Necroptosis is the major cause of death in alveolar epithelial cells (AECs) during acute lung injury (ALI). Here, we report a previously unrecognized mechanism for necroptosis. We found an accumulation of mitochondrial citrate (citratemt) in lipopolysaccharide (LPS)-treated AECs because of the downregulation of Idh3α and citrate carrier (CIC, also known as Slc25a1). shRNA- or inhibitor–mediated inhibition of Idh3α and Slc25a1 induced citratemt accumulation and necroptosis in vitro. Mice with AEC-specific Idh3α and Slc25a1 deficiency exhibited exacerbated lung injury and AEC necroptosis. Interestingly, the overexpression of Idh3α and Slc25a1 decreased citratemt levels and rescued AECs from necroptosis. Mechanistically, citratemt accumulation induced mitochondrial fission and excessive mitophagy in AECs. Furthermore, citratemt directly interacted with FUN14 domain-containing protein 1 (FUNDC1) and promoted the interaction of FUNDC1 with dynamin-related protein 1 (DRP1), leading to excessive mitophagy-mediated necroptosis and thereby initiating and promoting ALI. Importantly, necroptosis induced by citratemt accumulation was inhibited in FUNDC1-knockout AECs. We show that citratemt accumulation is a novel target for protection against ALI involving necroptosis.
Phytochemical Constituents from Metasequoia glyptostroboids Leaves

Phytochemical investigation of Metasequoia glyptostroboids leaves resulted in the isolation of ten compounds. The structures were determined to be isoquercitrin (1), quercitrin (2), myricitrin (3), amentoflavone (4), sciadopitysin (5), isoginkgetin (6), 2,3-dihydrosciadopitysin (7), 2,3-dihydroisoginkgetin (8), 3β-acetoxy- 8(17),13E-labdadien-15-oic acid (9) and β-sitosterol (10) by spectroscopic analyses. Isoquercitrin (1) was isolated from this plant for the first time.

<P>Pancreatic insulin-producing beta-cell dysfunction and death plays central roles in the onset and progression of both type 1 and type 2 diabetes. Current antidiabetic drugs cannot halt the ongoing progression of beta-cell dysfunction and death. In diabetes, a major cause for the decline in beta-cell function and survival is endoplasmic reticulum (ER) stress. Here, we identified quinazoline derivatives as a novel class of beta-cell protective agents against ER stress-induced dysfunction and death. A series of quinazoline derivatives were synthesized from dichloroquiazoline utilizing a sequence of nucleophilic reactions. Through SAR optimization, 2,4-diaminoquinazoline compound 9c markedly protects beta-cells against ER stress-induced dysfunction and death with 80% maximum rescue activity and an EC50 value of 0.56 mu M. Importantly, 9c restores the ER stress-impaired glucose-stimulated insulin secretion response and survival in primary human islet beta-cells. We showed that 9c protects beta-cells by alleviating ER stress through the suppression of the induction of key genes of the unfolded protein response and apoptosis.</P>
Correlation analysis of the wind of a cable-stayed bridge based on field monitoring

This paper investigates the correlation of wind characteristics monitored on a cable-stayed bridge. Total five anemoscopes are implemented into the bridge. Two out of 5 anemoscopes in inflow and two out of 5 anemoscopes in wake-flow along the longitudinal direction of the bridge are installed. Four anemoscopes are respectively distributed at two cross-sections. Another anemoscope is installed at the top of the tower. The correlation of mean wind speed and direction, power spectral density, the turbulent intensity and integral length of wind in flow at two cross-sections are investigated. In addition, considering the non-stationary characteristics of wind, the spatial correlation in time-frequency is analyzed using wavelet transform and different phenomenon from those obtained through FFT is observed. The time-frequency analysis further indicates that intermittence, coherence structures and self-similar structures are distinctly observed from fluctuant wind. The flow characteristics around the bridge deck at two positions are also investigated using the field measurement. The results indicate that the mean wind speed decrease when the flow passing through the deck, but the turbulence intensity become much larger and the turbulence integral lengths become much smaller compared with those of inflow. The relationship of RMS (root mean square) of wake-flow and the mean wind speed of inflow is approximately linear. The special structures of wake-flow in time-frequency domain are also analyzed using wavelet transform, which aids to reveal the forming process of wake-flow.

