        Foxp3 is a key downstream regulator of p53-mediated cellular senescence

        <P>The downstream events and target genes of p53 in the process of senescence are not fully understood. Here, we report a novel function of the forkhead transcription factor Foxp3, which is a key player in mediating T-cell inhibitory functions, in p53-mediated cellular senescence. The overexpression of Foxp3 in mouse embryonic fibroblasts (MEFs) accelerates senescence, whereas Foxp3 knockdown leads to escape from p53-mediated senescence in p53-expressing MEFs. Consistent with these results, Foxp3 expression resulted in the induction of senescence in epithelial cancer cells, including MCF7 and HCT116 cells. Foxp3 overexpression also increased the intracellular levels of reactive oxygen species (ROS). The ROS inhibitor N-acetyl-L-cysteine rescued cells from Foxp3-expression-induced senescence. Furthermore, the elevated ROS levels that accompanied Foxp3 overexpression were paralleled by an increase in p21 expression. Knockdown of p21 in Foxp3-expressing MEFs abrogated the Foxp3-dependent increase in ROS levels, indicating that Foxp3 acts through the induction of p21 and the subsequent ROS elevation to trigger senescence. Collectively, these results suggest that Foxp3 is a downstream target of p53 that is sufficient to induce p21 expression, ROS production and p53-mediated senescence.</P>

      • Kinetics determination of electrogenerated hydrogen peroxide (H<sub>2</sub>O<sub>2</sub>) using carbon fiber microelectrode in electroenzymatic degradation of phenolic compounds

        The kinetics of electrogenerated hydrogen peroxide (H<SUB>2</SUB>O<SUB>2</SUB>), which can activate peroxidases in an electroenzymatic process, was examined by an amperometric technique using a carbon fiber microelectrode that was modified by polyaniline (PAn) film and platinum particles. The electrogeneration of H<SUB>2</SUB>O<SUB>2</SUB> was found to be dependent on the pH and applied potential, and resulting in a variable current response of the carbon fiber microelectrode. The highest amount of H<SUB>2</SUB>O<SUB>2</SUB> was electrogenerated when 2.3V was applied between the Pt/Ti anode and a reticulated vitreous carbon (RVC) cathode at pH 6.0, with a current response of 0.0190μAmin<SUP>-1</SUP>. Phenol was completely degraded by the electroenzymatic reaction of the immobilized horseradish peroxidase (HRP), and the time required for the electrogeneration of H<SUB>2</SUB>O<SUB>2</SUB> increased according to the initial concentration of phenol. The degradation stoichiometric ratio between the electrogenerated H<SUB>2</SUB>O<SUB>2</SUB> and the aqueous phenol under HRP immobilized on RVC was found to be 1:1.

        <P>By combining hydrolysis, it was observed that the maximum concentration of xylose + mannose + galactose (xmg) was 10.7 g/L at a CS of 1.72. The aliphatic acid, aromatic acid and aldehydes were released 29.8, 0.45 and 0.90 from the black liquor at CS of 1.19, 1.72, and 2.19, respectively. In the gasification using bench-scale bubbling fluidized bed (BFB) reactor, high-quality syngas (H-2/CO: 1.47) was obtained from the steam of acid-insoluble lignin (AIL), suitable for conventional energy systems such as gas engines, turbines, and boilers.       • A new class of COX-2 inhibitor, rutaecarpine from Evodia rutaecarpa

