        Study of the Higher-order Shock Excitations in a Degenerate Quantum Plasma

        We propose a comprehensive theory for one dimensional ion-acoustic shock waves (IASWs) in anunmagnetized degenerate quantum plasma that is assumed to contain inertial ions and relativisticdegenerate electrons. The modified Burgers (mB) and the Gardner equations have been derived byemploying the reductive perturbation method and have been analyzed in order to identify the basicfeatures (amplitude, width, speed, etc.) of the IASWs. The basic features of these shocks obtainedfrom this analysis are observed to be significantly different from those obtained from the standardBurgers equation. The implications of our results in space and interstellar compact objects (viz. white dwarfs, non-rotating neutron stars, active galactic nuclei, cometary tails, etc.) are brieflymentioned.

        In vitro antioxidative and anti-inflammatory effects of the compound K-rich fraction BIOGF1K, prepared from Panax ginseng

        Background: BIOGF1K, a compound K-rich fraction prepared from the root of Panax ginseng, is widely used for cosmetic purposes in Korea. We investigated the functional mechanisms of the anti-inflammatory and antioxidative activities of BIOGF1K by discovering target enzymes through various molecular studies. Methods: We explored the inhibitory mechanisms of BIOGF1K using lipopolysaccharide-mediated inflammatory responses, reporter gene assays involving overexpression of toll-like receptor adaptor molecules, and immunoblotting analysis. We used the 2,2-diphenyl-1-picrylhydrazyl (DPPH) assay to measure the antioxidative activity. We cotransfected adaptor molecules, including the myeloid differentiation primary response gene 88 (MyD88) and Toll/interleukin-receptor domain containing adaptor molecule-inducing interferon-${\beta}$ (TRIF), to measure the activation of nuclear factor (NF)-${\kappa}B$ and interferon regulatory factor 3 (IRF3). Results: BIOGF1K suppressed lipopolysaccharide-triggered NO release in macrophages as well as DPPH-induced electron-donating activity. It also blocked lipopolysaccharide-induced mRNA levels of interferon-${\beta}$ and inducible nitric oxide synthase. Moreover, BIOGF1K diminished the translocation and activation of IRF3 and NF-${\kappa}B$ (p50 and p65). This extract inhibited the upregulation of NF-${\kappa}B$-linked luciferase activity provoked by phorbal-12-myristate-13 acetate as well as MyD88, TRIF, and inhibitor of ${\kappa}B$ ($I{\kappa}B{\alpha}$) kinase ($IKK{\beta}$), and IRF3-mediated luciferase activity induced by TRIF and TANK-binding kinase 1 (TBK1). Finally, BIOGF1K downregulated the NF-${\kappa}B$ pathway by blocking $IKK{\beta}$ and the IRF3 pathway by inhibiting TBK1, according to reporter gene assays, immunoblotting analysis, and an AKT/$IKK{\beta}$/TBK1 overexpression strategy. Conclusion: Overall, our data suggest that the suppression of $IKK{\beta}$ and TBK1, which mediate transcriptional regulation of NF-${\kappa}B$ and IRF3, respectively, may contribute to the broad-spectrum inhibitory activity of BIOGF1K.

        <P><B><I>Introduction:</I></B> 3-Phosphoinositide-dependent kinase 1 (PDK1) is a master regulator of the AGC protein kinase family and is a critical activator of multiple pro-survival and oncogenic protein kinases, for which it has garnered considerable interest as an oncology drug target.</P><P><B><I>Areas covered:</I></B> This manuscript reviews small molecule patent literature disclosures between October 2011 and September 2014 for both PDK1 activators and inhibitors and restates the selective patents published before September 2011. PDK1 modulators are organized according to pharmaceutical company and chemical structural class.</P><P><B><I>Expert opinion:</I></B> Many academic institutions and pharmaceutical companies continue to research into the development of small molecules that can function as PDK1 inhibitors or modulators. To date, > 50 patent publications on PDK1 disruptors and modulators have been published since the protein was first discovered in 1998. Most of these molecules act as ATP mimetics, forming similar hydrogen bonding patterns to PDK1 as ATP and functioning as hydrophobic pharmacophores. To achieve selectivity in PDK1 inhibition, the discovery of binding pockets structurally distinctive from the ATP site is a challenging but promising strategy.</P>

        AP-1-Targeted Inhibition of Macrophage Function and Lipopolysaccharide/D-Galactosamine-Induced Hepatitis by Phyllanthus acidus Methanolic Extract.

