      • Self-Assembled Hierarchical Superstructures from the Benzene-1,3,5-Tricarboxamide Supramolecules for the Fabrication of Remote-Controllable Actuating and Rewritable Films

        <P>The well-defined hierarchical superstructures constructed by the self assembly of programmed supramolecules can be organized for the fabrication of remote-controllable actuating and rewritable films. To realize this concept, we newly designed and synthesized a benzene-1,3,5-tricarboxamide (BTA) derivative (abbreviated as BTA-3AZO) containing photoresponsive azobenzene (AZO) mesogens on the periphery of the BTA core. BTA-3AZO was first self-assembled to nanocolumns mainly driven by the intermolecular hydrogen-bonds between BTA cores, and these self-assembled nanocolumns were further self-organized laterally to form the low-ordered hexagonal columnar liquid crystal (LC) phase below the isotropization temperature. Upon cooling, a lamello-columnar crystal phase emerged at room temperature via a highly ordered lamello-columnar LC phase. The three-dimensional (3D) organogel networks consisted of fibrous and lamellar superstructures were fabricated in the BTA-3AZO cyclohexane-methanol solutions. By tuning the wavelength of light, the shape and color of the 3D networked thin films were remote-controlled by the conformational changes of azobenzene moieties in the BTA-3AZO. The demonstrations of remote-controllable 3D actuating and rewritable films with the self-assembled hierarchical BTA-3AZO thin films can be stepping stones for the advanced flexible optoelectronic devices.</P>

        RNA sequencing reveals that Prx II gene knockout can down-regulate the allograft rejection of dermal mesenchymal stem cells

        In this study, we used RNA sequencing (RNA-seq) to analyze and compare bulk cell samples from wild-type (WT) dermal mesenchymal stem cells (DMSCs) (n = 3) and Prx II knockout DMSCs (n = 3). The purpose of the study was to elucidate the role of Prx II on allogeneic immune rejection of transplanted DMSCs. The results revealed differential expression of 472 genes (176 up-regulated and 296 down-regulated; p ≤ 0.05) between the PrxII+/+ (WT) and PrxII−/− sample groups. When highly regulated genes were categorized according to the Gene Ontology (GO) molecular function classification and the Kyoto Encyclopedia of Genes and Genomes (KEGG) pathway analysis, the PrxII−/− samples showed a robust downward trend in allograft rejection. The study identified 43 all immunologically rejected differentially expressed genes, of which 41 showed lower expression in the PrxII−/− vs. PrxII+/+ (WT) samples. These findings suggest that Prx II gene knockout may down-regulate the allograft rejection that occurs during DMSCs transplantation and improve the survival rate of DMSCs in the host. This study provides a new perspective on the clinical treatment of stem cell transplantation.

        Anti-inflammatory effect of hispidin on LPS induced macrophage inflammation through MAPK and JAK1/STAT3 signaling pathways

        Severe inflammatory reactions caused by macrophage activation can trigger a systemic immune response. In the present study, we observed the anti-inflammatory properties of hispidin on LPS induced RAW264.7 macrophage cells. Our results showed that hispidin treatment significantly reduced the production of cellular NO, IL-6 and reactive oxygen species (ROS) while has not inhibitory effect on TNF-α productions. Excitingly, hispidin treatment retains the phagocytosis ability of macrophages which enabling them to perform the function of removing foreign invaders. Signaling studies showed, hispidin treatment dramatic suppressed the LPS induced mitogen activated protein kinases (MAPK) and JAK/STAT activations. In conclusion, our findings suggest that hispidin may be a new therapeutic target for clinical treatment of macrophages-mediated inflammatory responses.

        Identification and expression specificity of chemosensory genes in the male reproductive system of Spodoptera exigua

        Some chemosensory genes are found to be expressed specifically or highly in the male reproductive system, suggesting their importance in male reproductive physiology. In this paper, we identified chemosensory genes expressed in the male reproductive system of the moth pest Spodoptera exigua by transcriptome and proteome sequencing, and further determined expression patterns of these genes regarding antennae and different tissues of the male reproductive system (testes, vas deferens, seminal vesicles, ejaculatory ducts, male accessory glands) by the real-time quantitative PCR (qPCR). Transcriptome analysis showed that 46 chemosensory genes were expressed in the male reproductive system of S. exigua, including 18 odorant binding proteins (OBPs), 12 che mosensory proteins (CSPs), 10 odorant receptors (ORs) and 6 other chemosensory genes. Further qPCR mea surements revealed that 14 chemosensory genes were expressed specifically or highly in the male reproductive tissues, relative to that in male antennae. Of the 14 genes, OBP17, OBP22, OBP27 and GR43 were specifically expressed in the testes; OBP20 was specifically expressed in the vas deferens; and PBP4 was specifically expressed in the male accessory glands. In addition, proteome data of the spermatophore confirmed that at least 4 OBPs and one CSP were transmitted from males to females, suggesting their function in the reproductive physiology of mated females. The results provide insights into the functional diversity of chemosensory genes, and bases for exploring functions of these chemosensory genes in male reproductive physiology.

