Turbulent flow and mixing performance of a novel six-blade grid disc impeller
Feng Ling Yang,Shenjie Zhou,Cui Xun Zhang 한국화학공학회 2015 Korean Journal of Chemical Engineering Vol.32 No.5
A novel six-blade grid disc impeller (RT-G) was designed and the single-phase turbulent flow and mixing in a baffled stirred tank agitated by this impeller were numerically studied by detached eddy simulation (DES) model. For comparison, a standard Rushton impeller (RT) with the same dimension was also investigated. The numerical results were compared with the reported experimental data and good agreements were obtained. Comparisons of the mean velocity, turbulent kinetic energy, power consumption and mixing time of RT and RT-G were performed. Results show that, for the tank stirred with RT-G, the velocity components can be increased in comparison with RT when the same power is consumed. The increase of the turbulent kinetic energy is about 20-30%. Besides, the mixing time for the tank stirred with RT-G is about 11% shorter than that of RT stirred tank operated at the same condition.

Mixing of two stratified miscible fluids in an eccentric stirred tank agitated by a four pitched-blade turbine was studied by using the detached eddy simulation (DES) model and volume of fluid (VOF) method. The fluids were operated in the transitional and mildly turbulent flow regimes. Interfaces between the two miscible fluids during the mixing processes were captured and mixing times were computed. Effects of the Richardson number and eccentricity on the mixing times were quantificationally analyzed. Results show that the spatial and temporal variations of volume fractions of the fluids can be well captured by the method presented in this study. Mixing time increases with the increase of Richardson number. Effect of eccentricity on mixing time depends on Richardson number and the eccentric agitation scheme is not advisable to use to blend the low-viscosity miscible fluids starting from a stratified state, especially for lower Richardson values.

The capsular polysaccharide (CPS) of pyogenic liver abscessKlebsiella pneumoniaeconsists of repeating units of the trisaccharide (→3)-□-D-Glc-(1→4)-[2,3-(S)-pyruvate]-□-D-GlcA-(1→4)-□-L-Fuc-(1→) and has the unusual feature of extensive pyruvation of glucuronic acid and acetylation of C2-OH or C3-OH of fucose. The present study investigates how CPS activates human monocyte-derived dendritic cells (DCs). Our experimental results show that CPS activates DCs by (1) increasing the expression of CD11c, CD40, CD80, CD83, CD86, and MHC-II (2) increasing the production of TNF-□, IL-1, IL-6, and IL-12p70 (3) increasing DC-elicited allogeneic T-cell proliferation, and (4) increasing the DC-driven Th1 response. In addition, CPS activates DCs through TLR4 and the pyruvation and the acetylation of CPS are important for its cytokine induction activity. Further, our results show that CPS activates TNF-□ and IL-6 secretion through JNK1/2, p38, NF-B,- PKC and ROS pathways in DCs.

Liver cancer is one of leading digestive malignancies with high morbidity and mortality. There is an urgent need for the development of novel therapies for this deadly disease. It has been proven that asparagus polysaccharide, one of the most active derivates from the traditional medicine asparagus, possesses notable antitumor properties. However, little is known about the efficacy of asparagus polysaccharide as an adjuvant for liver cancer chemotherapy. Herein, we reported that asparagus polysaccharide and its embolic agent form, asparagus gum, significantly inhibited liver tumor growth with transcatheter arterial chemoembolization (TACE) therapy in an orthotopic hepatocellular carcinoma (HCC) tumor model, while significantly inhibiting angiogenesis and promoting tumor cell apoptosis. Moreover, asparagine gelatinous possessed immunomodulatory functions and showed little toxicity to the host. These results highlight the chemotherapeutic potential of asparagus polysaccharide and warrant a future focus on development as novel chemotherapeutic agent for liver cancer TACE therapy.

