        Stimulation of angiogenesis and survival of endothelial cells by human monoclonal Tie2 receptor antibody

        <P><B>Abstract</B></P> <P>Angiopoietin-1 (Ang1) and its endothelium-specific receptor, tyrosine kinase with Ig and epidermal growth factor homology domain 2 (Tie2), play critical roles in vascular development. Although the Ang1/Tie2 system has been considered a promising target for therapeutic neovascularization, several imitations of large-scale production have hampered the development of recombinant Ang1 for therapeutics. In this study, we produced a fully human agonistic antibody against Tie2, designated 1–4h, and tested the applicability of 1–4h as an alternative to native Ang1 in therapeutic angiogenesis. 1–4h significantly enhanced the phosphorylation of Tie2 in a dose- and time-dependent manner in human Tie2-expressing HEK293 cells and human umbilical vein endothelial cells. Moreover, 1–4h induced the activation of Tie2-mediated intracellular signaling such as AKT, eNOS, MAPK, and Focal Adhesion Kinase p125<SUP>FAK</SUP>. In addition, 1–4h increased the chemotactic motility and capillary-like tube formation of endothelial cells <I>in vitro</I> and enhanced the survival of serum-deprived endothelial cells. Taken together, our data clearly suggest that a human Tie2 agonistic antibody is a potentially useful therapeutic approach for the treatment of several ischemic diseases including delayed-wound healing and ischemic heart and limb diseases.</P>

        압력카트리지는 화약으로 고온-고압의 가스를 형성하는 일종의 에너지 발생 장치이다. 압력카트리지는 너트, 볼트, 밸브, 사출시스템, Pressure-Actuated Pump, Switch 및 Pyrotechnic Cutter와 같은 피스톤 장치 등 항공 우주 산업 분야에서 다양하게 사용된다. 압력카트리지는 에너지원인 주장약의 재료에 따른 성능의 차이가 크다. 본 논문은 주장약 재료에 따른 압력카트리지 성능을 비교 하였다. The pressure cartridge is kind of energy generating device to form the high-pressure and high-temperature gas using pyrotechnic charge. The pressure cartridge is used in the military and aerospace industry such as nut, bolt, valve, ejection system, pressure-actuated pump, switch and pyrotechnic cutter. The pressure cartridge shows performance variation depending on the main charge, which is the energy source. This paper compares the main charge characteristics according to the main charge.

        중대재해법 관련 사회적 담론 구조: 트위터와 신문 기사를 중심으로

        트위터 데이터와 보수언론 및 진보언론의 기사를 대상으로, 토픽모델링, 워드 임베딩 및 소셜 네트워크 분석 방법론을 통해 중대재해법과 관련된 논의를 다양한 층위에서 분석하고자 하였다. 분석결과, 언론에서의 논의와 구별되는 중대재해법에 관해 트위터 유저들이 전개하고 있는 시민 관점의 독자적 논의의 내용이 확인되었다. 한편 보수언론과 상대적 진보언론으로 구분하여 분석한 결과로부터 보수언론의 경우 상대적으로 경영자, 기업의 관점이 전면에 드러나는 모습을 발견할 수 있었던 반면, 진보언론의 경우 노동계의 관점이 강조되는 경향을 발견할 수 있었다. 특히 보수언론의 경우 보수정당의 인물-기관, 진보언론의 경우 상대적 진보정당의 인물-기관이 전면에 드러나는데, 이는 결국 정치적 이념적 관점이 중대재해법과 관련된 사회적 논의에서도 상당 부분 작용하였을 가능성을 시사한다. To capture the structure of social discussion on “Serious Accidents Punishment Law”, this study analyzed Twitter’s tweets and online news articles from conservative and progressive media at various levels through topic modeling, word embedding, and social network analysis methodologies. The result of this paper shows that the uniqueness of citizens’ perspectives represented by Twitter users’ tweets, which is distinct from that of the news media, were confirmed. In the case of conservative media, it was prominent that the perspectives of corporate managers and companies were relatively more exposed, while in the case of progressive media, it was possible to find a tendency that the perspectives of the group of workers were more emphasized. Through the analysis of the social network, similarity between the conservative party’s perspective and that of conservative media was confirmed. Likewise, relationship between liberal media and liberal party’s perspectives are revealed, suggesting that political and ideological perspectives may have been reflected in social discussions related to the law.

        Recently, train control system is adopting computer system replacing mechanical system and its software is taking more responsibility than ever. Train control system software is a safety-critical embedded software with realtime and high reliability requirements. In this paper, we propose a safety assessment method for the train control system software. We review characteristics of train control system software and analyze related international software safety standards to derive requirements for safety assessment. Testing tools used for embedded software are surveyed to find a feasible safety assessment architecture. The proposed safety assessment method is to use safety activity results generated during development processes and feed them to the runtime embedded software testing tool.

        KLHL3 deficiency in mice ameliorates obesity, insulin resistance, and nonalcoholic fatty liver disease by regulating energy expenditure

        Obesity is a growing global epidemic that can cause serious adverse health consequences, including insulin resistance (IR) and nonalcoholic fatty liver disease (NAFLD). Obesity development can be attributed to energy imbalance and metabolic inflexibility. Here, we demonstrated that lack of Kelch-like protein 3 (KLHL3) mitigated the development of obesity, IR, and NAFLD by increasing energy expenditure. KLHL3 mutations in humans cause Gordon’s hypertension syndrome; however, the role of KLHL3 in obesity was previously unknown. We examined differences in obesity-related parameters between control and Klhl3−/− mice. A significant decrease in body weight concomitant with fat mass loss and improved IR and NAFLD were observed in Klhl3−/− mice fed a high-fat (HF) diet and aged. KLHL3 deficiency inhibited obesity, IR, and NAFLD by increasing energy expenditure with augmentation of O2 consumption and CO2 production. Delivering dominant-negative (DN) Klhl3 using adeno-associated virus into mice, thereby dominantly expressing DN-KLHL3 in the liver, ameliorated diet-induced obesity, IR, and NAFLD. Finally, adenoviral overexpression of DN-KLHL3, but not wild-type KLHL3, in hepatocytes revealed an energetic phenotype with an increase in the oxygen consumption rate. The present findings demonstrate a novel function of KLHL3 mutation in extrarenal tissues, such as the liver, and may provide a therapeutic target against obesity and obesity-related diseases.
