Crocetin Induces Cytotoxicity in Colon Cancer Cells Via p53-independent Mechanisms

Objective: Crocin has been proposed as a promising candidate for cancer chemoprevention. The purpose of this investigation was to investigate the chemopreventive action and the possible mechanisms of crocin against human colon cancer cells in vitro. Methods: Cell proliferation was examined using MTT assay and the cell cycle distribution fractions were analyzed using fow cytometric analysis after propidium iodide staining. Apoptosis was detected using theTUNEL Apoptosis Detection Kit with laser scanning confocal microscope. DNA damage was assessed using the alkaline single-cell gel electrophoresis assay, while expression levels of p53, cdk2, cyclinA and P21 were examined by Western blot analysis. Results: Treatment of SW480 cells with crocetin (0.2, 0.4, 0.8 mmol/L) for 48 h signifcantly inhibited their proliferation in a concentration-dependent manner. Crocetin (0.8 mmol/L) signifcantly induced cell cycle arrest through p53-independent mechanisms accompanied by P21 induction. Crocetin (0.8 mmol/L) caused cytotoxicity in the SW480 cells by enhancing apoptosis and decreasing DNA repair capacity in a time-dependent manner. Conclusions: This report provides evidence that crocetin is a potential anticancer agent, which may be used as a chemotherapeutic drug.

Serum miR-21 Expression in Human Esophageal Squamous Cell Carcinomas

To investigate the relationship between serum miRNA-21 (miR-21) expression in esophageal squamous cell carcinomas (ESCCs) and its clinicopathologic features, a 1:1 matched case-control study including 21 patients with ESCC and 21 age- and gender-matched healthy controls was carried out. Serum specimens were taken from all subjects. Total RNA was extracted and the stem-loop real time polymerase chain reaction was used to measure serum miR-21 in both groups. Clinical parameters were assessed to determine associations with serum miR-21 concentrations. Serum miR-21 expression in ESCC samples was significantly higher than in paired cancer-free samples (P<0.05). Metastasis was associated with mir-21 expression in serum (P<0.05), ESCC patients with metastasis having 8.4-fold higher serum miR-21 concentrations than healthy controls. There were no statistically significant associations between miR-21 expression and clinicopathologic parameters, such as gender (P>0.05), age (P>0.05), tumor location (P>0.05), cell differentiation (P>0.05), TNM staging (P>0.05), whether chemo/radiotherapy had been administered (P>0.05), or whether surgery had been performed (P>0.05). These findings suggest that the detection of microRNA-21 in serum might serve as a new tumor biomarker in diagnosis and assessment of prognosis of ESCCs.

Background miRNAs play an important role in regulating normal animal development. Muscle tissue and fat metabolismare important for maintaining energy balance in animals. Yak has important agricultural and economic importance as itprovides milk, meat, and hair. It is used for transportation as well. However, the miRNA expression profiles of their muscleand adipose tissue are currently unknown. Objective To explore the regulatory roles of miRNAs in the skeletal muscle and adipose tissues of yak. Methods A total of 12 small RNA libraries were constructed from the skeletal muscle and adipose samples from yak aged 0.5,2.5, 4.5, and 7.5 years. High-throughput sequencing and bioinformatics analysis were used to determine the dynamic expressionprofile of miRNA, and a miRNA regulatory network related to muscle and adipose tissue development was established. Results miR-1-3p and miR-143-3p showed the highest expression during yak skeletal muscle and fat development, respectively. The MAPK and Ras signaling pathways were the pivotal pathways. miR-181-5p, miR-542-3p, and miR-424-5p mayhave key roles in skeletal muscle development, and CREBRF, GRB10, CDK1, RFX3, and EPC2 were the core target genes. While miR-127-5p, miR-379-3p, and miR-494-3p may play important regulatory roles in adipose deposition, and ETV1,XPO7, and C5AR2 were the core target genes. Conclusion This study provides valuable resources for further study of the molecular mechanisms underlying yak skeletalmuscle and adipose tissue development, and also a basis for studying the interactions between genes and miRNAs.
Dynamic Strain-Induced Ferrite Transformation during Hot Compression of Low Carbon Si-Mn Steels

<P>The dynamic strain-induced transformation (DSIT) of austenite to ferrite was investigated under different undercooling conditions using three low carbon Si-Mn steels. The undercooling of austenite (Δ<I>T</I>) was controlled by varying the cooling rate between austenitization and deformation temperatures. Uniform DSIT ferrite grains (∼2.3 μm) were produced at a relatively high deformation temperature above 840°C using a low carbon high Si steel (0.077C-0.97Mn-1.35Si, mass%) in connection with a larger Δ<I>T</I>. The critical conditions for DSIT were determined based on the flow stress-strain curves measured during hot compression tests. Influence of deformation temperature on DSIT of low carbon Si-added steel was also discussed.</P>

Strong laws are established for linear statistics that are weighted sums of a random sample. We show extensions of the Marcinkiewicz-Zygmund strong laws under certain moment conditions on both the weights and the distribution. The result obtained extends and sharpens the result of Sung ([12]).
Three-Dimensional Surface Rendering and Computer-aided Diagnosis based on CT Images

According to the actual application requirements of computer-aided diagnosis, it’s necessary to segment a group of continues two-dimensional tomographic images and construct the three-dimensional surface modeling of human organ, in this case doctors can fully understand the lesions situation of patient by 3D models. But it’s common that those surface patches are defined on different parameter spaces in the process of surface rendering, which causes smoothness problem of human organ surface. This paper presents a new method to define boundary conditions, which defined by the new method have the same parameter space if the three given surface patches can be converted into the same form through affine transformation. Experimental results show that the method of parametric triangular patch has the same interpolation accuracy and smoother surface with existing methods, especially all three-dimensional lung image reconstruction examples show that this method can provide effective quantitative and qualitative diagnosis in clinical practice.
Analysis and Modeling of AC–AC Switched Capacitor Converters

A new modeling method for AC–AC switched capacitor converters (SCCs) is introduced in this study. The proposed analyticalmethod aims to accurately describe the input–output characteristics of AC–AC SCCs and establish a mathematical model forstatic voltage conversion ratio and equivalent resistance, which are key performance metrics for SCCs. A quantitative analysis ofconverter regulation capability is addressed on the basis of the modeling method. In this analysis, the effects of the controlparameters and individual components on SCCs are illustrated extensively. Component stresses, such as the peak value andtransient variation of the voltage/current of the converter, are also presented. The effectiveness of the proposed method is verifiedby comparing it with the existing modeling method and applying it to an AC–AC SCC with a conversion ratio of three. Two 1kW prototypes are built in a laboratory, and their experimental results exhibit good agreement with the theoretical analysis.
