Functional Development of the Inside of Microtubules Based on a Tau-derived Peptide

Microtubules are tubular cytoskeletons with a 15 nm inner diameter, which are formed by the polymerization of tubulins. Although microtubule-based nanomaterials have been developed by functionalization of their outer surface, the inside of microtubules has not been used because of no methodology. The encapsulation of nanostructures will be a new concept to functionalize microtubules. Herein, we developed a methodology to encapsulate various molecules inside microtubules. Firstly, we constructed a Tau-derived peptide (TP) which binds to a pocket inside microtubules (Chem. Eur. J., 2018). By conjugating TP, green fluorescent protein (GFP) (Chem. Commun., 2019) and magnetic nanoparticles (Nano Lett., 2020) were successfully encapsulated inside microtubules. Also, we showed the binding of TP to the microtubules in living cells (ACS Omega, 2019). These results indicate that the peptide-based encapsulation is a promising approach to construct new microtubule-based materials.

Background: Type 1 diabetes mellitus induced by immune-checkpoint inhibitors (ICI-T1DM) is a rare critical entity. However, the etiology of ICI-T1DM remains unclear. Methods: In order to elucidate risk factors for ICI-T1DM, we evaluated the clinical course and immunological status of patients with ICI-T1DM who had been diagnosed during 2016 to 2021. Results: Seven of 871 (0.8%, six men and one woman) patients developed ICI-T1DM. We revealed that the allele frequencies of human leukocyte antigen (HLA)-DPA1*02:02 and DPB1*05:01 were significantly higher in the patients with ICI-T1DM In comparison to the controls who received ICI (11/14 vs. 10/26, P=0.022; 11/14 vs. 7/26, P=0.0027, respectively). HLA-DRB1*04:05, which has been found to be a T1DM susceptibility allele in Asians, was also observed as a high-risk allele for ICI-T1DM. The significance of the HLA-DPB1*05:01 and DRB1*04:05 alleles was confirmed by an analysis of four additional patients. The absolute/relative neutrophil count, neutrophils-lymphocyte ratio, and neutrophil-eosinophil ratio increased, and the absolute lymphocyte count and absolute/relative eosinophil count decreased at the onset as compared with 6 weeks before. In two patients, alterations in cytokines and chemokines were found at the onset. Conclusion: Novel high-risk HLA alleles and haplotypes were identified in ICI-T1DM, and peripheral blood factors may be utilized as biomarkers.

Distribution of marine type of Aquabirnavirus (MABV) was examined in shellfish and fish from Okinawa and Ishigaki Islands, Japan, where water temperature is higher than 25°C through the year. Genome detection and virus isolation were performed for shellfish and fish samples, and the results revealed the prevalent distribution of MABV in diverse species in the area, although isolation was not frequently. Detection rate of MABV genome in bivalves was higher than gastropods, which was similar result to former report in mainland of Japan. Furthermore, the unique five-nucleotide deletion was found with a high rate of occurrence in the MABV genome from shellfish and fish. This study showed distribution status of MABV in organisms in subtropical waters by wide monitoring, and discovered new genome variation in VP2/NS region of this virus.

In order to evaluate the water quality (N, P and C) and the biological mass balance of semi-enclosed brackish Lake Obuchi, Japan, an ecosystem model was developed and applied to the lake, using the flow field calculated by a hydrodynamic model. The time series data of the observed tide level, river discharge and meteorological parameters from January 2001 to December 2002 were incorporated as the parameters of the hydrodynamic model. Water quality and biomass balance were estimated by the ecosystem model, and simulated fluctuations in water quality agreed with our observations. The carbon contents of POC, phytoplankton and zooplankton in the lake were calculated by the model at an average 7200, 1500 and 22 ㎏, respectively, which somewhat agreed with our observations of POC (5900 ㎏), phytoplankton (3800 ㎏), and zooplankton (150 ㎏).
