support, informational support) had a positive effect on trust in sellers. Second, it was found that trust in sellers had a positive effect on brand trust. Third, both seller trust and brand trust have a positive effect on consumers continuous purchase intention. Research implications or Originality - When consumers gain emotional and informational support from sellers, trust in sellers will be effectively improved. Companies wishing to improve brand credibility of their products will have to outsource the sale of their products to trusted sellers. The C2C social commerce platform should build its own trust rating system, recommend sellers with high reliability ratings, and encourage sellers to provide consumers with a lot of information about their brand.
