Background miRNAs play an important role in regulating normal animal development. Muscle tissue and fat metabolismare important for maintaining energy balance in animals. Yak has important agricultural and economic importance as itprovides milk, meat, and hair. It is used for transportation as well. However, the miRNA expression profiles of their muscleand adipose tissue are currently unknown. Objective To explore the regulatory roles of miRNAs in the skeletal muscle and adipose tissues of yak. Methods A total of 12 small RNA libraries were constructed from the skeletal muscle and adipose samples from yak aged 0.5,2.5, 4.5, and 7.5 years. High-throughput sequencing and bioinformatics analysis were used to determine the dynamic expressionprofile of miRNA, and a miRNA regulatory network related to muscle and adipose tissue development was established. Results miR-1-3p and miR-143-3p showed the highest expression during yak skeletal muscle and fat development, respectively. The MAPK and Ras signaling pathways were the pivotal pathways. miR-181-5p, miR-542-3p, and miR-424-5p mayhave key roles in skeletal muscle development, and CREBRF, GRB10, CDK1, RFX3, and EPC2 were the core target genes. While miR-127-5p, miR-379-3p, and miR-494-3p may play important regulatory roles in adipose deposition, and ETV1,XPO7, and C5AR2 were the core target genes. Conclusion This study provides valuable resources for further study of the molecular mechanisms underlying yak skeletalmuscle and adipose tissue development, and also a basis for studying the interactions between genes and miRNAs.

TIM-1 (also known as KIM-1 and HAVcr-1) is a type I transmembrane glycoprotein member of the TIM family that may play important roles in innate and adaptive immune responses. The overexpression of proteins associated with membrane proteins is a major obstacle to overcome in studies of membrane protein structures and functions. In this study, we successfully coupled the overexpression of the TIM-1 protein with a C-terminal enhanced green fluorescent protein (GFP) tag in Escherichia coli. To the best of our knowledge, this report is the first to describe the overexpression of human TIM-1 in E. coli. The purified TIM-1-EGFP fusion protein recognized and bound directly to apoptotic cells and did not to bind to viable cells. Furthermore, we confirmed that the interactions of TIM-1-EGFP with apoptotic cells were blocked by TIM-1-Fc fusion proteins. This fusion protein represents a readily obtainable source of biologically active TIM-1 that may prove useful in future studies of human TIM-1.

Tetrandrine (Tet), a bisbenzylisoquinoline alkaloid, has been reported to have a radiosensitization effect on tumors. However, its effects on human glioma and the specific molecular mechanisms of these effects remain unknown. In this study, we demonstrated that Tet has a radiosensitization effect on human glioma cells. It has been hypothesized that Tet has a radiosensitization effect on glioma cells by affecting the glioma cell cycle and DNA repair mechanism and that ERK mediates these activities. Therefore, we conducted detailed analyses of the effects of Tet on the cell cycle by performing flow cytometric analysis and on DNA repair by detecting the expression of phosphorylated H2AX by immunofluorescence. We used western blot analysis to investigate the role of ERK in the effect of Tet on the cell cycle and DNA repair. The results revealed that Tet exerts its radiosensitization effect on glioma cells by inhibiting proliferation and decreasing the expression of phosphorylated ERK and its downstream proteins. In summary, our data indicate that ERK is involved in Tet-induced radiosensitization of glioma cells via inhibition of glioma cell proliferation or of the cell cycle at G0/G1 phase.

Background: Psoriasis is an autoimmune disease that is caused by a shift in the Th1/Th2 balance toward Th1- dominant immunity. It has been established as an effective treatment to counteract psoriasis by subcutaneous injection of recombinant interleukin (IL)-4, and IL-4 gene therapy by topical transdermal penetration has shown its antipsoriatic effect in mice. Retinoic acid (RA) and dimethylsulfoxide can increase the efficiency of gene transfection in the topical transdermal delivery system. Objective: We investigated whether RA could improve anti-psoriasis efficiency using IL-4 expression plasmid pORF-mIL-4 (pIL-4) via transdermal delivery system in K14-vascular endothelial growth (K14- VEGF) factor transgenic mice. Methods: After pretreatment with RA, plasmid pIL-4 in 10% dimethylsulfoxide was applied to the ear skin by topical transdermal penetration. Hematoxylin- eosin staining and immunohistochemistry were performed with ear samples to evaluate anti-psoriasis efficiency in mice. Results: The psoriasis pathological features were relieved and psoriasis-associated factors were significantly reduced. Conclusion: Our results reveal that topical application of pIL-4 in dimethylsulfoxide by transdermal delivery with RA pretreatment can improve psoriasis significantly. Fruit quality traits like heat, color and flavor are unique and important for pepper cuisines and industrial use. Pepper pigments include chlorophyll, carotenoids, anthocyanin and red pigments like capsanthin are unique carotenoids to capsicum spp. So developing carotenoids-rich peppers and use of red pigments extraction along with capsaicinoids are one of interest pepper breeding goals in the world. Horticultural traits of 113 germplasms introduced by international cooperative research with YASS of China were evaluated to select promissing materials for high quality peppers during the past 3 years. All of germplasms including 60 local peppers were belong to C. annuum and fruit characteristics were diverse. Especially fruit chemical compositions like ASTA color, capsaicinoids and sugars were evaluated compared with korean commercial peppers. Average contents of ASTA color was 98±37, 55±63mg in capsaicinoids and 12±6% in total sugars. ASTA color which generally means red pigments content was over 120 in 25 germplasms including over 200 in 2 germplasms. Higher ASTA colors were observed in different fruit types. Capsaicinoids content was over 90mg/100g in 24 germplasms including 5 germpalsms over 200mg. Especially higher capsaicinoids were higher in local peppers with round and wrinkled fruit shape. And total sugar content was over 20% in 10 germplasms. Germplasms of different fruit shapes and characteristics will be useful materials for diverse fruit quality breeding.

The effects of MgO loading and calcination temperature of the catalysts on the carbon dioxide reforming of methane (CDR) have been extensively investigated over Ni/MgO/Al2O3 catalysts in a fixed-bed reactor system. Results indicate that the activity and stability of the catalysts were significantly influenced by both MgO loading and calcination temperature, i.e., higher calcination temperature and higher MgO loading are essential for high activity and stability of the catalyst. Characterization results indicate that mixed spinel phase composed of MgAl2O4 and NiAl2O4 is effective to produce active and stable Ni species having strong Ni to support interaction resulting in active and stable catalytic performance in CDR.

A novel non-viral gene delivery system, Procationic-Liposome-Protamine-DNA complexes (PLPD) which could further adsorb transferrin on the surface as a targeting ligand to form Tf-PLPD, was prepared and characterized before and after lyophilization. The size distribution of Tf-PLPD was in the range of $240{\pm}12nm$ and the zeta potential was $-24.10{\pm}2.5mV$. The transfection efficiencies of PLPD and Tf-PLPD were $12.18{\pm}3.8\;and\;24.26{\pm}2.6mU\;{\beta}-galactosidase/mg$ protein respectively. The lyophilization and the presence of serum didn't affect the tansfectivities of PLPD or Tf-PLPD. Compared to $Lipofectamine^{TM}$ 2000 (Invitrogen, U.S.A.), the procationic liposomes had less cytotoxicity to cells. In summary the procationic lipoplex described here, combining the condensing effect of protamine and the targeting capability of Tf, was a perspective non-viral vector for gene delivery system.
