Interobserver Variability of Ki-67 Measurement in Breast Cancer

<P><B>Background:</B></P><P>As measurement of Ki-67 proliferation index is an important part of breast cancer diagnostics, we conducted a multicenter study to examine the degree of concordance in Ki-67 counting and to find factors that lead to its variability.</P><P><B>Methods:</B></P><P>Thirty observers from thirty different institutions reviewed Ki-67–stained slides of 20 different breast cancers on whole sections and tissue microarray (TMA) by online system. Ten of the 20 breast cancers had hot spots of Ki-67 expression. Each observer scored Ki-67 in two different ways: direct counting (average vs. hot spot method) and categorical estimation. Intraclass correlation coefficient (ICC) of Ki-67 index was calculated for comparative analysis.</P><P><B>Results:</B></P><P>For direct counting, ICC of TMA was slightly higher than that of whole sections using average method (0.895 vs 0.858). The ICC of tumors with hot spots was lower than that of tumors without (0.736 vs 0.874). In tumors with hot spots, observers took an additional counting from the hot spot; the ICC of whole sections using hot spot method was still lower than that of TMA (0.737 vs 0.895). In categorical estimation, Ki-67 index showed a wide distribution in some cases. Nevertheless, in tumors with hot spots, the range of distribution in Ki-67 categories was decreased with hot spot method and in TMA platform.</P><P><B>Conclusions:</B></P><P>Interobserver variability of Ki-67 index for direct counting and categorical estimation was relatively high. Tumors with hot spots showed greater interobserver variability as opposed to those without, and restricting the measurement area yielded lower interobserver variability.</P>

Green tea was known which regulated adipocyte differentiation metabolism. The mecha nism on the lipid decreased contents of TAG in the plasma. In addition, green tea increased the expression leptin mRNA, PPAR δ mRNA and TGF β. The tea tested was korean powdered green tea. In this experiment, Sprague-Dawley(SD) rats were fed 3 % green tea(powdered) for 3 weeks on the basal diet and obese diet and green tea grouts-fed pork meats, duck meats. The expression of leptin mRNA and PPAR δ mRNA were up-regulated in the green tea-fed groups compared with those of the not green tea-fed groups. There were no significantly difference on the expression of leptin mRNA and PPAR δ mRNA in green tea grouts-fed pork meats, duck meats as compared with the not fed green tea grouts meats. TGF β mRNA, TNF α mRNA and adipsin mRNA were not expressed in the pork meats, duck meats. The expression of TGF β mRNA, TNF α mRNA and adipsin mRNA were observed in the experimental rats but no significantly difference on the contents. Physiologic regulated genes were not expressed in the green tea grout-fed pork meats and duck meats.

포식성이리응애류는 섭식행동이 왕성한 천적군으로 알려져 있으며, 다수의 종들이 상업적으로 개발되고 있다. 이에 우리는 국내 토착 포식성이리응애의 이용도를 탐색하였다. 2012년까지 국내에서 서식이 보고된 포식성이리응애류은 47종을 대상으로 구글 학술검색을 이용하여 논문수를 조사한 결과, 사막이리응애가 3,560개로 가장 많은 수가 검색되었다. 천개 이상의 논문이 검색된 종은 서양이리응애(3,160개), 나팔이리응애(1,590개), 순이리응애(1,510개), 대중이리응애(1,260개), 긴털이리응애(1,190개), 동양이리응애 (1,020개)이었다. 백개 이상의 논문이 검색된 종은 긴꼬리이리응애(314개), 알락이리응애(109개), 엷은줄이리응애(345개), 북방이리응애(143개), 꽃무늬이리응애(200개) 등 5종이었다. 이 중 전세계적으로 상품화가 보고된 종은 사막이리응애, 서양이리응애, 나팔이리응애이다. 국내에 서식이 보고되지 않았으나, 국내외 농업현장에서 가장 많이 사용되는 지중해이리응애는 5,060개가 보고되었다. 긴꼬리이리응애는 국내에서 47종의 식물에서 서식하여 가장 많은 식물 종에서 발견된 포식성이리응애로 농업해충인 점박이응애, 총채벌레류, 가루이류를 포식하며, 농업해충 외에 농작물해충이 아닌 긴털가루응애, 설탕응애, 꽃가루 등의 섭식을 확인하여 앞으로 국내에서 농업해충을 방제하기 위한 천적으로 활용여부에 대한 적극적인 검토가 요구된다.
HPLC를 이용한 carbamate계 농약의 다성분 동시분석

