      • Characterization of an activation-tagged mutant with delayed flower abscission in Arabidopsis

        Toward molecular understanding of flower senescence/abscission, we have identified a mutant, designated as dea1-1D (dealyed abscission1-1D), with delayed flower senescence/abscission syndrome from activation-tagged pools. Phenotypic analysis revealed pleiotropic effects of dea1-1D mutation including delayed flowering as well as smaller serrated leaves. Genetic analysis showed that it is a dominant mutation. Molecular analysis on the flower senescence syndrome indicated that dea1-1D might define novel regulatory branch of flower abscission, controlling expression of ethylene-responsive AP2 transcription factor. On the contrary, triple responses was not affected by dea1-1D mutant. Though the penetrance was not complete, the mutant phenoytpes was shown to be tightly linked with the T-DNA selection marker, BASTA-resistance. We identified the T-DNA insertion site through molecular cloning of the T-DNA flanking genomic DNA and found that a neighboring gene was overexpressed in the dea1-1D mutant. Together with gene expression analysis, we will discuss possible function of DEA1 during flower senescence and abscission.

      • Wound Healing Effects of <i> Prunus yedoensis</i> Matsumura Bark in Scalded Rats

        <P>Pruni Cortex has been used to treat asthma, measles, cough, urticaria, pruritus, and dermatitis in traditional Korean medicine. The objective of this study was to investigate the effects of<I> Prunus yedoensis</I> Matsumura bark methanol extract (PYE) on scald-induced dorsal skin wounds in rats. Scalds were produced in Sprague-Dawley rats with 100°C water and treated with 5% and 20% PYE (using Vaseline as a base), silver sulfadiazine (SSD), and Vaseline once a day for 21 days, beginning 24 hours after scald by treatment group allocation. The PYE-treated groups showed accelerated healing from 12 days after scald, demonstrated by rapid eschar exfoliation compared to the control and SSD groups. PYE-treated groups showed higher wound contraction rates and better tissue regeneration in comparison with the control group. Serum analysis showed that transforming growth factor beta 1 and vascular endothelial growth factor levels remained high or gradually increased up to day 14 in both PYE groups and then showed a sharp decline by day 21, implying successful completion of the inflammatory phase and initiation of tissue regeneration. These findings suggested that PYE is effective in promoting scald wound healing in the inflammation and tissue proliferation stages.</P>

        <P>Nanostar and nanonetwork crystals were prepared from fully conjugated poly(3-(2-ethylhexyl)thiophene)-<I>block</I>-polythiophene (P3EHT-<I>b</I>-PT) via a simple INCP process.       • Supramolecular Coordination Polymer Formed from Artificial Light-Harvesting Dendrimer

        <P>We report the formation of supramolecular coordination polymers formed from multiporphyrin dendrimers (<B>P</B><SUB><B>Zn</B></SUB><B>P</B><SUB><B>M</B></SUB>; <B>M</B> = <B>FB</B> or <B>Cu</B>), composed of the focal freebase porphyrin (<B>P</B><SUB><B>FB</B></SUB>) or cupper porphyrin (<B>P</B><SUB><B>Cu</B></SUB>) with eight zinc porphyrin (<B>P</B><SUB><B>Zn</B></SUB>) wings, and multipyridyl porphyrins (<B>PyP</B><SUB><B>M</B></SUB>; <B>M</B> = <B>FB</B> or <B>Cu</B>), <B>P</B><SUB><B>FB</B></SUB> or <B>P</B><SUB><B>Cu</B></SUB> with eight pyridyl groups, through multiple axial coordination interactions of pyridyl groups to <B>P</B><SUB><B>Zn</B></SUB>s. UV–vis absorption spectra were recorded upon titration of <B>PyP</B><SUB><B>FB</B></SUB> to <B>P</B><SUB><B>Zn</B></SUB><B>P</B><SUB><B>FB</B></SUB>. Differential spectra, obtained by subtracting the absorption of <B>P</B><SUB><B>Zn</B></SUB><B>P</B><SUB><B>FB</B></SUB> without guest addition as well as the absorption of <B>PyP</B><SUB><B>FB</B></SUB>, exhibited clear isosbestic points with saturation binding at 1 equiv addition of <B>PyP</B><SUB><B>FB</B></SUB> to <B>P</B><SUB><B>Zn</B></SUB><B>P</B><SUB><B>FB</B></SUB>. Job’s plot analysis also indicated 1:1 stoichiometry for the saturation binding. The apparent association constant between <B>P</B><SUB><B>Zn</B></SUB><B>P</B><SUB><B>FB</B></SUB> and <B>PyP</B><SUB><B>FB</B></SUB> (2.91 × 10<SUP>6</SUP> M<SUP>–1</SUP>), estimated by isothermal titration calorimetry, was high enough for fibrous assemblies to form at micromolar concentrations. The formation of a fibrous assembly from <B>P</B><SUB><B>Zn</B></SUB><B>P</B><SUB><B>FB</B></SUB> and <B>PyP</B><SUB><B>FB</B></SUB> was visualized by atomic force microscopy and transmission electron microscopy (TEM). When a 1:1 mixture solution of <B>P</B><SUB><B>Zn</B></SUB><B>P</B><SUB><B>FB</B></SUB> and <B>PyP</B><SUB><B>FB</B></SUB> (20 μM) in toluene was cast onto mica, fibrous assemblies with regular height (ca. 2 nm) were observed. TEM images obtained from 1:1 mixture solution of <B>P</B><SUB><B>Zn</B></SUB><B>P</B><SUB><B>FB</B></SUB> and <B>PyP</B><SUB><B>FB</B></SUB> (0.1 wt %) in toluene clearly showed the formation of nanofibers with a regular diameter of ca. 6 nm. Fluorescence emission measurement of <B>P</B><SUB><B>Zn</B></SUB><B>P</B><SUB><B>M</B></SUB> indicated efficient intramolecular energy transfer from <B>P</B><SUB><B>Zn</B></SUB> to the focal <B>P</B><SUB><B>FB</B></SUB> or <B>P</B><SUB><B>Cu</B></SUB>. By the formation of supramolecular coordination polymers, the intramolecular energy transfer changed to intermolecular energy transfer from <B>P</B><SUB><B>Zn</B></SUB><B>P</B><SUB><B>M</B></SUB> to <B>PyP</B><SUB><B>M</B></SUB>. When the nonfluorescent <B>PyP</B><SUB><B>Cu</B></SUB> was titrated to fluorescent <B>P</B><SUB><B>Zn</B></SUB><B>P</B><SUB><B>FB</B></SUB>, fluorescence emission from the focal <B>P</B><SUB><B>FB</B></SUB> was gradually decreased.       • Vasorelaxant Effect of Osterici Radix Ethanol Extract on Rat Aortic Rings

