Functional Analysis of Tmie in the Inner Ear and Gene Therapy using tmie Transgenic Mice

The circling (cir/cir ) mouse is a murine model for human non‐syndromic deafness DFNB6. The causative gene is transmembrane inner ear (tmie), in which the mutation is a 40‐kilobase genomic deletion including tmie. The function of Tmie is unknown. To better understand the function of Tmie, we observed the spatiotemporal expression of tmie in the mouse cochlea using a Tmie‐specific antibody during postnatal inner ear development. Tmie was expressed in the cochlear hair cells of the mouse inner ear from embryonic days to adult. It is postulated that Tmie protein is involved in the hair cell structural formation and maturation before hearing onset (around P14), and maintenance of organ of Corti tissues after that. The cochlear hair cells of the circling mouse showed a basal‐to‐apical gradient of outer hair cell degeneration. The hair cell stereocilia bundles revealed the abnormal structure and it expanded to the apical region. In order to find the exact localization of Tmie protein inside the cell, we transfected the plasmids expressing GFP‐Tmie fusion protein into the HEI‐OC1 auditory hair cells. Tmie protein was colocalized with Calnexin (Canx), ER marker protein, but not with beta‐COP, Golgi marker protein. We next produced the Myo7a promoterdirected tmie expression transgenic mice to induce the phenotypic rescue of circling mice in a gene therapeutic way. Some circling mice with tmie transgene showed the normal behavior and hearing ability. These results indicate that tmie has a critical role in the inner ear development and hearing ability in the mice.
담론분석과 정치경제학의 조우 가능성에 대한 탐색적 연구

이 논문은 질적, 비판적, 해석학적 커뮤니케이션 연구에 있어 담론분석이 비언어적 요소, 특히 구조와 맥락, 사회적 총체성 등을 포괄하는 구조적ㆍ산업적 분석이나 미디어 정치경제학과 맺는 유기적 관계, 나아가 양자 간의 유사한 인식론 및 문제의식을 밝혀, 향후 비판 커뮤니케이션 및 미디어 문화연구의 방법론적 논의를 확장하고자 하는 의도를 지닌다. 이를 위해 푸코의 담론이론을 포함한 비판적 담론분석의 다양한 해석 및 지류에 대해 논의하였다. 또한 이러한 방법론적 전통이 포착하고 있는 담론이 담론 아닌 것들과 맺는 관계, 이를테면 언어와 상징작용이 미디어 정치경제학의 주된 관심사인 구조와 물질, 생산과 제도 등과 맺는 교호적 관계에 대해 방법론은 물론 인식론적 견지에서도 논의하고, 이들이 창발적으로 접합할 수 있는 가능성 및 커뮤니케이션 연구의 질적 방법론으로서 활용 가능한 방법에 대해 메타 비평적 차원에서 논의하였다. The purpose of this essay is to expand the scale and scope of the methodological considerations of qualitative, critical, and hermeneutic communication studies and media-oriented cultural studies by collaborating discourse analysis with nonlinguistic elements, especially structural - and/or political economic analysis emphasizing the structure, context, social totality, and the like. For this, a wide variety of pertinent renditions and implications of critical discourse analysis including the Foucauldian discourse theory were discussed. Futhermore the organic relationship between these seemingly antagonistic perspectives, that is to say, discursive with non-discursive criteria pursued by such a methodological tradition was carefully examined. I contend that there is high potential of collaborative and interchangeable relationship between discourse analysis and media political economy, focusing on the structure, material, production, and institution, both in methodological and epistemological level and the possibility of their creative and constructive articulation, despite their notable differences.

이 논문은 한류 현상으로 대변되는 한국 대중문화의 미디어 문화연구적 입장에서 미디어 텍스트나 콘텐츠, 혹은 수용자 중심주의에서 도출되는 문화의 흔종성, 창의적이고 저항적인 텍스트 읽기의 편재성에 보다 섬세한 결을 더하여 논의를 확장하기 위한 의도를 가지고 있다. 그 하나가 문화현상을 이해하는 분석의 엄정성과 총체성을 담보하기 위해 기존의 텍스트나 수용자에 중심을 둔 문화주의적 접근에 비판적 정치경제학(critical political economy) 시각의 접합이다. 가령 한류 현상을 분석하는 흔종성 개념의 유용함과 의의를 이전의 단선적인 시각들로부터 차별화하고 문화연구의 전통 내에서 그 장점과 공헌을 부각시키는 동시에, 그것이 만족스럽게 설명하기 어려운 부분에 대한 보완을 위한 하나의 틀로서 비판적 정치경제학 패러다임의 복원에 대한 이론적이며 방법론적 요청에 대해 논구하고 있다. 더 나아가 수용자의 문화적인 상상과 수용과정 등을 강조하는 미디어 문화연구가 초국적 문화자본이나 국가 그리고 문화산업 의 역할과 그들간의 상호작용을 다양한 층위에서의 접합을 통해 보다 비판적이며 동시에 실천적인 문화연구의 본래적 기능에 다가갈 수 있는 가능성에 대해 모색하고자 하였다. The purpose of this essay is to expand the scope and horizon of the discourse surrounding a popular cultural phenomenon, Korean Wave, especially by complementing partial perspectives emphasizing the hybridization thesis and creative reading of the text, often found in media-oriented cultural studies. My central contention is to incorporate the critical political economy perspective with the culturalist approach focusing on text or the audiences. For example, I highlight the virtue of hybridity concept in analyzing the Korean Wave, and how it differs from more linear models. At the same time, I call for more astute attention to critical political economy paradigm as a useful framework to supplement some blind spots found in the hybridization thesis. Futhermore, I suggest a potential and progressive node for more critical, practical and holistic cultural studies by articulating the role of transnational cultural capital, state, and culture industry, and mutual relationship among them.