        <P>Traditionally, Phyllanthus acidus (Phyllanthaceae) has been used for the treatment of rheumatism, bronchitis, asthma, respiratory disorders, and hepatitis. Recently, we showed that a methanol extract of Phyllanthaceae (Pa-ME) has a potent anti-inflammatory activity in RAW264.7 cells and strongly ameliorates HCl/EtOH-induced gastric ulcers in mice by targeting the Src/Syk of NF-kappa B. In the present study, we explored the molecular mechanism of Pa-ME on the AP-1 activation pathway and evaluated its potential hepatoprotective effects. To do this, we employed lipopolysaccharide (LPS)-stimulated RAW264.7 cells and U937 cells and an LPS/D-galactosamine (D-GaIN)-induced acute hepatitis mouse model. We utilized a multitude of assays, including immunoblotting analysis, reporter gene assays, and mRNA expression analysis, to determine the effect of Pa-ME on the AP-1 pathway. Pa-ME strikingly suppressed the production of LPS-induced pro-inflammatory cytokines including interleukin (IL)-1 beta, IL-6, and tumor necrosis factor-alpha (TNF-alpha). Furthermore, Pa-ME also strongly inhibited activator protein-1 (AP-1) activation and mitogen-activated protein kinase (MAPK) phosphorylation in LPS-stimulated RAW264.7 macrophages cells and the U937 monocyte like human cell line. Moreover, pre-treatment with Pa-ME exhibited strong hepatoprotective and curative effects in an LPS/D-Gal-induced mouse hepatitis model as evidenced by a decrease in elevated serum AST and ALT levels and the amelioration of histological damage. Taken together, our data suggest that Pa-ME might play a crucial ethnopharmacological role as a hepatoprotective herbal remedy by suppressing MAPK signaling and the activity of the downstream transcription factor AP-1.</P>

        Concept Selection of NPP Construction Delay Risk Assessment Methodology Using Systems Engineering Approach

        Construction industry faces a lot of inherent uncertainties and issues and the construction phase of nuclear power project is not free from this risk. This paper investigates promising methodologies to be used on nuclear power plant (NPP) construction schedule delay risk assessment by using entry level systems engineering approach. This study contains how the initial concept for the risk assessment methodology has been developed. In this point of view, this work structured on three main phases: needs analysis (NA), concept exploration (CE), and concept definition (CD) through systems engineering (SE) approach. Traditionally, the SE process is applied to technical development programs but this study opens up a new avenue that SE can also be successfully applied to the development and optimization of the risk assessment model. This study provides a rational and systematic process for developing and selecting the best risk assessment model. This paper selects analytic hierarchy process (AHP) method to assess NPP construction schedule delay risk for international project. As conclusion, the proposed concept and selected method can discriminate successfully and clearly among schedule delay risk assessment methods.

        Development of early flowering, short life-spanned jute (Corchorus spp.) mutant via ethyl methane sulfonate mutagenesis

        Polyethene is a global environmental threat, whereas jute (Corchorus spp.) fber is biodegradable, eco-friendly and can be used as a substitute for polyethene. Jute is a short-day plant and cultivated in March to July in Indian subcontinent for bast fber production. This season-bound cultivation nature hampers continuous supply of bast fber in the industry. In addition, its long cultivation period creates difculties to accommodate other high-value crops. To address these issues, a short life span (early fowering) jute genotype is extremely felt in jute growing areas of the world. But, unfortunately such variety has not been developed yet through both conventional and biotechnological approach. Accordingly, we adopted chemical mutagenesis by establishing LD50 (Lethal Dose) of EMS (Ethyl Methane Sulfonate) at 150 mM with 4h’ incubation for Capsularis jute seed. A novel mutant was found named—Komola, which showed a shorter lifespan than its ancestor by fowering at 70 days instead of 120 days. In addition, the mutant's stem and petiole was coppery-red instead of green. Molecular analysis revealed four SNPs in PMIR1 (Plastid Movement Impaired 1 Related 1) and two clade deletions in ELF3 (Early Flowering 3) genes from stable M4 generation. Histochemical and biochemical analyses explained this genotype’s lower content of lignin. This mutant could be used as future breeding material for the development of year-round cultivable jute genotype along with ofer up accommodation of other high-value agricultural crops in cropping pattern with a good source of year-round supply of bast fber to the industry

        Selection of Architect Engineering Concept for Barge Mounted SMR Using Systems Engineering Approach

        The trade-off studies in the concept development stage to assess the relative goodness of alternative systems concepts for AE (architect engineering) design for the Barge Mounted SMR (BMSMR) is introduced. With respect to design margin, system performance, schedule and risk, the design selection is cond ucted using the following characteristics; barge mobility, system safety under the natural disaster (seismic), power output, interfacing with the other system, and the additional supporting functions as desalination. There are three findings that should be remedied; deficiencies in the assumed characteristics of the system being modeled, deficiencies in the test model, and excessively stringent system requirements. This study is performed using systems engineering approach with trade off matrix method. In order to execute this work, concept development stage is divided into three (3) phases as NA (needs analysis), CE (concept exploration), and CD (concept definition).

      • Stereoscopic Video Transmission Over DVB-T2 System Using Future Extension Frame

        <P>This paper proposes a complete framework for end-to-end transmission of stereo video for regular services using a digital video broadcasting-terrestrial version 2 (DVB-T2) system. The proposed system incorporates existing services (such as fixed and mobile) to deliver stereoscopic 3-D content in order to maintain backward compatibility without requiring any additional bandwidth. The system is based on a single stream using the future extension frame part of the T2 frame in the DVB-T2 system, which provides the flexibility to carry both (T2-base and T2-lite) profiles of DVB-T2 on a single stream. T2-base and T2-lite are used for left-and right-eye views of stereoscopic video. Finally, a service scenario for the proposed system considering target receivers and the required data rate is analyzed. A case study is given to represent practical applications of the proposed system in scenarios that could be deployed at different times in the mid-term future in high-definition environment.</P>
