        Pazopanib, a Novel Multitargeted Kinase Inhibitor, Shows Potent <i>In Vitro</i> Antitumor Activity in Gastric Cancer Cell Lines with <i>FGFR2</i> Amplification

        <P>Pazopanib is an orally bioavailable, ATP-competitive, multitargeted tyrosine kinase inhibitor mainly targeting VEGFR2 and PDGFR tyrosine kinases, but the biologic sequences of pazopanib activities beyond antiangiogenesis are poorly defined. We used a panel of 38 gastric cancer cell lines to test the efficacy of pazopanib. In a growth inhibition assay, genomic changes indicated that pazopanib had differential effects on cell growth. Treatment of the KATO-III, OCUM-2M, SNU-16, and HSC-39 gastric cancer cell lines harboring <I>FGFR2</I> amplification with pazopanib resulted in marked decreases of cell survival with IC<SUB>50</SUB> in ranges of 0.1 to 2.0 μmol/L, whereas the same treatment of those cell lines without <I>FGFR2</I> amplification had no growth-inhibitory effects. In the ectopic FGFR2-expressing model, treatment with the indicated concentrations of pazopanib significantly inhibited cell growth and colony formation by FGFR2-expressing NIH 3T3 cells with wild-type (WT) <I>FGFR2</I> and mutant <I>FGFR2</I> (S252W). Pazopanib also selectively suppressed constitutive FGFR2 signaling and phosphorylation of downstream effectors. In cell-cycle analysis, <I>FGFR2</I>-amplified cells underwent cell-cycle arrest at the G<SUB>1</SUB>–S phase after pazopanib treatment, whereas there were no significant effects on cell-cycle progression in cells without <I>FGFR2</I> amplification treated with pazopanib. In addition, pazopanib increased a substantial fraction of sub-G<SUB>1</SUB> only in <I>FGFR2</I>-amplified cells. These findings show that the activation of FGFR2 signaling by amplification may be a critical mediator of cell proliferation in a small subset of gastric cancer patients and that pazopanib may provide genotype-correlated clinical benefits beyond the setting of highly vascular tumors. <I>Mol Cancer Ther; 13(11); 2527–36. ©2014 AACR</I>.</P>

        Distribution of Staphylococcus pseudintermedius in clinical cases of dogs

        Staphylococcus intermedius has been most common agent associated with dog diseases, such as pyoderma and otitis externa. Recently, S. schleiferi subsp. coagulans has been also detected in suppurative diseases of dog and the organism formerly known as S. intermedius is called as S intermedius group (SIG), consisting of S. intermedius, Staphylococcus pseudintermedius and Staphylococcus delphini. From a clinical point of view, S. pseudintermedius is emerged as the most significant species of the SIG. A total of 72 SIG tested were positive for the presence of SInuc gene specific to S. intermedius and 67 (93.1%) isolates were S. pseudintermedius and only 5 (6.9%) S. intermedius, differentiating on the basis of arginine dyhydrolase production. None of the strains was identified as S. schleiferi subsp. coagulans. As results, S. pseudintermedius was found to be dominant in SIG from dogs in Jeju.

        Colorectal adenocarcinoma‐derived EGFR mutants are oncogenic and sensitive to EGFR‐targeted monoclonal antibodies, cetuximab and panitumumab

        <P>Somatic mutations of epidermal growth factor receptor (EGFR) occur in ~3% of colorectal cancer (CRC) patients. Here, through systematic functional screening of 21 recurrent EGFR mutations selected from public data sets, we show that 11 colon cancer‐derived EGFR mutants (G63R, E114K, R165Q, R222C, S492R, P596L, K708R, E709K, G719S, G724S and L858R) are oncogenic and able to transform cells in a ligand‐independent manner. We demonstrate that cellular transformation by these mutants requires receptor dimerization. Importantly, the EGF‐induced and constitutive oncogenic potential of these EGFR mutants are inhibited by cetuximab or panitumumab <I>in vivo</I> and <I>in vitro</I>. Taken together, we propose that a subset of EGFR mutations can serve as genomic predictors for response to anti‐EGFR antibodies and that metastatic CRC patients with such mutations may benefit from these drugs as part of the first‐line therapy.</P>