        <P>The root of <I>Ostericum koreanum</I> Maximowicz has been used as a traditional medicine called “Kanghwal” in Korea (or “Qianghuo” in China). The purpose of this study was to investigate the vasorelaxant activity and mechanism of action of an ethanol extract of the <I>O. koreanum</I> root (EOK). We used isolated rat aortic rings to assess the effects of EOK on various vasorelaxant or vasoconstriction factors. EOK induced vasorelaxation in phenylephrine hydrochloride (PE) or KCl precontracted aortic rings in a concentration-dependent manner. However, the vasorelaxant effects of EOK on endothelium-intact aortic rings were reduced by pretreatment with <SMALL>L</SMALL>-NAME or methylene blue. In Ca<SUP>2+</SUP>-free Krebs-Henseleit solution, pretreatment with EOK (0.3 mg/mL) completely inhibited PE-induced constriction. In addition, EOK (0.3 mg/mL) also completely inhibited vasoconstriction induced by supplemental Ca<SUP>2+</SUP> in aortic rings that were precontracted with PE or KCl. Furthermore, the EOK-induced vasorelaxation in PE-contracted aortic rings was inhibited by preincubation with nifedipine. These results indicate that the vasorelaxant effects of EOK are responsible for the induction of NO formation from <SMALL>L</SMALL>-Arg and NO-cGMP pathways, blockage of the extracellular Ca<SUP>2+</SUP> entry via the receptor-operative Ca<SUP>2+</SUP> channel and voltage-dependent calcium channel, and blockage of sarcoplasmic reticulum Ca<SUP>2+</SUP> release via the inositol triphosphate pathway.</P>

        Cultural Contacts and Intercultural Sensitivity of KFL College Students in the US

