        Nonlinear response of laterally loaded rigid piles in sand

        This paper investigates nonlinear response of 51 laterally loaded rigid piles in sand. Measured response of each pile test was used to deduce input parameters of modulus of subgrade reaction and the gradient of the linear limiting force profile using elastic-plastic solutions. Normalised load - displacement and/or moment - rotation curves and in some cases bending moment and displacement distributions with depth are provided for all the pile tests, to show the effect of load eccentricity on the nonlinear pile response and pile capacity. The values of modulus of subgrade reaction and the gradient of the linear limiting force profile may be used in the design of laterally loaded rigid piles in sand.

        Ultimate Lateral Bearing Capacity and Group Effect of Belled Wedge Pile Groups

        Pile foundation of ports, high-voltage transmission line tower are subjected to amount of lateral loading, prediction on lateral bearing capacity is one of the most important projects in structure design. This paper pertains to the model tests on pile-soil interaction of single pile, 2 × 1 and 2 × 2 belled wedge pile groups embedded in sand under lateral load. The load versus displacement, and the soil pressures along depth surrounding piles are measured, the ultimate lateral bearing capacities and group effects of belled wedge piles are analyzed and discussed. An simplified theoretical calculation method on predicting the lateral bearing capacities of shaped pile groups with considering group pile p-y curves, and longitudinal cross-section variation are proposed. The accuracy and reliability of this developed calculation method are verified through the comparative analysis with model test results obtained in this study and previous literature. The predicted values of lateral bearing capacities have suitable agreement with the measured data. It also shows that the ultimate lateral bearing capacities of belled wedge pile group are nearly 1.8 − 2.0 times of those of traditional belled pile with the same concrete usage in this study’s condition.

        Limit analysis of 3D rock slope stability with non-linear failure criterion

        The non-linear Hoek-Brown failure criterion has been widely accepted and applied to evaluate the stability of rock slopes under plane-strain conditions. This paper presents a kinematic approach of limit analysis to assessing the static and seismic stability of three-dimensional (3D) rock slopes using the generalized Hoek-Brown failure criterion. A tangential technique is employed to obtain the equivalent Mohr-Coulomb strength parameters of rock material from the generalized Hoek-Brown criterion. The least upper bounds to the stability number are obtained in an optimization procedure and presented in the form of graphs and tables for a wide range of parameters. The calculated results demonstrate the influences of 3D geometrical constraint, non-linear strength parameters and seismic acceleration on the stability number and equivalent strength parameters. The presented upper-bound solutions can be used for preliminary assessment on the 3D rock slope stability in design and assessing other solutions from the developing methods in the stability analysis of 3D rock slopes.

        Amplification of transglutaminase 2 enhances tumor-promoting inflammation in gastric cancers

        Tumor-promoting inflammation is a hallmark of cancer and is highly associated with tumor progression, angiogenesis, and metastasis. Tumor-associated macrophages (TAMs) are major drivers of tumor-promoting inflammation, but due to the complexity of the tumor microenvironment, the detailed regulatory mechanisms are still under investigation. Here, we investigated a novel role for transglutaminase 2 (TGM2) in the development of tumor-promoting inflammation and recruitment of TAMs to gastric cancer (GC) tissues. When estimated by array comparative genomic hybridization and droplet digital PCR, the copy numbers of the TGM2 gene were amplified in 13.6% (14/103) of GC patients and positively associated with TGM2 expression. Gene set enrichment analysis of expression microarray data for GC samples with high or low TGM2 expression showed that increased TGM2 expression was associated with tumor-promoting inflammation in GC. In addition, the expression of TGM2 was correlated with the expression of markers for macrophages, neutrophils, blood vessels, and lymphatic vessels. Overexpression of TGM2 in GC cells augmented the IL-1β-induced secretion of macrophage-recruiting chemokines and NF-κB activation. TGM2 protein levels were associated with the expression levels of the macrophage marker CD163 in human GC tissue samples. Moreover, GC patients with high expression of TGM2 had a worse prognosis than those with low expression of TGM2. These results suggest TGM2 as a novel regulator of the tumor microenvironment of GC and provide a promising target for constraining tumor-promoting inflammation.