        Evaluation of Lactobacillus plantarum KCTC 3928 in Fermentation of Korean Soybean Paste (Doenjang)

        Lactobacillus plantarum KCTC 3928 was evaluated for use in Doenjang fermentation with four-fold approaches. Firstly, amino acids-profile of L. plantarum KCTC 3928 was investigated using MRS growth medium and found that this strain was the arginine pathway-deficient strain. Secondly, cell growth of L. plantarum KCTC 3928 in the sterilized Doenjang was carried out to determine susceptibility to inhibition by high concentration of salt (13.2%) in Doenjang. Thirdly, optimum addition ratio of L. plantarum KCTC 3928 and Doenjang upon investigation were 12.5 mg of cells and 10 g of Doenjang. Finally, conversion efficiency from glycoside isoflavone to aglycone isoflavone in Doenjang by L. plantarum KCTC 3928 was measured. The results indicate that L. plantarum KCTC 3928 can be used for low-salt Doenjang and Cheonggukjang fermentations.

      • User Experience of Bicycle Riders in Ulsan Grand Park

        Objective: In order to develop an E-bike system for Ulsan Grand Park, the user experience of an existing bicycle system was examined. Background: Within near future, it is expected that more and more people start to use e-bikes. In this regards, Ulsan Grand Park is a good place to test the usability of an e-bike system with number of bike users. Through practical user research, probable directions can be suggested in order to prepare the e-bike market expansion. Method: The researchers selected four user research methods from IDEO Method cards’ four different categories respectively; Learn, Look, Ask and Try. These methods were efficiently combined and applied to the user research process. Also the experience of traditional bike user and e-bike users were compared. Results: Through the user experience analysis, both advantages and disadvantages of the e-bike system in Ulsan Grand Park were identified. Also traditional bike users and e-bike users displayed different experiences and needs. Conclusion: The results imply a useful insight to develop a competitive e-bike system and activate its usage in terms of usability. Especially developing an e-bike system requires different perspectives and novel approaches compared to traditional bike-systems. Application: The results of this research provide a direction to vitalize the usage of e-bikes. In addition to that, the results assist people to build a guideline when they try to develop an e-bike system for a much wider area.

      • Spatial and temporal variations in nutrient and chlorophyll-<i>a</i> concentrations in the northern East China Sea surrounding Cheju Island

        <P><B>Abstract</B></P><P>Nutrients, chlorophyll-<I>a</I> (Chl<I>-a</I>), and environmental conditions were extensively investigated in the northern East China Sea (ECS) near Cheju Island during five research cruises from 2003 to 2007. In the eastern part of the study area, surface waters were characterized only by the Tsushima Current Water (TCW) during all five cruises. However, the western surface waters changed with season and were characterized by the Yellow Sea Cold Water (YSCW) in spring, the Changjiang Diluted Water (CDW) in summer, and the Yellow Sea Mixed Water (YSMW) in autumn. In spring and autumn, relatively high concentrations of nitrate and phosphate were observed in the surface waters in the western part of the study area, where vertical mixing brought large supplies of nutrients from deep waters. Changes in wind direction occasionally varied the inflow of the Changjiang plume in summer, clearly causing the annual variation in surface nitrate and phosphate concentrations in summer. In summer, the surface distribution of nitrate and phosphate did not coincide with that of silicate in the study area, which probably resulted from the significant drop in the Si:N ratio in the Changjiang plume since construction of the Three Gorges Dam (TGD). Despite large temporal and spatial variations in surface Chl-<I>a</I> concentrations, depth-integrated Chl-<I>a</I> concentrations exhibited little variation temporally and spatially. In the study area, surface Chl-<I>a</I> concentration did not well reflect the standing stocks of phytoplankton. The vertical distribution of Chl-<I>a</I> showed large temporal and spatial variations, and the main factor controlling the vertical distribution of Chl-<I>a</I> in summer was the availability of nitrate. The thermohaline front may play an important role for accumulation of phytoplankton biomass in spring and autumn.</P>

        Characteristics of a large outbreak arising from a school field trip after COVID-19 restrictions were eased in 2022