Background: P53 has been reported to be involved with tumorigenesis and has also been implicated as a significant biomarker in oral squamous cell carcinoma(OSCC). However, the diagnostic value of p53 antibodies remains controversial; hence, we comprehensively and quantitatively assessed the potential in the present systematic review. Materials and Methods: A comprehensive search was performed using PubMed and Embase, up to October 31, 2014, without language restriction. Studies were assessed for quality using QUADAS (quality assessment of studies of diagnostic accuracy). The positive likelihood ratio (PLR) and negative likelihood ratio (NLR) were pooled separately and compared with overall accuracy measures using diagnostic odds ratios (DORs) and symmetric summary receiver operating characteristic (SROC) curves. Results: Of 150 studies initially identified, 7 eligible regarding serum p53 antibodies met the inclusion criteria. Some 85.7% (6/7) were of relatively high quality (QUADAS $score{\geq}7$). The summary estimates for quantitative analysis of serum p53 antibody in the diagnosis of squamous cell carcinoma were: PLR 2.06 [95% confidence interval (CI) : 1.35-3.15], NLR 0.85 (95%CI: 0.80-0.90) and DOR 2.47 (95%CI: 1.49-4.12). Conclusions: This meta-analysis suggests that the use of s-p53-antibodies has potential diagnostic value with relatively high sensitivity and specificity for OSCC particularly with serum specimens for discrimination of OSCCs from healthy controls. However, its discrimination power is not perfect because of low sensitivity.
Atrasentan (ABT-627) enhances perfusion and reduces hypoxia in a human tumor xenograft model.
Yang, Kwang Mo,Russell, James,Lupu, Mihaela E,Cho, Hyungjoon,Li, Xiao-Feng,Koutcher, Jason A,Ling, C Clifton Landes Bioscience 2009 Cancer Biology & Therapy Vol.8 No.20

The endothelin-1 antagonist, Atrasentan (ABT-627) was used to modify perfusion in the human tumor xenograft model, HT29, growing in nude mice. Atrasentan produced a significant increase in perfusion, as measured in vivo by Gd-DTPA DCE-MRI. Changes in tumor hypoxia were assessed by comparing the binding of two hypoxia tracers, pimonidazole and EF5 given before and after Atrasentan administration. In vehicle-treated controls, the distribution of EF5 and pimonidazole was very similar. However, Atrasentan treatment was associated with decreased uptake of the second hypoxia tracer (EF5), relative to the first (pimonidazole). Although Atrasentan had no independent effect on the growth of HT29 tumors, Atrasentan combined with 20 Gy radiation led to a modest but significant increase in tumor growth delay compared to radiation alone.

Necroptosis is the major cause of death in alveolar epithelial cells (AECs) during acute lung injury (ALI). Here, we report a previously unrecognized mechanism for necroptosis. We found an accumulation of mitochondrial citrate (citratemt) in lipopolysaccharide (LPS)-treated AECs because of the downregulation of Idh3α and citrate carrier (CIC, also known as Slc25a1). shRNA- or inhibitor–mediated inhibition of Idh3α and Slc25a1 induced citratemt accumulation and necroptosis in vitro. Mice with AEC-specific Idh3α and Slc25a1 deficiency exhibited exacerbated lung injury and AEC necroptosis. Interestingly, the overexpression of Idh3α and Slc25a1 decreased citratemt levels and rescued AECs from necroptosis. Mechanistically, citratemt accumulation induced mitochondrial fission and excessive mitophagy in AECs. Furthermore, citratemt directly interacted with FUN14 domain-containing protein 1 (FUNDC1) and promoted the interaction of FUNDC1 with dynamin-related protein 1 (DRP1), leading to excessive mitophagy-mediated necroptosis and thereby initiating and promoting ALI. Importantly, necroptosis induced by citratemt accumulation was inhibited in FUNDC1-knockout AECs. We show that citratemt accumulation is a novel target for protection against ALI involving necroptosis.
Separable Recursive Gradient Algorithm for Dynamical Systems Based on the Impulse Response Signals
Ling Xu,Feng Ding,Erfu Yang 제어·로봇·시스템학회 2020 International Journal of Control, Automation, and Vol.18 No.12
The identification for process control systems is considered in this paper based on the impulse response signals from the discrete measurements. By taking advantage of impulse signals and through the model parameter decomposition, two dependent identification models are constructed and two identification sub-algorithms are presented based on the nonlinear gradient optimization. In terms of the associated items of the parameters to be estimated between two derived sub-algorithms, a separable recursive gradient parameter estimation method is proposed by designing an interactive and recursive estimation. The performance tests and the comparison experiments are carried out by the simulation examples.

Aim: The purpose of this study was to determine whether susceptibility to oral tongue squamous cell carcinoma (OSCC) is related to polymorphisms in the u-PA gene. Methods: We examined the rs2227564 C/T and rs2227562 G/A single nucleotide polymorphisms (SNPs) in 196 OSCC patients and 201 age- and gender-matched controls via direct sequencing and PCR-RFLP methods. Results: Significant differences were found in allelic and genotypic distributions of the rs2227564 and rs2227562 loci when comparing cases and controls. In addition, logistic analyses indicated that the rs2227564 C/T genotype was related to a 1.52-fold increased risk of developing OSCC (adjusted OR=1.521, 95%CI: 1.144~2.022, P=0.004). Linkage disequilibrium analysis was conducted and no association between the two loci was found (D'=0.031, $r^2$=0.000). Conclusions: Our findings provide evidence that the rs2227564 C/T SNP in the u-PA gene is associated with the development of OSCC.
