        Evaluation of dietary <i>Hybanthus enneaspermus</i> (Linn F. Muell.) as a growth and haemato-immunological modulator in <i>Labeo rohita</i>

        <P><B>Abstract</B></P> <P>This study evaluated the effects of <I>Hybanthus enneaspermus</I> aqueous extract (HEE) as a feed supplement on growth, immune responses, haematological, and biochemical parameters of <I>Labeo rohita</I> and its susceptibility to <I>Aeromonas hydrophila</I> infection. Diets containing five variable concentrations of HEE (g kg<SUP>−1</SUP>) (0 [basal diet], 1.0 [H1], 2.0 [H2], 3.0 [H3], and 4.0 [H4]) were fed to fish (average weight: 17.2 ± 0.21 g) for 6 weeks. Growth parameters, immune parameters, and haemato-biochemical parameters were examined 6 weeks post-feeding. The results showed that growth parameters, such as final weight (39.47 ± 3.1 g) and specific growth rate (1.92 ± 0.2), were significantly higher in the H3 group than in the control. Immunological parameters such as serum lysozyme (24.73 ± 2.16 U mg<SUP>−1</SUP>), phagocytic activity (19.14± 0.26%), and respiratory burst activity (0.307 ± 0.005) were significantly higher in the H3 group, compared with the control. Among the haematological parameters examined, red blood cells, white blood cells, neutrophils, total serum protein, and albumin levels were significantly higher in the H3 group. However, HEE administration had no significant effect on haematocrit, haemoglobin, monocytes, or eosinophils levels. Blood biochemical parameters, including superoxide dismutase and alkaline phosphatase activities, were higher (<I>P</I> < 0.05) in the H3 group than in the control; however, the opposite result was found for aspartate transaminase, alanine transaminase, glucose, and cholesterol. Supplementation with 3 g kg<SUP>−1</SUP> HEE increased the activities of intestinal digestive enzyme (amylase, protease, and lipase) in fish. Moreover, fish fed diets supplemented with 3 g kg<SUP>−1</SUP> HEE exhibited the highest disease resistance against <I>A. hydrophila</I> infection. These results suggest that dietary administration of HEE (at 3 g kg<SUP>−1</SUP>) has several positive effects on growth, immune and haematological responses in <I>L. rohita</I>.</P> <P><B>Highlights</B></P> <P> <UL> <LI> <I>Labeo rohita</I> diets were supplemented with <I>Hybanthus enneaspermus</I> extract (HEE). </LI> <LI> Dietary administration of 3 g kg<SUP>−1</SUP> HEE improved growth performance in <I>L. rohita.</I> </LI> <LI> Dietary HEE had several positive effects on haematological, biochemical and immune responses of fish. </LI> <LI> HEE at 3 g Kg<SUP>−1</SUP> enhanced the intestinal enzymatic activities in fish. </LI> <LI> Diets with 3 g kg<SUP>−1</SUP> HEE provided the highest protection against pathogen challenge. </LI> </UL> </P>

        Evaluation of the Compatibility of Repair Materials for Concrete Structures

        This study evaluates the compatibility of repair materials for concrete bridge decks. A new compatibility test set-up was designed and tested based on the concrete bridge deck cracking and delamination mechanism theory. The repair materials used in this study include lab formulated inorganic nano-aluminum silicates and commercially available organic two-part epoxy systems. Two different lab test-setups are proposed in this study: a prototype and a full-scale test. The developed test procedures were effective in communicating results in terms of compatibility of material properties, performance and quality. The prototype beams test can successfully serve as a small scale screening test providing insights on materials selection for the full-scale beam tests. The full-scale beams demonstrated the compatibility of the repaired system by providing data on authentic field conditions. Based on the observations it can be concluded that the proposed test setup is effective in examining the concrete bridge deck repair materials performance and selection, and compatibility in terms of mechanical properties and further guarantee the repaired structure safety.

      • The modified Yule-Walker method for multidimensional infinite-variance periodic autoregressive model of order 1

        The time series with periodic behavior, such as the periodic autoregressive (PAR) models belonging to the class of the periodically correlated processes, are present in various real applications. In the literature, such processes were considered in different directions, especially with the Gaussian-distributed noise. However, in most of the applications, the assumption of the finite-variance distribution seems to be too simplified. Thus, one can consider the extensions of the classical PAR model where the non-Gaussian distribution is applied. In particular, the Gaussian distribution can be replaced by the infinite-variance distribution, e.g. by the α-stable distribution. In this paper, we focus on the multidimensional α-stable PAR time series models. For such models, we propose a new estimation method based on the Yule-Walker equa-tions. However, since for the infinite-variance case the covariance does not exist, thus it is replaced by another measure, namely the covariation. In this paper we propose to apply two estimators of the covariation measure. The first one is based on moment representation (moment-based) while the second one—on the spectral measure representation (spectral-based). The validity of the new approaches are verified using the Monte Carlo simulations in different contexts, including the sam-ple size and the index of stability of the noise. Moreover, we compare the moment-based covariation-based method with spectral-based covariation-based technique. Finally, the real data analysis is presented.

