<P>Discoidin domain receptor 1 (DDR1) is activated by fibrillar (triple-helical) collagens and collagen IV, which are major components of tumor stroma; thus, DDR1 might be a critical mediator of communication between cancer cells and stroma. The aim of this study was to investigate the effect of DDR1 inhibition on stroma-induced peritoneal metastasis in gastric carcinoma. We analyzed by immunohistochemistry the correlation between DDR1 expression and the pattern of recurrence in gastric carcinoma tissues from a previously characterized and established gastric carcinoma patient cohort. We also cocultured human gastric carcinoma cell lines with gastric cancer–associated fibroblasts (CAF) and investigated DDR1 expression and activation. We evaluated CAF-induced tumorigenic properties of gastric carcinoma cell lines and the effect of a DDR1-specific inhibitor in organotypic cultures and in a peritoneal seeding xenograft animal model. The expression of DDR1 in gastric cancer tissues was positively associated with early recurrence (<I>P</I> = 0.043) and a high incidence of peritoneal recurrence (<I>P</I> = 0.036). We confirmed that coculturing with CAFs elevated DDR1 protein expression in gastric carcinoma cell lines and enhanced gastric carcinoma cell line spheroid formation in organotypic cultures in a tumor cell DDR1-dependent manner. Coimplantation of CAFs with gastric carcinoma cells enhanced peritoneal tumor formation <I>in vivo</I>, an effect that was sensitive to pharmacologic inhibition of DDR1.</P><P><B>Implications:</B> This study highlights that CAF-induced elevation of DDR1 expression in gastric carcinoma cells enhances peritoneal tumorigenesis, and that inhibition of DDR1 is an attractive strategy for the treatment of gastric carcinoma peritoneal metastasis. <I>Mol Cancer Res; 16(10); 1590–600. ©2018 AACR</I>.</P>

Today, elder population has been increasing as it becomes an aging society. South Korea announced by UN as an aging society in 2000 as well, the percentage is still increasing rapidly. However, the residential facility for eledr people is deteriorative compare to other advanced country. In USA, Europe and our neighborhood country Japan, an advanced, active and convenience design for potential users is providing. As well as, it is developed active. Nowadays, the design market that is faster and applied new technology for younger users is hard to use and inconvenience for elder people who is analogue age. This problem has been shown up in many differnet kinds of field. It also has been shown in home network that is made to convenience residential facility. The residential automatic systems, home automation, network, smart home, is the most necessary and reliable technique however, it has difficulty to use that because of the access way and advanced aspect. So, in this thesis, a model of smart home that has easy interface for elder people is going to provided, furthermore it would suggest an easy to use and access design for the asing society that is coming to our near future
Determination of acibenzolar-S-methyl and acibenzolar acid in agricultural commodities by LC-MS/MS

Acibenzolar-s-methyl is a well-known plant activator and fungicide, which is synthetic analog of salicylic acid. Usually, it is not directly toxic to fungi but works by inducing systemic acquired resistance, the natural defence system of plants. The residue definition set in MRL regulation for all agricultural commodities is the sum of acibenzolar-Smethyl(ester form) and acibenzolar acid, expressed as acibenzolar-S-methyl. The current official analytical method(MFDS Food Code) is to hydrolyze acibenzolar-S-methyl in a sample with acibenzolar acid and analyze it with LC-UVD. In this study, a method based on QuEChERS and LC-MS/MS was developed for the simultaneous determination of acibenzolar-S-methyl and acibenzolar acid in agricultural commodities. Two compounds were extracted from matrix with acetonitrile containing 1% formic acid and treated with QuEChERS original salts. Recovery experiments were conducted on 3 agricultural commodities(rice, mandarin, and pepper) and main performance parameters investigated were linearity, recovery, relative standard deviation(RSD), and limit of quantitation(LOQ). The recoveries were within 71.1∼116.9% with the RSD of 0.7∼14.8% and the LOQ of methods were below 0.01mg/kg, and the coefficient of determination (R2) of matrix-matched standards were > 0.995. The proposed method was reproducible and sensitive enough to determine the residues of acibenzolar-S-methyl and acibenzolar acid in agricultural commodities.
Müllerian inhibiting substance gene expression and fertility in Pagrus major

Müllerian inhibiting substance (MIS) is a protein that encoded by MIS gene. It has also been called Müllerian inhibiting factor (MIF) and anti-Müllerian hormone (AMH). Mis expression occurs in ovarian granulose cells of females postpartum, and serves as a molecular biomarker for relative size of the ovarian reserve. In humans, the number of cells in the follicular reserve can be used to evaluate the reproductive function and fertility of female. Pagrus major is typical cultured fish in Korea but there is no clear evidence for their gene identification. However, in many teleost, MIS genes were demonstrated already. Present study aimed to identify the Mis gene in Pagrus major and seasonal difference of its expression. Using conserved sequence of the other known teleost Mis genes, we make conserved primers. Pagrus major’s ovary samples were obtained from the sea rim farm (Geoje, Korea) and kept in RNAlater®Solution or fixed in 4% paraformaldehyde containing 0.16% picric acid. RNA was isolated from kept sample and cDNA was synthesized. The PCR products were performed ligation with TOPO vector and transformation in TOP10 cell and sequenced the Mis mRNA fragment. MIS was localized in the follicle cells. Its mRNA levels were higher in summer than spring or fall. Based on them, it is suggested that MIS can be used to estimate the fertility of this fish.

In Brassica campestris, self-incompatibility (SI) can be overcome by CO2 gas treatment. Previously, we reported inhibition of the SI in the Brassica campestris line, 734, by CO2 gas. In this study, we used a cDNA microarray comprising 1,184 unique cDNA ESTs from a pistil-specific cDNA library of the Brassica line to examine the expression of the pistil genes in response to CO2. Microarray analysis revealed that expression of 2% of the pistil genes was altered by exposure to CO2 gas. Surprisingly, the SLG gene of the susceptible line was one of the down-regulated genes. Transmission electron microscopic analysis showed that the CO2 caused morphological changes in the papillary cell. Our results demonstrate that the overcoming of SI by CO2 gas involves reduced expression of the SLG gene in Brassica.
