      • A new mid-late maturing japonica rice variety Hyeonpum with a good grain quality and eating quality

        ‘Hyeonpum’ is a new japonica rice variety developed by a cross breeding between Iksan469 having a good canopy architecture and Sindongjin and Musashino 7 having a good eating-quality with a view to develop a new variety having high quality of grain and palatability by the rice breeding team of Rice Breeding and Cultivation Research Division, Department of Rice and Winter Cereal Crop, NICS, RDA in 2012. The heading date of this variety is August 18 and later than that of check variety, Nampyeongbyeo, by four days. ‘Hyeonpum’ has 74cm of culm length and 101 spikelets per panicle. This variety showed resistance to bacterial leaf blight and rice stripe virus, but susceptible to leaf blast and planthoppers. The milled rice of this variety exhibits translucent and very clear non-glutinous endosperm. ‘Hyeonpum’ has much better palatability of cooked rice than that of Nampyeongbyeo. The whole grain rate of milled rice and milled rice recovery of ‘Hyeonpum’ are slightly higher than those of Nampyeongbyeo as 91.8% and 75.2% respectively. The yield of ‘Hyeonpum’ is 5.6 MT/ha in milled rice. ‘Hyeonpum’ could be adaptable to the southern part plain area of Pyeongtaek and southwestern costal areas in Korea.

      • Transgenic pig breeding and effect of hDAF & US11 genes in immune response

        Until now, problems related to shortage of organ for transplantation have been continuing. Pigs are the most suitable animal for xenotransplantation. Although primates are most similar to humans, they are not suitable because they have low productivity. Pigs are more productive than primates, and their organ size and physiological characteristics are similar to humans, with the exception of primates. In this study, we breeding the transgenic minipigs using natural mating to produce transgenic pigs. And, transgenic pigs has transmission rate that follow mendel’s rule. There are 20% hDAF gene, 20% US11 gene and 50% both hDAF and US11 gene in transgenic offsprings. Furthermore, transgenic pigs followed normal litter size, and piglets also has normal sex ratio. To suppress the immune function, experiments were performed using porcine ear fibroblast that transfected with hDAF and US11gene. In Cytotoxicity experiment against human complement, hDAF gene and double transgenic cell with both hDAF and US11 gene showed effect to reduce cytotoxicity rate in all of human complement condition. US11 gene and double transgenic cell were significantly reduce the cytotoxicity ratio in human NK cell. Besides, hDAF gene transgenic cell also reduce immune response in 10:1 concentration of human NK cell. In conclusion, natural mating was efficient method for breeding transgenic pigs. And, hDAF and US11 genes has effect for reduce cytotoxicity against human NK cell and human complement conditions.

        Quantitative analysis of mitochondrial DNA in porcine-mouse cloned embryos

        The aim of the research is to identify that porcine oocytes can function as recipients for interspecies cloning and have the ability to develop to blastocysts. Furthermore each mitochondrial DNA (mtDNA) in interspecises cloned embryos was analyzed. For the study, mouse-porcine and porcine-porcine cloned embryos were produced with mouse fetal fibroblasts (MFF) and porcine fetal fibroblasts (PFF), respectively, introduced as donor cells into enucleated porcine oocytes. The developmental rate and cell numbers of blastocysts between intraspecies porcine-porcine and interspecies mouse-porcine cloned embryos were compared and real-time polymerase chain reaction (PCR) was performed for the estimate of mouse and porcine mtDNA copy number in mouse-porcine cloned embryos at different stages.There was no significant difference in the developmental rate or total blastocyst number between mouse-porcine cloned embryos and porcine-porcine cloned embryos (11.1 ± 0.9%, 25 ± 3.5 vs. 10.1 ± 1.2%, 24 ± 6.3). In mouse-porcine reconstructed embryos, the copy numbers of mouse somatic cell-derived mtDNA decreased between the 1-cell and blastocyst stages, whereas the copy number of porcine oocyte-derived mtDNA significantly increased during this period, as assessed by real-time PCR analysis. In our real-time PCR analysis, we improved the standard curve construction-based method to analyze the level of mtDNA between mouse donor cells and porcine oocytes using the copy number of mouse beta-actin DNA as a standard. Our findings suggest that mouse-porcine cloned embryos have the ability to develop to blastocysts in vitro and exhibit mitochondrial heteroplasmy from the 1-cell to blastocyst stages and the mouse-derived mitochondria can be gradually replaced with those of the porcine oocyte in the early developmental stages of mouse-porcine cloned embryos.