      • CYP1A1 Gene Polymorphisms: Modulator of Genetic Damage in Coal-Tar Workers

        Aim: It is well known that polycyclic aromatic hydrocarbons (PAHs) such as benzo (a) pyrene have carcinogenic properties and may cause many types of cancers in human populations. Genetic susceptibility might be due to variation in genes encoding for carcinogen metabolizing enzymes, such as cytochrome P-450 (CYP450). Our study aimed to investigate the effect of genetic polymorphisms of CYP1A1 (m1 and m2) on genetic damage in 115 coal-tar workers exposed to PAHs at their work place. Methods: Genetic polymorphisms of CYP1A1 were determined by the PCR-RFLP method. Comet and buccal micronucleus assays were used to evaluate genetic damage among 115 coal tar workers and 105 control subjects. Results: Both CYP1A1 m1 and CYP1A1 m2 heterozygous and homozygous (wt/mt+mt/mt) variants individually as well as synergistically showed significant association (P<0.05) with genetic damage as measured by tail moment (TM) and buccal micronuclei (BMN) frequencies in control and exposed subjects. Conclusion: In our study we found significant association of CYP1A1 m1 and m2 heterozygous (wt/mt)+homozygous (mt/mt) variants with genetic damage suggesting that these polymorphisms may modulate the effects of PAH exposure in occupational settings.

      • Protective effects of leucine against lipopolysaccharide-induced inflammatory response in Labeo rohita fingerlings

        <P>The present study investigated the protective effects of leucine against lipopolysaccharide (LPS)-induced inflammatory responses in Labeo rohita (rohu) in vivo and in vitro. Primary hepatocytes, isolated from the hepatopancreas, were exposed to different concentrations of LPS for 24 h to induce an inflammatory response, and the protective effects of leucine against LPS-induced inflammation were studied. Finally, we investigated the efficiency of dietary leucine supplementation in attenuating an immune challenge induced by LPS in vivo. Exposure of cells to 10-25 mu g mL(-1) of LPS for 24 h resulted in a significant production of nitric oxide and release of lactate dehydrogenase to the medium, whereas cell viability and protein content were reduced (p < 0.05). LPS exposure (10 mu g mL(-1)) increased mRNA levels of the pro inflammatory cytokines TNF-alpha, IL-1 beta and IL-8 in vitro (p < 0.05). However, pretreatment with leucine prevented the LPS-induced upregulation of TNF-alpha, IL-1 beta and IL-8 mRNAs by downregulating TLR4, MyD88, NF-kappa Bp65, and MAPKp38 mRNA expression. Interestingly, mRNA expression of the anti-inflammatory cytokine, IL-10, which was increased by LPS treatment, was further enhanced (p < 0.05) by leucine pretreatment. The enhanced expression of IL-10 might inhibit the production of other pro inflammatory cytokines. It was found that leucine pretreatment attenuated the excessive activation of LPS-induced TLR4-MyD88 signaling as manifested by lower level of TLR4, MyD88, MAPKp38, NF-kappa Bp65 and increased level of I kappa B-alpha protein in leucine pre-treatment group. In vivo experiments demonstrated that leucine pre-supplementation could protect fish against LPS-induced inflammation through an attenuation of TLR4-MyD88 signaling pathway. Taken together, we propose that leucine pre-supplementation decreases LPS-induced immune damage in rohu by enhancing the expression of IL-10 and by regulating the TLR4-MyD88 signaling pathways.         Dietary emodin affects the growth performance, immune responses, and disease resistance of Labeo rohita against Aeromonas hydrophila

        <P>Healthy Labeo rohita (mean bodyweight of 20.1 g) were divided into four groups before being fed for 60 days on diets supplemented with 0 (control), 20 (E1), 30 (E2), or 40 (E3) mg kg(-1) emodin. Various growth and immune parameters were measured after 15, 30, 45, and 60 days of feeding. Fish fed E2 diet exhibited accelerated (p < 0.05) weight gain after 30-60 days of feeding. The most significant improvements (p < 0.05) in immune parameters, such as lysozyme activity, alternative complement pathway activity, respiratory bursts activity, phagocytic activity, superoxide dismutase activity, and myloperoxidase activity, were observed in the E2-fed group after 30 and 45 days of feeding. However, fish groups fed E2 or E3 diets exhibited significantly lower malondialdehyde, aspartate aminotransferase, and alanine aminotransferase activities than did the control group after 30 and 45 days of feeding. The IgM level was significantly elevated in treatment groups after 30 and 45 days of feeding. Further, fish fed E2 diet for 45 days had the highest (p < 0.005) post-challenge survival rate (83.3 %), followed by fish fed E2 diet for 30 days (75 %). Therefore, dietary feeding of emodin at 30 mg kg(-1) to L. rohita for 30-45 days is optimal to enhance the immunity and disease resistance against A. hydrophila.</P>
