        Berberine Prevents Intestinal Mucosal Barrier Damage During Early Phase of Sepsis in Rat through the Toll-Like Receptors Signaling Pathway

        Our previous study has shown berberine prevents damage to the intestinal mucosal barrier during early phase of sepsis in rat through mechanisms independent of the NOD-like receptors signaling pathway. In this study, we explored the regulatory effects of berberine on Toll-like receptors during the intestinal mucosal damaging process in rats. Male Sprague-Dawlay (SD) rats were treated with berberine for 5 d before undergoing cecal ligation and puncture (CLP) to induce polymicrobial sepsis. The expression of Toll-like receptor 2 (TLR 2), TLR 4, TLR 9, the activity of nuclear factor-kappa B (NF-κB), the levels of selected cytokines and chemokines, percentage of cell death in intestinal epithelial cells, and mucosal permeability were investigated at 0, 2, 6, 12 and 24 h after CLP. Results showed that the tumor necrosis factor-α (TNF-α) and interleukin-6 (IL-6) level were significantly lower in berberine-treated rats compared to the control animals. Conversely, the expression level of tight junction proteins, percentage of cell death in intestinal epithelial cells and the mucosal permeability were significantly higher in berberine-treated rats. The mRNA expression of TLR 2, TLR 4, and TLR 9 were significantly affected by berberine treatment. Our results indicate that pretreatment with berberine attenuates tissue injury and protects the intestinal mucosal barrier in early phase of sepsis and this may possibly have been mediated through the TLRs pathway.

        LMI Stability Criterion with Less Variables for Time-delay Systems

        Slack variables approach is an important technique for tackling the delay-dependent stability problem for systems with time-varying delay. In this paper, a new delay-dependent stability criterion is presented without introducing any slack variable. The technique is based on a simply integral inequal-ity. The result is shown to be equivalent to some existing ones but includes the least number of vari-ables. Thus, redundant selection and computation can be avoided so that the computational burden can be largely reduced. Numerical examples are given to illustrate the effectiveness of the proposed stabil-ity conditions.

        The complete mitochondrial genome of Hemigrapsus sinensis (Brachyura, Grapsoidea, Varunidae) and its phylogenetic position within Grapsoidea

        Background In this study, the complete mitogenome of Hemigrapsus sinensis was the first identified and analyzed. Objective The complete mitochondrial genome of Hemigrapsus sinensis (Brachyura, Grapsoidea, Varunidae) and its phylogenetic position within Grapsoidea. Methods The sample of Hemigrapsus sinensis was collected and DNA was extracted. After sequencing, NOVOPlasty was used for sequence assembly. Annotate sequences with MITOS WebServer, tRNAscan-SE2.0, and NCBI database. MEGA was used for sequence analysis and Phylosuite was used for phylogenetic tree construction. DnaSP was used to calculate Ka/Ks. Results This mitochondrial genome shows that it was 15,900 bp and encoded 13 PCGs, 22 tRNA genes, two rRNA genes, and one control region. The genome composition tends to A + T (74.34%) and presents a negative GC-skew (− 0.22) and AT-skew (− 0.03). The PCGs initiation codon was the typical ATN and termination codon was the typical TAN, incomplete T or missing. The ML and BI trees showed that H. sinensis was most closely related to Hemigrapsus and clustered together with the Varunidae. And our phylogenetic trees provide proof that Ocypodoidea and Grapsoidea may be of common origin. Meanwhile, in the phylogenetic tree, parallel mixing of Chiromantes and Orisarma raised doubts over the traditional classification system. Besides, Incomplete Lineage sorting (ILS) was observed in Varunidae. In the subsequent analysis of evolution rate, we found that all of the PCGs (NAD4 was not calculated) had undergone negative selections, indicating the conservation of mitochondrial genes of H. sinensis during the evolution. Conclusion Therefore, researching the complete mitogenome of H. sinensis would be contributing to molecular taxonomy, phylogenetic relationship, and breeding optimization within the Grapsoidea superfamily.

      • Clinical Significance of Quantitative Analysis of Plasma Epstein-Barr Virus DNA in Patients of Xinjiang Uygur Nationality with Hodgkin's Lymphoma

        Objective: To investigate the relationship between plasma EBV-DNA concentration and clinicopathologic features of Hodgkin's lymphoma cases. Methods: At first, the positive rate of plasma EBV-DNA was determined with a nested-PCR method using 45 specimens from Uygur HL patients, as well as 110 healthy people sampled as normal controls. Secondly, using fluorescent quantitative nested-PCR, EBV viral load was assessed in the EBV-DNA positive plasma samples. Then, relationships between plasma EBV viral load and clinicopathologic features of HL patients were analyzed. Results: The positive rate of plasma EBV-DNA of HL patients was significantly higher than that of normal controls (53.3% vs 26.4%, P=0.001). There was no significant difference about plasma EBV viral load between EBV-associated HL and EBV-DNA positive normal people (P=0.490). Looking at patients' characteristics, plasma EBV viral load in 10-20 years EBV-associated HL was higher than in EBV cases which were less than 10 years or more than 35 years (P=0.025). Furthermore, in EBV-associated HL, concentration of plasma EBV-DNA was significantly higher in advanced stage disease (stages III-IV; P=0.013), and with B-symptoms (P=0.020). Conclusion: EBV-DNA levels were associated with part of clinicopathologic features of cases. It was of practical use to screen HL. Further etiological studies appear warranted.