      • Effect of US11 and hDAF Gene for Cytotoxicity Regulate and Transgenic Pigs Breeding

        This study aims to find effect of natural mating and cytotoxicity regulate from US11 and hDAF gene transgenic pig. Piglets that has US11, hDAF or US11&hDAF genes were born from natural mating of female US11 gene transgenic pig and male hDAF gene transgenic pig. Among 10 piglets were born, there are 2 piglets with US11 gene only, 2 piglets with hDAF gene only, 5 piglets with both US11 and hDAF gene, and 1piglets without transgenes. Sex ratio of piglets was 1:1, and transfection rate from natural breeding showed result that only US11 gene was 20%, only hDAF gene was 20%, and both US11 gene and hDAF gene was 50%. In cytotoxicity effect, US11 gene was showed a tendency of reduce human NK cell cytotoxicity in 10:1 ratio. In complement assay, hDAF gene was reduce human complement cytotoxicity in both 1:1 and 10:1 ratio of human complement. hDAF gene was also effected in reduce human NK cell cytotoxicity that showed similar result with US11 gene. Double transgenic piglets fibroblast that has both US11 and hDAF resulted that decrease both human NK cell and complement cytotoxicity compared with wild type cells. In conclusion, Natural mating was effected for US11 and hDAF gene transmitted into offsping piglets, and US11 and hDAF gene were expected to regulate xenograft rejections in transgenic tissues.

      • Cytotoxicity Assay of US11 & hDAF Genes, and Natural Breeding Effect in Transgenic Pigs

        Unlike allograft, xenotransplantation has problems such as hyperacute rejection (HAR), acute rejection and chronic rejection. HAR was rapid humoral immune responses against the pig carbohydrate Galα1,3-Gal epitope by human complement regulatory reaction, but HAR was almost overcome through knockout of α1,3-galactosyltransferase gene. The acute rejection is appeared within a few days to week caused by ischemia, diffuse intravascular coagulation and lack of cellular infiltrate. In this study, we bred the transgenic pigs containing US11 and hDAF (CD55) gene, evaluate transgene transmission ratio and efficiency of Reproduction rate in natural breeding. In addition, performed Cytotoxicity experiment about human NK cells and complement against each piglet ear fibroblast that have gene expression of US11, hDAF or both hDAF and US11. In breeding evaluate, sow has normal litter size, and piglets sex rate was followed 1:1. Transmission ratio of transgenes was indicated that wild type was 10%, US11 gene only was 20%, hDAF gene only was 20%, and both US11 gene and hDAF gene was 50% in 10 piglets. In cytotoxicity assay, US11 gene showed that effect to reduce cytotoxicity rate from human NK cell, and hDAF gene was significantly reduce the cytotoxicity ratio from complement. Besides, piglet cells that has both US11 gene and hDAF gene has effect to reduce cytotoxicity of NK cell and complement. In conclusion, our study showed that US11 and hDAF genes has effective to reduced Xenotransplantation rejection, and natural breeding was efficient to product transgenic pig.

        산전 우울 임부를 위한 긍정심리기반 프로그램의 효과

        Purpose: This study examined the effects of a positive psychology-based program for depressed pregnant women. Methods: Of the 221 pregnant women who were screened, 24 were recruited from 4 local women’s clinic in Daegu city, South Korea. Subjects were at 6-29 weeks of gestation and scored 9-13 points on the Korean version of the Edinburgh Postnatal Depression Scale. Subjects were assigned to experimental (n= 12) and control (n=12) groups. The program consisted of ten 120-minute sessions conducted twice a week. Data were collected during the period between December 21, 2017 and April 12, 2018 and analyzed using IBM SPSS/WIN (ver. 21.0). Homogeneity of the experimental and control groups was evaluated by chi-square test and t-test. Hypotheses were tested by repeated measure analysis of variance. Results: The results revealed that in terms of stress coping styles, the active coping style (group*time= 15.23, p<0.001) increased, while the passive coping style decreased (group*time=15.27, p<0.001). Perceived spouse support (group*time=19.45, p<0.001), self-esteem (group*time=17.39, p<0.001), perinatal depression (group*time=78.77, p<0.001), and flourish (group*time=57.56, p<0.001) have significantly improved in the experimental group compared with the control group. Conclusion: These findings indicate that positive psychology-based program can be useful for pregnant women already suffering from depression to prevent perinatal depression.