        Objective and Design: We investigated the effect of a new class of COX-2 inhibitor, rutaecarpine, on the production of PGD, in bone marrow derived mast cells(BMMC) and PGE_(2) in bone marrow derived mast cells (BMMC) and PGE_(2) in COX-2 transfected H다293 cells. Inflammation was induced by λ-carrageenan in male Splague-Dawley(SD) rats. Material: Rutaecarpine(8, 13-Dihydroindolo[2', 3':3,4]pyridol[2,1-b]quinazolin-5(7H)-one) was isolated from the fruits of Evodia rutaecarpa. BMMC were cultured with WEHI-3 conditioned medium. c-Kit ligand and IL-10 were obtained by their expression in baculovirus. Methods: The generation of PGD_(2) and PGE_(2) were determined by their assay kit. COX-1 and COX-2 protein and mRNA expression was determined by BMMC in the presence of KL, LPS and IL-10. Treatment: Rutaecarpine and indomethacin dissolved in 0.1% carboxymethyl cellulose was administered intraperitoneally and, 1h later, λ-carrageenan solution was in-jucted to right hind paw of rats. Paw volumes were measured using plethysmometer 5h after λ-carrageenan injection. Results: Rutaecarpine inhibited COX-2 and COX-1 dependent phases of PGD_(2) generation in BMMC in a concentration-dependent manner with an IC_(50) of 0.28 μM and 8.7μM, respectively. It inhibited COX-2-dependent conversion of exogenous arachidonic acid to PGE_(2) in a dose-dependent manner by the COX-2-transfected HEK293 cells. However, rutaecarpine inhibited neither PLA_(2) and COX-1 activity nor COX_(2) protein and the mRNA expression up to the concentration of 30μM in BMMC, indicating that retaecarpine directly inhibited COX-2 activity. Furthermore, rutaecarpine showed in vivo anti-inflammatory activity on rat λ-carrageen-an induced paw edema by intraperitoneal administration. Conclusion: Anti-inflammatory activity of Evodia rutaecarpa could be attributed at least in part by inhibition of COS-2.

      • Pollen morphology of the tribe Sorbarieae (Rosaceae)

        <P>The pollen morphology of the tribe Sorbarieae (Adenostoma, Chamaebatiaria, Sorbaria, and Spiraeanthus) and two related genera Gillenia and Lyonothamnus was investigated by light, scanning electron, and transmission electron microscopy. Sorbarieae pollen was monad, tri-colporate, small to medium in size (P = 10.2-40.0 mu m, E = 10.8-32.4 mu m), and oblate to prolate in shape (P/E = 0.74-1.88). The sexine ornamentation was striate, but four different types could be divided: striate-psilate, striate-plicate, striate-microechinate, and striate-perforate. The pollen wall stratification typically consisted of unbranched columellae and a continuous endexine. As an additional palynological characteristic, orbicules (small sporopollenin granules) were observed in all taxa and thus a possible synapomorphic character of Sorbarieae as a whole. The pollen characteristics are useful to recognize certain taxa. Gillenia is the only genus with both a pore flap on the aperture and the perforated sexine. Lyonothamnus has a significantly thick exine and relatively larger pollen grains, which is compared to that of Sorbarieae. Some quantitative characteristics (e.g., pollen and colpus size, exine thickness, and ridge width of striae) identified using principal components analysis (PCA) may have diagnostic importance among the taxa in the tribe.</P>

        p34 is a novel regulator of the oncogenic behavior of NEDD4-1 and PTEN

        PTEN is one of the most frequently mutated or deleted tumor suppressors in human cancers. NEDD4-1 was recently identified as the E3 ubiquitin ligase for PTEN; however, a number of important questions remain regarding the role of ubiquitination in regulating PTEN function and the mechanisms by which PTEN ubiquitination is regulated. In the present study, we demonstrated that p34, which was identified as a binding partner of NEDD4-1, controls PTEN ubiquitination by regulating NEDD4-1 protein stability. p34 interacts with the WW1 domain of NEDD4-1, an interaction that enhances NEDD4-1 stability. Expression of p34 promotes PTEN poly-ubiquitination, leading to PTEN protein degradation, whereas p34 knockdown results in PTEN mono-ubiquitination. Notably, an inverse correlation between PTEN and p34/NEDD4-1 levels was confirmed in tumor samples from colon cancer patients. Thus, p34 acts as a key regulator of the oncogenic behavior of NEDD4-1 and PTEN.

      • Hydrogen sulfide diminishes the levels of thymic stromal lymphopoietin in activated mast cells