        C-Phycocyanin supplementation during <i>in vitro</i> maturation enhances pre-implantation developmental competence of parthenogenetic and cloned embryos in pigs

        <P><B>Abstract</B></P> <P>C-Phycocyanin (C-PC), a protein from green microalgae, has been suggested to possess various biological activities, including antioxidant and free radical scavenging properties. The aim of the current study was to explore the effects of C-PC on the maturation of porcine oocytes and subsequent developmental competence after parthenogenetic activation and somatic cell nuclear transfer (SCNT) as well as the underlying mechanisms. There was no significant improvement in nuclear maturation rates between the control and C-PC supplementation groups (1, 3, 5, 10 μg/mL). However, supplementation of 5 μg/mL C-PC in the maturation medium significantly increased blastocyst formation and hatching rates after parthenogenetic activation (59.6 ± 3.6% and 33.0 ± 2.6% vs. 49.8 ± 3.5% and 27.4 ± 2.4%, respectively). In addition, the presence of C-PC during the maturation period significantly improved blastocyst formation rates and total cell numbers after SCNT (24.8 ± 1.9% and 42.2 ± 3.3 vs. 21.6 ± 2.2% and 39.5 ± 3.4, respectively) compared to the control group. Furthermore, cellular proliferation and the expression of pluripotency-related genes (<I>SOX2</I> and <I>NANOG</I>) were increased in cloned blastocysts derived from the C-PC supplemented group. Importantly, C-PC supplementation during maturation not only improved cumulus expansion and increased the expression of cumulus expansion-related genes (<I>HAS2</I>, <I>PTX3</I>, and <I>PTGS2</I>), but also enhanced antioxidant capacity, improved mitochondria function, and decreased cathepsin B activity in porcine oocytes. These results demonstrate that C-PC may be useful for improving porcine oocyte quality and subsequent developmental competence in embryos.</P> <P><B>Highlights</B></P> <P> <UL> <LI> C-Phycocyanin significantly increased the developmental competence of porcine parthenogenetic and cloned embryos. </LI> <LI> C-Phycocyanin enhanced antioxidant capacity, improved mitochondria function, and decreased cathepsin B activity in porcine oocytes. </LI> </UL> </P>

      • 林蛙(Rana temporaria chinensis David) 皮膚細胞의 徵細構造

        中國 長白山에서 採集된 林蛙(Rana temporaria chinensis David)의 背部와 腹部 皮膚 組織을 pH 7.2에서 0.1M phosphate buffer로 緩衝시킨 2.5% glutaraldehyde와 2% osmium tetroxide 에 前.後 固定하여,各級 ethyl alcohol과 acetone으로 脫水, Epon 812 misture에 包埋하여 LKB ultram-icrotome으로 超薄切片을 만들어 uranyl acetate와 lead citrate,로 染色하여 JEM 100 CX-Ⅱ 電子顯微鏡으로 觀察하여 다음과 같은 結果를 얻었다. 1. 林蛙의 皮膚는 多層으로 種子層, 棘細胞層,顆粒層 및 角質層으로 構成되었는데 角質層은 완전히 角質化한 細胞와 semi-cornified cell도 있다. 2. 種子層 細胞는 背部와 腹部 공히 細胞質 대부분을 차지하는 rough endoplasmic reticulum이 他層과 區分되며, mitochondria,Golgi complex, 電子密度를 달리하는 顆粒 等이 發達하였으며 密集된 microvilli 發達하였다. 3. 棘細胞層은 腹部와 背部가 공히 多角形의 細胞로 이루어졌으며 他層에 비하여 desmosome 이 發達하였고 細胞質 周邊部에는 tonofibril이 있다. 背部細胞는 腹部 棘細胞보다 많은 顆粒을 나타낸다. 4. 顆粒層은 內外 二層으로 構成되었고, 대부분의 細胞質들은 電子密度가 비슷하나,內層에서 間歇的으로 電子密度가 낮은 細胞가 많이 있다. 5. 角質層은 角質細胞 사이에 微細管이 zig zag로 顆粒層까지 連結하였고 角質層에는 semi-cornified cell이 있다. 角質細胞質에는 microfilament, 細胞顆粒으로 차 있으며 멜라닌顆粒이 觀察된다. The authors observed the ultrastructure of the dorsal and ventral skin of the frog (Rana temporaria chinensis David)during the active phases. The specimens from the skin were fixed in 2.5% glutaraldehyde-paraformadehyde fixative in 0.1M phosphate buffer at pH 7.2 prior to fixation in 2% osmium tetroxide, dehydrated in graded ethyl alcohol and acetone, embedded in Epon 812 mixture,and sectioned with LKB-Ultramicrotome. The ultrathin section were contrasted with uranyl acetate and lead citrate and observed with a JEOL-100cx-Ⅱelectron microscope. The results were as follows. 1. Frog epidermis consisted stratified epithelium and was divided into four strata; stratum germinativum,stratum spinosum,stratum granulosum and stratum corneum. 2. Stratum germinativum: In the dorsal and ventral, the cells were columnar in form, rough endoplasmic reticula were specially developed in the whole cytoplasm and mitochondria,Golgi complex and granules were well developed in the cytoplasm. Many microvilli were ag-gregated densely in the intercellular space. 3. Stratum spinosum,:In the dorsal and ventral, the cells were polygonal in form, and desmosome and tonofibrils were well developed in the pericytoplasm. In the dorsal cells were many granules more than the ventral cells. 4. Stratum granulosum: In the dorsal and ventral. it consisted of the inner and outer layers with granules of the same electron density, but intermittently in the inner layer with lower electron density. 5. Stratum corneum: In the dorsal and ventral. it was separated from the underling outer stratum granulosum by the subcorneal space and in the layers with zigzag channel to the un-derling outer stratum granulosum. Semi-cornifid and conified cell were squamous in form.