        산전우울 임부를 위한 인지행동치료 프로그램의 효과

        본 연구는 임부의 산전우울에 대한 인지행동치료 중재의 효과를 검증하기 위해 수행된 체계적 문헌고찰 및 메타분석 연구이다. 데이터베이스는 CINAHL, PubMed, EMBASE 및 Koreamed, 국회도서관, 한국학술정보(Korean studies Information Service System, KISS), 한국의학논문데이터베이스 등 국내외 domain을 활용하였다. 검색은 ((Perinatal OR Antenatal OR Antepartum OR Pregnant) AND Depression) AND Women AND (CBT OR (Cognitive behavioral AND (Therapy OR Treatment)))와 ‘산전우울’, ‘임부’, ‘인지행동치료’ 등의 검색어로 2016년 5월까지 출판된 문헌을 검토하였다. 분석은 R을 이용한 임의효과모형을 적용하여 산전우울과 불안의 효과크기를 각각 산출하였으며, 효과크기의 이질성을 검증하기위해 메타 ANOVA를 활용하여 조절효과분석을 실시하였다. 그리고 funnel plot, Egger’s regression test, fail-safe N, trim-and-fill 분석을 활용한 출간오류분석과 민감성 분석을 실시하여 전체 연구 결과의 타당성을 검증하였다. 연구결과 총 180편의 문헌이 검색되었으며, 선택배제기준에 따라 최종적으로는 clinical trials 16편을 분석하였다. 분석에 포함된 개별문헌은 Scottish Intercollegiate Guideline Network (SIGN)의 checklist를 통해 비뚤림 위험을 평가하였으며, 대체로 비뚤림 위험은 낮았다. 연구결과 본 연구에서 중재의 산전우울에 대한 효과크기는 Hedges’ g=-0.55(95% CI: -0.76∼-0.33)로 통계적으로 유의하게 낮았으며, 불안에 대한 효과크기는 Hedges’ g=-0.20(95% CI: -0.48∼-0.08)이었으나, 통계적으로 유의한 차이는 없었다. 문헌의 이질성, 출판오류의 위험성 등은 낮았다. 본 메타분석결과에 의하면 인지행동치료는 임신부의 산전우울 증세 완화에 보통 정도의 효과가 있음이 밝혀졌다. This study was carried out to evaluate the efficacy of CBT for perinatal depression through systematic literature review and meta-analysis. The following databases were used to search the literature: CINAHL, PubMed, EMBASE, Koreamed, Library of Korean Congress, KISS, and Korean Academic Publication Database. Keywords included "perinatal depression," "pregnant women," and "cognitive behavioral therapy," and the evaluated articles were published up to May 2016. Using the R program, the effect size of perinatal depression and anxiety were calculated by random-effects model. The heterogeneity of the effect size was analyzed by data moderator analysis using the meta-ANOVA. Furthermore, the funnel plot, Egger"s regression test, fail-safe N, trim-and-fill test, and publication bias analysis were conducted and used to verify the results. Out of the 180 selected articles, 16 clinical trial studies were meta-analyzed. Each articles were evaluated for the risk of bias by the checklist of SIGN; the overall risk of bias was low. The effect size of CBT for perinatal depression was Hedges" g=-0.55 (95% CI: -0.76∼-0.33), which was a moderate level, while for anxiety reduction, Hedges" g=-0.20 (95% CI: -0.48∼-0.08) and it was not statistically significant. Heterogeneity or risk of publication bias were low. This meta-analytic study found that CBT is moderately effective in reducing perinatal depression in pregnant women.