To explore the most sensitive and useful method for simultaneous determination of 8 carbamate pesticides in livestock productions, the mixture of 8 carbonates was determined by HPLC with scanning fluorescence detector. For mobile phase water, acetonitrile and methanol program gradient showed more applicable sensitivity than water and acetonitrile program gradient used ExW 339nm and EmW 445nm for fluorescence detector. On using carbamate columns, the retention time was within 4 to 20 minutes. And this made it possible to separate and detect simultaneously. Therefore we could analyze it exactly and efficiently and reduce time. The preparation process of MSPD method showed that the high recovery rate was more than 88.5% in most of the carbamate pesticides. Therefore we could say that it was an efficient and fast method analyzing out of lots of samples.

Owing to climate change, the global resurgence of vector-borne infectious diseases has emerged as a critical public health issue. Orientia tsutsugamushi is the etiological agent of tsutsugamushi disease (scrub typhus) a mite-borne acute febrile disease occurring in the Asia-Pacific region. We investigated the prevalence of tsutsugamushi disease transmitted by chigger mite vectors living on rodents. Using sticky-type chigger traps for three months during 2016–2018, 1,057 chigger mites were collected (chigger mite index, 1.31) from four locations in the Hwaseong-si area of Gyeonggi-do, Republic of Korea. Five species distributed among three genera were identified. In addition, 94 rodents were captured (collection rate: 7.83%) using Sherman live traps over the course of three months (April, October, and November) during 2016–2017. Three rodent species were captured and identified and the striped field mouse (Apodemus agrarius) was the dominant rodent host species in the surveyed area. A total of 10,469 ectoparasitic chigger mites were recovered from the 94 rodents, from which 13 species distributed among four genera were identified. Of the 5,250 chigger mites examined, Leptotrombidium pallidum was most abundant (n = 2,558), followed by L. orientale, L. scutellare, L. zetum, Euschoengastia koreaensis, L. subintermedium, and Neotrombicula tamiyai. Of the examined chigger mites, no groups recovered from rodent hosts tested positive for O. tsutsugamushi. This study provides fundamental regional information on vector-borne disease data collection in the Hwaseong-si area, Gyeonggi-do, and will further contribute to formulating disease control and prevention strategies.
A survey on the incidence rate of foot diseases in dairy cattle

This study was carried out to investigate the status of hoof diseases and to develope treatment method for cattle with hoof diseases during the period of January to December in 2000. Out of 438 heads, 34 heads(7.8%) had hoof diseases. In respect to season, incidence rate was higher in August to September than that of other seasons. The incidence rates of hoof rot, pododermatitis verrucosae, laminitis, other cases and trauma were 14 cases(41%), 10 cases(29%), 7 cases(21%), 2 cases(6%) and 1 case(3%), respectively. In respect to age and milk production, incidence rate was higher in the cattle with high milk production. Incidence rate of hoof diseases on the hind-limbs was higher than that of the fore-limbs. E coli(8 strains), Clostridium spp(8 strains), Staphylococcus spp(6 strains), Fusobacterium spp(6 strains), and Bacteroides spp(6 strains) were isolated from the hoof lesions. All isolates were sensitive to composite preparation made of mainly formalin with picric acid and phenol. (Treatment against each strains isolated was sensitive to composite preparation made out of the main constitutions of formalin with picric acid and phenol).