        본 연구에서는 미국대학 KFL 학습자들의 한국 문화 접촉과 상호문화 감수성, 성취도의 상관관계를 살피고자 하였다. 이를 위해 82명의 초급 한국어 학습자를 대상으로 상호문화 감수성 조사 및 한국 문화 접촉 조사를 실시하고, 접촉경험과 상호문화 감수성의 상관관계, 접촉 경험 및 상호문화 감수성과 성취도의 상관관계를 분석하였다. 분석 결과, 미국 대학 KFL 학습자들은 비교적 높은 상호문화 감수성을 갖고 있었으며, 상호문화 감수성 조사의 다섯 가지 요인 중에서 `문화 차이에 대한 존중` 요인의 평균이 가장 높았고 `상호작용 자신감` 평균이 가장 낮았다. 한국인과의 접촉이 쉽지 않은 KFL 환경에서도 원어민과의 대화 경험의 유무는 학습자의 상호문화 감수성에 중요한 영향을 끼치고 있었으며, 특히 디지털 환경의 영향으로 소셜미디어를 통한 직접 접촉 역시 유의미한 상관을 보였다. 또한 대중매체는 학습자들에게 익숙한 한국 문화 접촉 방법이었으며, 학습자의 상호문화 감수성 및 성취도와 유의미한 상관관계를 보였다. 마지막으로, 모든 참가자는 대학에서의 한국어 강좌를 수강하였는데, 한국어 강좌 수강 및 한국어 학습 동료들과의 의사소통을 문화적 접촉으로 판단하는 학습자가 다수 있었던 반면 그렇지 않은 학습자들도 있었다. 한국어 수업 및 동료 학습자와의 의사소통에 대한 다른 인식을 가진 두 집단은 상호문화 감수성에서도 유의미한 차이를 보였다. 선행 연구에서 다루지 않았던 다양한 한국 문화 접촉 경험을 조사하고 상호문화 감수성과의 상관을 살폈다는 점, 성취도와의 상관관계를 일부 확인하였다는 점, 이를 통해 미국 대학의 KFL 학습자에 대한 이해를 높였다는 점에서 본고의 의의를 찾을 수 있을 것이다. The present study investigates the relationship among cultural contacts, intercultural sensitivity, and proficiency of US-based beginning level KFL college students. Data was collected from eighty two learners through the cultural contact survey and Intercultural Sensitivity Scale (Chen & Starostar, 2000). The results showed the following; 1) “meeting Koreans in person” and “chatting via social media” were popular ways to communicate with Korean native speakers and showed the relationship between cultural contact and intercultural sensitivity; 2) indirect contact via Korean popular media was a common way to be exposed to Korean culture. When compared by t-test, the high proficiency group and less proficiency group showed a statistically significant difference only in “Watching Korean dramas and TV shows.” This may indicate that being in contact with popular mainstream media such as K-dramas has a significant influence on learners` proficiency level; 3) the response to contact via Korean language course and communication with non-native peers in Korean class showed the students` beliefs about their thoughts on the connection between Korean culture through Korean course and their respective peers.         MFN1 deacetylation activates adaptive mitochondrial fusion and protects metabolically challenged mitochondria

        <P>Fasting and glucose shortage activate a metabolic switch that shifts more energy production to mitochondria. This metabolic adaptation ensures energy supply, but also elevates the risk of mitochondrial oxidative damage. Here, we present evidence that metabolically challenged mitochondria undergo active fusion to suppress oxidative stress. In response to glucose starvation, mitofusin 1 (MFN1) becomes associated with the protein deacetylase HDAC6. This interaction leads to MFN1 deacetylation and activation, promoting mitochondrial fusion. Deficiency in HDAC6 or MFN1 prevents mitochondrial fusion induced by glucose deprivation. Unexpectedly, failure to undergo fusion does not acutely affect mitochondrial adaptive energy production; instead, it causes excessive production of mitochondrial reactive oxygen species and oxidative damage, a defect suppressed by an acetylation-resistant MFN1 mutant. In mice subjected to fasting, skeletal muscle mitochondria undergo dramatic fusion. Remarkably, fasting-induced mitochondrial fusion is abrogated in HDAC6-knockout mice, resulting in extensive mitochondrial degeneration. These findings show that adaptive mitochondrial fusion protects metabolically challenged mitochondria.</P>

        <▼1><P>A missense mutation of <I>KARRIKIN-INSENSITIVE2</I>, <I>KAI2</I><SUP><I>ply2</I></SUP>, compromises its ligand-binding activity, which subsequently impairs KAI2-signaling and multiple aspects of light-dependent responses.</P></▼1><▼2><P><B>Abstract</B></P><P>A smoke-derived compound, karrikin (KAR), and an endogenous but as yet unidentified KARRIKIN INSENSITIVE2 (KAI2) ligand (KL) have been identified as chemical cues in higher plants that impact on multiple aspects of growth and development. Genetic screening of light-signaling mutants in <I>Arabidopsis thaliana</I> has identified a mutant designated as <I>ply2</I> (<I>pleiotropic long hypocotyl2</I>) that has pleiotropic light-response defects. In this study, we used positional cloning to identify the molecular lesion of <I>ply2</I> as a missense mutation of <I>KAI2</I>/<I>HYPOSENSITIVE TO LIGHT</I>, which causes a single amino acid substitution, Ala219Val. Physiological analysis and genetic epistasis analysis with the KL-signaling components <I>MORE AXILLARY GROWTH2</I> (<I>MAX2</I>) and <I>SUPPRESSOR OF MAX2 1</I> suggested that the pleiotropic phenotypes of the <I>ply2</I> mutant can be ascribed to a defect in KL-signaling. Molecular and biochemical analyses revealed that the mutant KAI2<SUP>ply2</SUP> protein is impaired in its ligand-binding activity. In support of this conclusion, X-ray crystallography studies suggested that the <I>KAI2</I><SUP><I>ply2</I></SUP> mutation not only results in a narrowed entrance gate for the ligand but also alters the structural flexibility of the helical lid domains. We discuss the structural implications of the Ala219 residue with regard to ligand-specific binding and signaling of KAI2, together with potential functions of KL-signaling in the context of the light-regulatory network in <I>Arabidopsis thaliana</I>.</P></▼2>