        Objectives: This study analyzed a large outbreak of coronavirus disease 2019 (COVID-19) that occurred during a high school field trip in the Jeonbuk region and aimed to identify risk factors for COVID-19 infection, with the goal of preventing such outbreaks in the future.Methods: A retrospective cohort study of 737 participants, including 668 students and 69 staff at High School A, was designed to describe the epidemiological characteristics of this large COVID-19 outbreak. Logistic regression analysis was performed to calculate relative risks (odds ratios [ORs]) and 95% confidence intervals (CIs).Results: There were 190 confirmed cases (174 students, 16 staff), with an attack rate of 25.8%. Small outbreaks were decreasing before the field trip, but this trend reversed after the trip, leading to larger outbreaks. Logistic regression showed an OR of 2.39 (95% CI, 1.66–3.43; p<0.05) for COVID-19 infection among field trip participants. Among them, 11th graders had an OR of 2.32 (95% CI, 1.53–3.52; p<0.05) compared to 10th graders, while no significant risk difference was found within same-grade teams.Conclusion: There was a high risk for COVID-19 transmission during extracurricular activities with a large number of participants, such as field trips, even after the nationwide Omicron variant epidemic subsided. Even when students are separated into teams and follow different routes, it is challenging to design routes that entirely prevent contact between teams. Thus, programs should be designed carefully, and students with symptoms should be identified before and during the program to isolate them promptly.

        Experiences of Latent Tuberculosis Infection Treatment for the North Korean Refugees

        Background: Tuberculosis (TB) is increasing in immigrants. We aimed to investigate the current status of latent tuberculosis infection (LTBI) treatment for North Korean Refugees (NKR) compared to South Koreans Contacts (SKC). Methods: TB close contacts in a closed facility of SKC and NKR who underwent LTBI screening in a settlement support center for NKR were analyzed retrospectively. Results: Among tuberculin skin test (TST) ${\geq}10mm$ (n=298) reactors, the males accounted for 72.2% in SKC (n=126) and 19.5% in NKR (n=172) (p<0.01). The mean age was higher in South Korea ($42.8{\pm}9.9years$ vs. $35.4{\pm}10.0years$, p<0.01). Additionally, the mean TST size was significantly bigger in NKR ($17.39{\pm}3.9mm$ vs. $16.57{\pm}4.2mm$, p=0.03). The LTBI treatments were initiated for all screened NKR, and LTBI completion rate was only 68.0%. However, in NKR, LTBI treatment completion rate was significantly increased by shorter 4R regimen (odds ratio [OR], 9.296; 95% confidence interval [CI], 4.159-20.774; p<0.01) and male (OR, 3.447; 95% CI, 1.191-9.974; p=0.02). Conclusion: LTBI treatment compliance must be improved in NKR with a shorter regimen. In addition, a larger study regarding a focus on LTBI with easy access to related data for NKR should be conducted.

        A clinical pilot study of jawbone mineral density measured by the newly developed dual-energy cone-beam computed tomography method compared to calibrated multislice computed tomography

        Purpose: This clinical pilot study was performed to determine the effectiveness of dual-energy cone-beam computed tomography (DE-CBCT) in measuring bone mineral density (BMD). Materials and Methods: The BMD values obtained using DE-CBCT were compared to those obtained using calibrated multislice computed tomography (MSCT). After BMD calibration with specially designed phantoms, both DE-CBCT and MSCT scanning were performed in 15 adult dental patients. Three-dimensional (3D) Digital Imaging and Communications in Medicine data were imported into a dental software program, and the defined regions of interest (ROIs) on the 3-dimensional surface-rendered images were identified. The automatically-measured BMD values of the ROIs (g/㎤), the differences in the measured BMD values of the matched ROIs obtained by DE-CBCT and MSCT 3D images, and the correlation between the BMD values obtained by the 2 devices were statistically analyzed. Results: The mean BMD values of the ROIs for the 15 patients as assessed using DE-CBCT and MSCT were 1.09±0.07 g/㎤ and 1.13±0.08 g/㎤, respectively. The mean of the differences between the BMD values of the matched ROIs as assessed using DE-CBCT and calibrated MSCT images was 0.04±0.02 g/㎤. The Pearson correlation coefficient between the BMD values of DE-CBCT and MSCT images was 0.982 (r=0.982, P<0.001). Conclusion: The newly developed DE-CBCT technique could be used to measure jaw BMD in dentistry and may soon replace MSCT, which is expensive and requires special facilities.
