B-cell lymphoma 3 (Bc13) is a proto-oncogene upregulated in a wide range of cancers, including breast cancer. Although Bc13 is known to promote cell proliferation and inhibit apoptosis, the molecular mech¬anisms underlying the proto-oncogenic function of Bc13 have not been completely elucidated. To gain insight into the oncogenic role of Bc13, we applied a proteomic approach, which led to the identification of C-terminal binding protein 1 (CtBP1) as a binding partner of Bc13. A PXDLS/R motif embedded in Bc13 was found to mediate the interaction between Bc13 and CtBP1, which caused the stabilization of CtBP1 by blocking proteasome-dependent degradation. Apoptotic stimuli-induced degradation of CtBP1 was sig¬nificantly abolished by the upregulation of Bc13, leading to the sustained repression of pro-apoptotic gene expression and subsequent inhibition of apoptosis. Intriguingly, a strong positive correlation between the protein levels of Bc13 and CtBP1 was detected in breast cancer patient samples. Our study reveals a novel combinatorial role for Bc13 and CtBP1, providing an explanation for the acquisition of resistance to apop¬tosis in cancer cells, which is a major requirement for cancer development.

The objective of this study is to identify the Korean copula ‘ida’ as an adjective and discuss grammatical issues regarding ‘ida’ by taking into account not only its multi-dimensional nature but also the whole Korean grammatical system. Until now, there have been many studies focusing on categorization issues, phonological dropping, conjugational characteristics of ‘ida’ and classification of ‘ida’ constructions. While the previous studies have examined individual topics on this copula, few studies have presented a balanced description of ‘ida’ within the whole system of the Korean grammar. Not only do we need to establish the category of ‘ida’ but also it is necessary to discuss the Korean POS and sentence complement systems in relation to the category of ‘ida’. This study is organized as follows. Chapter 2 describes the characteristics of ‘ida’ from the perspective of copula typology and provides an analysis of the most frequent patterns of ‘ida’, based on the lists of POS taggers and types in [n±1] of ‘ida’ in morphologically tagged Korean corpus. Furthermore, we show the conjugational similarities between ‘ida’ and ‘anida’. Chapter 3 contends that ‘ida’ finds a more appropriate place in the Korean POS and sentence complement systems if identified as an adjective. That is to say, the Korean copula ‘ida’ being described as an adjective provides answers to issues related to the Korean morpho-syntactical system.

<P><B>ABSTRACT</B></P><P>Correct temporal and spatial control of actin dynamics is essential for the cytotoxic T cell effector function against tumor cells. However, little is known whether actin engineering in tumor-targeted T cells can enhance their antitumor responses, thereby potentiating the adoptive T cell therapy. Here, we report that TAGLN2, a 22-KDa actin-stabilizing protein which is physically associated with lymphocyte function-associated antigen-1 (LFA-1), potentiates the <I>OTI TCR</I> CD8+ T cells to kill the intercellular adhesion molecule-1 (ICAM-1)-positive/OVA-presenting E0771 cells, but not ICAM-1-negative OVA-B16F10 cells, suggesting an ‘inside-out’ activation of LFA-1, which causes more efficient immunological synapse formation between T cells and tumor cells. Notably, recombinant TAGLN2 fused with the protein transduction domain (TG2P) overcame the disadvantages of viral gene delivery, leading to a significant reduction in tumor growth in mice. TG2P also potentiated the CD19-targeted, chimeric antigen receptor (CAR)-modified T cells to kill Raji B-lymphoma cells. Our findings indicate that activating the TAGLN2–actin–LFA-1 axis is an effective strategy to potentiate the adoptive T-cell immunotherapy.</P>

Binarized Neural Networks (BNNs) use only binary synapses of +1 and -1, not allowing any intermediate weights between -1 and +1. Though the recognition rate of BNNs is lower than the conventional Deep Neural Networks (DNNs), BNNs have attracted many interests nowadays, because BNNs do not need the complicated multiplication such as DNNs. Binary memristor crossbars can be very suitable to realize BNN hardware. This is because, in memristor BNNs, simple binary operation can be performed in bitwise manner for all the columns in memristor crossbars, simultaneously. In this paper, single-column and double-column memristor BNNs are presented, respectively. In addition, ReLU and sigmoid activation function circuits are also proposed with CMOS circuits. The designed Memristor-CMOS hybrid circuits of BNNs have been tested for MNIST vectors. The memristor BNNs could recognize almost 90% MNIST digits when the memristance variation is as large as 25%. For variation tolerance, the memristor BNNs are compared with the multi-valued memristor neural networks such as 4-bit, 6-bit, etc, in this paper. As a result, it has been confirmed the memristor BNNs become more variation-tolerant than the multi-valued memristor NNs when the variation becomes larger than 22%. Comparing the single-column and doublecolumn BNNs in this paper indicates that the singlecolumn BNN can save power consumption and array area almost by half than the double-column. This is because the single-column has just half memristors than the double-column. And, we measured the single-column and double-column BNNs using the fabricated memristor array. In this measurement, both the double-column and single-column BNNs were observed to work well.

Background/Aims: Little attention is paid to chronic lymphocytic leukemia/small lymphocytic lymphoma (CLL/SLL) in Korea due to the rarity of the disease. With its rising incidence, we aimed to evaluate recent changes in treatment patterns and survival outcomes of patients with CLL/SLL. Methods: A total of 141 patients diagnosed with CLL/SLL between January 2010 and March 2020 who received systemic therapy were analyzed in this multicenter retrospective study. Results: The median patient age was 66 years at diagnosis, and 68.1% were male. The median interval from diagnosis to initial treatment was 0.9 months (range: 0-77.6 months), and the most common treatment indication was progressive marrow failure (50.4%). Regarding first-line therapy, 46.8% received fludarabine, cyclophosphamide, plus rituximab (FCR), followed by chlorambucil (19.9%), and obinutuzumab plus chlorambucil (GC) (12.1%). The median progression-free survival (PFS) was 49.3 months (95% confidence interval [CI], 32.7-61.4), and median overall survival was not reached (95% CI, 98.4 mo-not reached). Multivariable analysis revealed younger age (≤ 65 yr) (hazard ratio [HR], 0.46; p < 0.001) and first-line therapy with FCR (HR, 0.64; p = 0.019) were independently associated with improved PFS. TP53 aberrations were observed in 7.0% (4/57) of evaluable patients. Following reimbursement, GC became the most common therapy among patients over 65 years and second in the overall population after 2017. Conclusions: Age and reimbursement mainly influenced treatment strategies. Greater effort to apply risk stratifications into practice and clinical trials for novel agents could help improve treatment outcomes in Korean patients.
