        Bioabsorbable bone plates enabled with local, sustained delivery of alendronate for bone regeneration

        <P>We prepared a bone plate enabled with the local, sustained release of alendronate, which is a drug known to inhibit osteoclast-mediated bone resorption and also expedite the bone-remodeling activity of osteoblasts. For this, we coated a bone plate already in clinical use (PLT-1031, Inion, Finland) with a blend of alendronate and a biocompatible polymer, azidobenzoic acid-modified chitosan (i.e., Az-CH) photo-crosslinked by UV irradiation. As we performed the in vitro drug release study, the drug was released from the coating at an average rate of 4.03 mu g/day for 63 days in a sustained manner. To examine the effect on bone regeneration, the plate was fixed on an 8 mm cranial critical size defect in living rats and the newly formed bone volume was quantitatively evaluated by micro-computed tomography (micro-CT) at scheduled times over 8 weeks. At week 8, the group implanted with the plate enabled with sustained delivery of alendronate showed a significantly higher volume of newly formed bone (52.78 +/- 6.84%) than the groups implanted with the plates without drug (23.6 +/- 3.81%) (p < 0.05). The plate enabled with alendronate delivery also exhibited good biocompatibility on H&E staining, which was comparable to the Inion plate already in clinical use. Therefore, we suggest that a bone plate enabled with local, sustained delivery of alendronate can be a promising system with the combined functionality of bone fixation and its expedited repair.         <P>The frequent economic losses incurred with H9N2 low pathogenic avian influenza viruses (LPAI) infection have raised serious concerns for the poultry industry. A 1-dose regimen with inactivated H9N2 LPAI vaccine could not prevent vaccinated poultry from becoming infected and from shedding wild viruses. A study was conducted to determine whether a 2-dose regimen of inactivated H9N2 LPAI vaccine could enhance the immunologic response in chickens. Such gel-primed and mineral oil-boosted regimen has produced encouraging results associated with improved immune responses to an H9N2 LPAI. This strategy could be cost effective and helpful for preventing avian influenza virus in the poultry industry.</P>

        ASXL2 promotes proliferation of breast cancer cells by linking ERα to histone methylation

        <P>Estrogen receptor alpha (ER alpha) has a pivotal role in breast carcinogenesis by associating with various cellular factors. Selective expression of additional sex comb-like 2 (ASXL2) in ER alpha-positive breast cancer cells prompted us to investigate its role in chromatin modification required for ERa activation and breast carcinogenesis. Here, we observed that ASXL2 interacts with ligand E2-bound ERa and mediates ERa activation. Chromatin immunoprecipitation-sequencing analysis supports a positive role of ASXL2 at ERa target gene promoters. ASXL2 forms a complex with histone methylation modifiers including LSD1, UTX and MLL2, which all are recruited to the E2-responsive genes via ASXL2 and regulate methylations at histone H3 lysine 4, 9 and 27. The preferential binding of the PHD finger of ASXL2 to the dimethylated H3 lysine 4 may account for its requirement for ERa activation. On ASXL2 depletion, the proliferative potential of MCF7 cells and tumor size of xenograft mice decreased. Together with our finding on the higher ASXL2 expression in ERa-positive patients, we propose that ASXL2 could be a novel prognostic marker in breast cancer.</P>

        포장방법과 저장조건이 돈육 등심의 저장성 및 보수력에 미치는 영향

        To investigate the effects of packaging method and storage condition on meat quality, a total of eight pork loins were selected from a commercial meat plant at 24 hr postmortem. The loins were cut to 2.5㎝ thick steaks and were randomly assigned to the five treatment combination; vacuum packaging and submersion in chilled salt water(SCSW)(T1), non-vacuum packaging and submersion in chilled salt water (T2), vacuum packaging and storage in conventional refrigerator(SCR)(T3), non-vacuum packaging and storage in conventional refrigerator(T4), and wrap packaging and storage in conventional refrigerator(TS). All samples were stored at 0℃ for 20 days to measure color of meat(CIE L*, a*. b*), pH, drip loss, purge loss %, lipid oxidation (TBARS; thiobarbituric acid reactive substances) and total plate counts (TPC). CIE L* and b* value of SCSW(T1, 2) samples were significantly(P$lt;0.05) lower than SCR samples when they were measured at storage 20 days. The sample of SCSW(T1, 2) showed significantly(P$lt;0.05) lower pH than those of SCR(T3, 4, 5) during storage. The sample of SCSW(T1, 2) showed significantly(P$lt;0.05) higher Drip loss than those of SCR(T3, 4, 5) during 5 days storage. T2 showed the lowest purge loss % whereas T3 remarked the highest purge loss %. The samples of SCSW showed significantly(P$lt;0.05) lower TBARS than those of SCR during storage. TPC was samples of SCSW(T1, 2) showed significantly(P$lt;0.05) lower than other treatments. These results suggested that NVP(non-vacuum packaging) and SCSW could reduce purge loss and discoloration, and extend shelf-life of pork loin.