        <P>Bamboo salt (BS) is a Korean traditional type of salt and has been reported to have therapeutic effects on allergic inflammation. Thymic stromal lymphopoietin (TSLP) aggravates inflammation in the pathogenesis of allergic reactions, such as allergic rhinitis (AR). To confirm an active compound of BS, we investigated the effect of sulfur, a compound of BS, on the levels of TSLP in a human mast cell line, HMC-1 cells and a mouse model of AR using hydrogen sulfide (H2S) donor, sodium hydrosulfide (NaSH). We treated NaSH or BS in HMC-1 cells and activated the HMC-1 cells with phorbol myristate acetate and calcium ionophore A23187 (PMACI). ELISA for the production measurement of TSLP, PCR for the mRNA expression measurement of TSLP, and western blot analysis for the expression measurement of upstream mediators were performed. Mice were treated with NaSH and sensitized with ovalbumin (OVA). The levels of TSLP were measured in serum and nasal mucosa tissue in an OVA-induced AR mouse model. NaSH or BS diminished the production and mRNA expression of TSLP as well as interleukin (IL)-6 and tumor necrosis factor (TNF)-alpha in the PMACI-activated HMC-1 cells. NaSH or BS diminished the level of intracellular calcium in the PMACI-activated HMC-1 cells. NaSH or BS reduced the expression and activity of caspase-1 in the PMACI-activated HMC-1 cells. And NaSH or BS inhibited the expression of receptor interacting protein-2 and the phosphorylation of extracellular signal-regulated kinase in the PMACI-activated HMC-1 cells. The translocation of NF-kappa B into the nucleus as well as the phosphorylation and degradation of I kappa B alpha in the cytoplasm were diminished by NaSH or BS in the PMACI-activated HMC-1 cells. Furthermore, NaSH inhibited the production of TSLP, IL-6, and IL-8 in TNF-alpha-activated HMC-1 cells. Finally, the administration of NaSH showed a decrease in number of rubs on mice with OVA-induced AR. And the levels of immunoglobulin E and TSLP in the serum and the level of TSLP in the nasal mucosa tissue of the OVA-induced AR mice were reduced by NaSH. In conclusion, these findings show that H2S, as an active compound of BS is a potential agent to cure allergic inflammation.</P>

        A new technique using search method based on fuzzy multi-criteria function is proposed for generator maintenance scheduling. Minimization of probabilistic production cost, maximization of system reliability level and air pollution are considered for fuzzy multi-criteria function. To obtain an optimal solution for generator maintenance scheduling under fuzzy environment, fuzzy multi-criteria relaxation method(fuzzy search method) is used. The practicality and effectiveness of the proposed approach are demonstrated by the simulation results of the real size model system.

        중 에너지 이온산란 분광장치의 제작 및 성능 평가

        이온-고체 표면 사이의 상호작용에 관한 연구를 수행하기 위하여 중 에너지 이온산란 분광장치를 개발하였고 그 특성 평가를 수행하였다. 제작된 MEIS의 에너지 분해능은 4×10^(-3)으로 측정되었다. MEIS의 표면분석의 응용으로 60 keV H^+을 Ta₂O_5(300 Å)/Si에 적용하여 에너지 손실인자와 깊이 분해능을 얻은 결과는 42eV/Å와 9.7 Å이었다. 또한, Si(100) 표면에 97.5 keV H^+이온을 random 방향으로 입사시켜 이차윈 스펙트럼을 얻었다. A medium energy ion scattering spectroscopy(MEIS) system has been developed and tested. In the MEIS system a toroidal electrostatic energy analyzer(TEA) and a two dimensional position sensitive detector(PSD) were used. The energy resolution of MEIS system was estimated to be less than 4×10^(-3) and the overall angular resolution was less than 0.3°. From the MEIS spectrum of Ta₂O_5(300 Å) on Si analyzed using 60 keV H^+, the energy loss factor[S] and depth resolution were estimated to be 42 eV/Å and 9.7 Å, respectively. Also Si(100) surface was analyzed using the MEIS system. A random MEIS spectrum was obtained from the Si(100) covered with native oxide layers. At the double alignment condition, MEIS spectrum showed a Si surface peak, a oxygen peak and a carbon peak.

        Anti-inflammatory effects of extract from Haliotis discus hannai fermented with Cordyceps militaris mycelia in RAW264.7 macrophages through TRIF-dependent signaling pathway

        In this study, Haliotis discus hannai (H. discus hannai) fermentation was attempted with Cordyceps militaris (C. militaris) mycelia using a solid culture. We tried to ferment H. discus hannai to determine the optimal conditions fermentation with regards to its anti-inflammatory effects. The extracts of H. discus hannai fermented with C. militaris mycelia (HFCM-5) showed higher nitric oxide inhibitory effects than H. discus hannai and C. militaris alone. HFCM-5 also decreased pro-inflammatory cytokines, TNF-α and IL-6 in a dose-dependent manner. HFCM-5 did not affect the MyD88-dependent pathway, but decreased phosphorylation of IRF3 and STAT1 which are involved in TRIF-dependent pathway. Taken together, our results suggest that HFCM-5 exerts its anti-inflammatory effects via TRIF signaling pathway and could potentially be used as a functional food in the regulation of inflammation.