        林蛙의 皮膚色素胞인 멜라닌色素胞,黃色素胞 및 紅色素胞를 電子顯微鏡으로 觀察하여 다음과 같은 結果를 얻었다. 멜라닌色素胞 : 멜라닌色素胞의 全 細胞質에는 電子密度가 같은 멜라닌 色素顆粒으로 충만되어 있으며, 核은 細胞質 한쪽으로 치우쳤고 楕圓形이거나 不定形이다. 대부분의 멜라닌色素胞는 細胞質突起를 가지며 黃色素胞와 紅色素胞 사이에 나타나는 경우가 많았다. 黃色素胞 : 黃色素胞는 다수의 pterinosome과 소수의 carotenoid vesicle의 色素小器官으로 構成되었다. 그중 pterinosomem 은 小囊 속의 內容物質에 따라, 內容物質이 없는 것을 第1型 pterinosome, 小囊 속에 纖維質이 散在한 것을 第 2 型 pterinosome. 小囊 속에 소수의 lamel-lae層을 形成한 것을 第 3 型 pterinosome,그리고 小囊 속에 다수의 lamellae를 形成한 것을 第 4 型 pterinosome으로 區別.觀察하였다. 紅色素胞 : 紅色素胞는 長方形 等 不定形의 反射小板으로 채워져 있다. 黃色素胞와 멜라닌色素胞,멜라닌色素胞와 멜라닌色素胞 사이 等에 連接하였다. The ultrastructures of the pigment cells in the Rana temporaria chinensis dorsal skin were observed with electron microscope. The results of the fine structure in the melanophores,xanthophores and iridophores were as follow: Melanophores:Epidermal melanophores were filled with melanin granules which ap-peared as the same electron density. A few melanin granules were observed in a cornified cell. Dermal melanophores contained numerous melanin granules extended up the lateral sides of the xanthophores and iridophores. Xanthophores:Xanthophores were filled with pterinosomes and carotenoid vesicles. Type I pterinosomes had a clear limiting membrane. Type Ⅱ pterinosomes had the inner fibrous structure. Type Ⅲ pterinosomes were charavterized by a few superficial lamellae and Type Ⅳ pterinosomes by multiple concentric lamellae. Iridophores:Iridophores were filled with reflective platelets,each of which is rectangle and shapelessness.

        Synthesis and negative inotropic effects evaluation of 7-substituted-4,5-dihydro-[1,2,4]oxadiazolo[4,3-a]quinolin- 1-ones

        A series of 7-alkoxy-4,5-dihydro-[1,2,4]oxadiazolo[4,3-a]quinolin-1-ones was synthesized and theirnegative inotropic effects were evaluated by measuring theleft atrium stroke volume in isolated rabbit heart preparations. All compounds moderated the cardiac workload bydecreasing heart rate and contractility (inotropic effects). Among them, compound 6 was found to be best potent witha -28.89 ± 1.91 % decrease in the stroke volume at aconcentration of 3 9 10-5 M in our in vitro study.