      • Working gas effect on properties of Al<sub>2</sub>O<sub>3</sub> film in plasma-enhanced atomic layer deposition

        <P><B>Abstract</B></P> <P>The effect of working gas on the properties of Al<SUB>2</SUB>O<SUB>3</SUB> films and SiO<SUB>x</SUB> interlayers was investigated in direct-type plasma-enhanced atomic layer deposition. The density of the Al<SUB>2</SUB>O<SUB>3</SUB> film was higher for Ar/O<SUB>2</SUB> plasma than for He/O<SUB>2</SUB> plasma, whereas the thicknesses of the Al<SUB>2</SUB>O<SUB>3</SUB> film and SiO<SUB>x</SUB> interlayer were greater for He/O<SUB>2</SUB> plasma than for Ar/O<SUB>2</SUB> plasma. For understanding these phenomena, the amounts of C– and H-containing impurities in the deposited Al<SUB>2</SUB>O<SUB>3</SUB> film were evaluated by using X-ray photoelectron spectroscopy (XPS). Further, differences between the plasma properties in He/O<SUB>2</SUB> and Ar/O<SUB>2</SUB> were analyzed using optical emission spectroscopy (OES); the consumption rate of O radicals and the production rate of H radicals were estimated from the time-resolved emission intensities for the O I and H<SUB>α</SUB> lines, respectively.       • 왕제 안티모니광상의 천열수 광화작용 : 광물 및 지구과학

        왕제 안티모니광상은 백악기 천부 관입암 복합채(역둔화강암체)의 영향을 받은 소규모의 탄산염암 교대형 천열수 광상이다. 광체는 풍촌석회암층의 층리를 교대하여 발달된 망토(manto) 또는 침치 또는 산점-망상세맥상으로 산출된다. 열수변질 및 광화작용을 받은 모암에는 Ag (0.1-0.6 ppm), As(34-241 ppm), Au(0.3-1.0 ppm), Cu(15-48 ppm), Pb(75-650 ppm), Sb(56->9,999 ppm) 및 Zn(331-370 ppm) 등이 부화되어 있다. 광화작용은 견운모 및 점토광물화 작용과 함께 단일 시기에 있었으며, 광석광물로는 자철석, 마그헤마이트, 황철석, 유비철석, 섬아연석, 황동석, 휘안석 등이 산출된다. 광화유체는 전반적으로 단순한 저염농도의 NaCh-H₂O 계의 것이었으나, CO₂- H₂O 계 유체의 일시적인 혼입이 있었다. 유체포유물의 균일화온도와 NaCl상당 염농도는 각각 164-246?Cdhk ??, ?? 값은 각각 6.2~8.6‰, 4.1~8.1‰alc -4.9~-9.1‰이었다. 따라서 광화작용의 초기에는 마그마 기원의 열수에 의한 광화작용이 진행되었으나, 후기로 가며 퇴적암 기원의 유체 또는 천수의 영향을 강하게 받은 것으로 보인다. 모암의 산소 및 탄소 안정동위원소비는 각각 12.1~20.0‰(SMOW)및 0.0~-2.8‰(PDB)이며, 광화대 및 열수변질대로 가면서 모두 가벼워지는 경향을 갖는다. 광상의 산출상태, 광석과 광물 및 생성조건으로 볼 때, 둔전-백전 지역에는 역둔화강암체의 중심에서 외각부로 가며 Au ->Au-Ag-> Au-Ag-Sb->Sb 광체순으로 수직적 및 수평적 대상분포가 인지된다. The Wangje antimony deposit, a small carbonate-hosted replacement deposit, was formed as a result of epithermal process that was associated with a shallow-seated Cretaceous Yeogdun granitoid intrusion. Mamo-to-chimney shaped orebodies were formed by the selective replacement of thin-bedded white grey limestone beds of the Cambrian Pungchon Limestone Formation. The ores occur as lens, patches and stringer-stockworks preferentially along the contact with coarse-grained calcite. Altered and/or mineralized host rock is enriched with Ag (0.1 to 0.6 ppm). As (34 to 241 ppm), Au (0.3 to 1,0 ppm), Cu (15 to 48 ppm), Pb (75 to 650 ppm). Sb (564 to >9,999 ppm) and Zn (331 to 370 ppm). The mineralization occurred within a single stage, consisting of ore minerals such as magnetite, magheinile, pyriic, arsenopyrite, sphalerite, chalcopyrite and Stibnite, and is associated with sericitic and argillic alteration. Fluid inclusion studies reveal that ore fluids were low saline with a simple NaCl-H2O system, but the temporary mixing with CO:-H;0 fluids occurred in the later period. Measured inclusion data indicate the homogenization temperatures and salinities of fluids are 164 to 246 C and 0.0 to 2.6 NaCI equivalent wt.%, respectively. The δ34SH2S values of hydrothennal fluid responsible for the deposition of sulfides such as pyrite and stibnile are calculated 10 be 6.2 to 8.6. The δ18H2o and δ13 Cco2 values for deposition of vein calcites range from 4.1 to 8.1. and -4.9 to 9.l. respectively. These stable isotope value are interpreted to indicate that ore fluids during the early period were derived from hydrothennal water of igneous origin, whereas the later fluids were highly influenced by mixing with oxidizing carbonatic and meteoric water. The oxygen and carbon isotope values of host rocks (sedimentary carbonates) are 12.1 to 20.0%,, vs. SMOW and 0.0 to -2.8%,, vs. PDB, respectively, and become progressively lighter toward the front of mineralized zone. Based upon the occurrence of orebodies, along with the mineralogy and depositional condition, a vertical and lateral orebody zoning as follows is recognized in the Dunjeon-Baegjeon mine area from the Yeogdun granitic complex toward the peripheral sites, Au→Au-Ag→Au-Ag-Sb→Sb ores.