The Wnt signaling pathway is well known to play major roles in skeletal development and homeostasis. In certain aspects, fracture repair mimics the process of bone embryonic development. Thus, the importance of Wnt signaling in fracture healing has become more apparent in recent years. Here, we summarize recent research progress in the area, which may be conducive to the development of Wnt-based therapeutic strategies for bone repair.
Matching Reconstruction Algorithms Performance Comparison based on Compressed Sensing in GPR Imaging

Compressed sensing (CS) provides a new solution for the problems of requiring large amount of measurements data and long data acquisition time in radar application, and both issues also exist in ground penetrating radar (GPR). Aiming at this problem, we adopt impulse radar with CS framework, and transform the GPR imaging into sparse constraint optimization problem performed on time-domain sub-sampling in this paper. Specifically, it focuses on the impulse GPR imaging method based on CS under double underground targets condition containing noise and abundant clutter. Furthermore, the performance of matching reconstruction algorithms under the different signal to noise ratios (SNR), measurement dimensions and sparseness values is also presented. The experimental results show that CS algorithms based on matching reconstruction can obviously reduce measurement data, improve the image quality and make a better anti-noise performance. When SNR of measurement data is 1dB, the probability of accurate imaging can still reach 95%. So we may reasonably conclude that the regularized orthogonal matching pursuit algorithm has a better performance than the other matching algorithms.

Aim: This study was to evaluate the effect of whole brain radiation (WBRT) combined with stereotactic radiotherapy (SRS) versus stereotactic radiotherapy alone for patients with brain metastases using a meta-analysis. Materials and Methods: We searched PubMed, EMBASE, Cochrane Library from their inception up to October 2013. Randomized controlled trials involving whole brain radiation combined with stereotactic radiotherapy versus stereotactic radiotherapy alone for brain metastases were included. Statistical analyses were performed using RevMan5.2 software. Results: Four randomized controlled trials including 903 patients were included. The meta-analysis showed statistically significant lowering of the local recurrence rate (OR=0.29, 95%CI: 0.17~0.49), new brain metastasis rate (OR=0.45, 95%CI: 0.28~0.71) and symptomatic late neurologic radiation toxicity rate (OR=3.92, 95%CI: 1.37~11.20) in the combined group. No statistically significant difference existed in the 1-year survival rate (OR=0.78, 95%CI: 0.60~1.03). Conclusions: The results indicate that whole brain radiotherapy combined with stereotactic radiotherapy has advantages in local recurrence and new brain metastasis rates, but stereotactic radiotherapy alone is associated with better neurological function. However, as the samples included were not large, more high-quality, large-sample size studies are necessary for confirmation.
An Improved NOx Emission Factor Model for Estimating Exhaust Emissions on Two Bulk Carriers

A dynamic NOx emission factors (EFNOx) model was developed based on on-board exhaust measurements of two bulk carriers over the course of a navigation. Missing AIS data processing, ship speed modification, fuel consumption estimation and on-board exhaust measurement data were used to develop dynamic EFNOx model. The ship speed in the cube of ratio of ship speed and maximum service speed was modified by wind, wave and current to estimate main engine loads. The results show that the modified speed (MS) can accurately predict the main engine loads, which verified by actual fuel consumption, and the absolute value of relative error for each cruise was range from 1.00% and 24.39%. The dynamic EFNOx model shown a better prediction of NOx emission quantity than that current inventory, and the absolute value of relative error for each cruise was between 5.69%~25.26%. The model is helpful to improve the calculation accuracy of emission inventory for typical bulk carriers in China.