      • INVESTIGATING THE DEVELOPMENT OF BRAND LOYALTY IN BRAND COMMUNITIES FROM A POSITIVE PSYCHOLOGY PERSPECTIVE

        Brand communities have been increasingly used by marketers to build brands. A brand community can be defined as a “…group of consumers with a shared enthusiasm for the brand and a well-developed social identity, whose members engage jointly in group actions to accomplish collective goals and/or express mutual sentiments and commitments” (Bagozzi and Dholakia, 2006, p. 45). Recent research on brand communities has begun to identify the importance of consumers’ psychological processes in regard to developing successful brand communities. Based on the flow theory from positive psychology, we propose that flow could be generated by brand community characteristics and plays an important role in influencing brand community members’ attitudes toward the brand. Specifically, we propose a model that identifies brand community characteristics (i.e., community cohesiveness and information quality) that produce flow experience and how the flow experience impacts brand identification and brand loyalty. Members from 31 automobile brand communities participated in this survey study, and 580 validated questionnaires were returned. Structural Equation Model was used to test the research hypotheses. The results show that community cohesiveness and information quality positively directly influence brand identification and also indirectly influence brand identification via flow. Flow also positively influences members’ brand identification and, subsequently, impacts brand loyalty. The results from our research contribute to the branding, brand community, and flow theory literature.

        Research on Mechanics of Longitudinal Joint in Shield Tunnel by the Nonlinear Spring Equivalent Method

        Shield tunnel lining is mainly composed of prefabricated segment, accurate description of longitudinal seam joint mechanics is important for design and analysis of shield tunnel segmental linings, however, due to its stress state is complex and internal force changes show the characteristics of nonlinear, there is no more rapid and accurate calculation model to analysis the mechanics. In the view of the problem of the entity unit simulating bolt bending with low computing speed, the model of shield tunnel longitudinal seam that using the nonlinear spring to simulate bending bolt joint is proposed. Combined with longitudinal seam joint test, analysis the entity model and spring bolt model, the study concluded that: 1) the calculation results of spring bolt model are closer to the test results than entity unit model; and 2) calculation efficiency of spring bolt model bolt up 1 times than the physical model. Based on the spring bolt model, further study the relationship between viscous coefficient, friction coefficient and calculation efficiency, and analyzes the regularity of deformation and damage of shield tunnel connection.

        Experimental Study of Composite Steel Plate Shear Wall with Flush End-Plate Connection

        This paper presents an investigation on the structural performance of steel plate shear wall (SPSW) with fl ush end-plate beam–column connections and infi ll precast reinforced concrete (precast RC) panels. Two single-span two-story SPSW specimens, including unstiff ened SPSW (NBRP) and precast RC panel restrained SPSW (Con-BRP), are fi rst tested, followed with a parametric study by fi nite element method. Precast RC cover panels are installed on both sides of the infi ll steel plate and are disconnected from steel frame. Test results indicate that the use of precast RC cover panels increases the load carrying and energy dissipation capacities of the SPSW structure, but decreases its ductility. It is also eff ective in reducing the inward fl exural deformation of columns. Moreover, the stiff ening eff ect of the infi ll steel plate on the beam–column connections remains, which is a result of the precast RC cover panel’s resistance to the local buckling and the tears of the infi ll steel plates. The infl uence of the gap size between the precast RC cover plate and frame members on the failure mode of the specimen Con-BRP is also investigated, based on which a maximum gap size is recommended. Conclusions are drawn that SPSW structure with fl ush end-plate beam–column connections and precast RC cover panels fully exploits the strength of infi ll steel plates and exhibits excellent structural performance.

      • Seismic response of combined retaining structure with inclined rock slope

        A gravity wall combined with an anchoring lattice frame (a combined retaining structure) is adopted at a typical engineering site at Dali-Ruili Railway Line China. Where, the combined retaining structure supports a soil deposit covering on different inclined rock slopes. With an aim to investigate and compare the effects of inclined rock slopes on the response of combined retaining structure under seismic excitation, three groups of shaking table tests are conducted. The rock slopes are shaped as planar surfaces inclined at angles of 20°, 30°, and 40° with the horizontal, respectively. The shaking table tests are supplemented by dynamic numerical simulations. The results regarding the horizontal acceleration response, vertical acceleration response, permanent displacement mode, and axial anchor force are comparatively examined. The acceleration response is more susceptible to outer structural profile of combined retaining structure than to inclined angle of rock slope. The permanent displacement decreases when the inclined angle of the rock slope increases within a range of 20°-40°. A critical inclined angle of rock slope exists within a range of 20°-40°, and induces the largest axial anchor force in the combined retaining structure.

      • Characterization and anti-tumor activity of saponin-rich fractions of South Korean sea cucumbers (Apostichopus japonicus)

        In this study, the saponin-rich fractions of five individual (two Red and three Black) sea cucumbers (Apostichopus japonicus) in South Korea were investigated for their antiproliferative effect against HL-60, B16F10, MCF-7, and Hep3B tumor cell lines. The red sea cucumber saponin-rich fraction (SSC) from Jeju Island (JRe) decreased the growth of HL-60 with an IC50 value of 23.55 ± 3.40 lg/mL, which represented the strongest anticancer activity among the extracts. Further, SSC downregulated B-cell lymphoma extra-large (Bcl-xL), while upregulating, to different degrees, Bcl-2-associated X protein (Bax), caspase-9, caspase-3, PARP cleavage, and apoptotic bodies in cancer cells. Evidence for SSC inducing apoptosis via the mitochondria-mediated pathway was found. The contents of SSCs were determined using ultra high-performance liquid chromatography coupled with a quadrupole orbitrap mass spectrometry to comparatively evaluate the regional influence. In West Sea, the total SSC content of A. japonicus was 15.5 mg/g, representing the highest content, while A. japonicus in the South Sea yielded the lowest content at 8 mg/g. The major saponin constituent in SSC was identified as Holotoxin A1, which may the anti-tumor compound in A. japonicus.
