<P>Increasing evidence indicates that advanced glycation end products (AGEs) contribute to the pathogenesis of diabetic nephropathy. The aim of this study was to investigate the protective effect of <I>L. japonica</I> extract (LJE) against renal damage in the db/db mouse. LJE (100 or 250 mg/kg per day) was given to diabetic mice for 12 weeks. Body weight, blood glucose levels, glycated hemoglobin (HbA1c) levels, and proteinuria were examined. In <I>in vitro</I> assay of the inhibition of AGE formation, immunohistochemical analysis of podocyte loss and AGE accumulations were performed. In 20-week-old db/db mice, severe hyperglycemia developed, and proteinuria was significantly increased. Diabetes induced markedly morphological alterations to the renal glomerular cells. AGE accumulations and podocyte loss were detected in renal glomeruli. LJE treatment significantly reduced proteinuria and AGE accumulations in diabetic mice. Moreover, the loss of nephrin, an important slit diaphragm component in the kidneys, was restored by LJE treatment. Our studies suggest that LJE might be beneficial for the treatment of diabetic nephropathy. The ability of LJE to attenuate proteinuria and podocyte dysfunction may be mediated by the inhibition of AGE accumulation in the context of diabetic nephropathy in db/db mice.</P>

<P>Podocyte injury contributes to renal damage and, eventually, to the occurrence of proteinuria in diabetic nephropathy. The aim of the present study was to investigate the effect of an ethanol extract from Rhizoma <I>Polygonum cuspidatum</I> (<I>P. cuspidatum</I>) on proteinuria and podocyte injury, and elucidate the underlying mechanism for streptozotocin (STZ)-induced diabetic nephropathy. The protective effects of <I>P. cuspidatum</I> extract (PCE) on renal podocytes in STZ-induced diabetic rats were also investigated. PCE (100 or 350 mg/kg/day) was administered to STZ-induced diabetic rats for 16 weeks, and blood glucose levels, body weight and proteinuria were measured. A double labeling technique with the terminal deoxynucleotidyl transferase dUTP nick end labeling assay was performed and synaptopodin expression was observed. In addition, cleaved caspase-3, methylglyoxal (MGO) and 8-hydroxydeoxyguanosine (8-OHdG) expression levels were measured. STZ-induced diabetic rats developed hyperglycemia and proteinuria. Increased apoptosis of the podocytes and increased cleaved caspase-3, MGO and 8-OHdG expression levels, as well as decreased synaptopodin expression were detected in the glomeruli of STZ-induced diabetic rats. However, treatment with PCE for 16 weeks restored protein levels to normal, and reduced podocyte loss and apoptosis. Levels of caspase-3 and MGO expression, as well as oxidative stress were ameliorated by PCE treatment. In addition, emodin, a biologically active ingredient of PCE, exerted an MGO scavenging effect and inhibited MGO-derived advanced glycation end-product formation. These findings indicate that PCE may be administered to prevent proteinuria and podocyte loss in STZ-induced diabetic rats partly by inhibiting podocyte apoptosis and cleaved caspase-3 expression, and by restoring the balance of oxidative stress and MGO expression.</P>
초등 예비교사의 핵심역량 진단도구 개발 및 타당화: 요인 분석 및 Rasch 모형의 적용

이 연구의 목적은 A교육대학교의 교육이념과 인재상에 기반 하여 초등 예비교사를 위한 핵심역량 진단도구를 개발하고 타당화 하는 데 있다. 이를 위해 관련 선행연구 및 전문가 검토를 거쳐 해당 대학의 인재상과 핵심역량의 개념을 조작적으로 정의하여 평가틀을 구안하고, 이를 바탕으로 예비검사 문항(총 240개)을 개발하였다. 예비검사를 통하여 본 검사는 총 7개 하위척도, 142개 문항으로 구성된 자기보고식 검사로 구성되었다. 본 연구에서는 A교육대학교 재학생 692명(32.9% 남학생)을 대상으로 확인적 요인분석 및 Rasch 측정모형 분석을 활용하여 본 검사 문항의 질을 점검하였다. 먼저 핵심역량별 요인구조의 타당성을 검토하고자 확인적 요인분석(Confirmatory Factor Analysis: higher-order CFA)을 실시하였고, 문항 수준에서의 분석을 위해 문항반응이론(Item Response Theory: IRT)에 근거한 1-모수 Rasch 모형을 적용하였다. 분석 결과, 하위척도별 신뢰도는 양호한 수준이었고, 핵심역량 CFA 모형의 적합도는 수용 가능한 수준으로 보고되었다. 또한 Rasch 모형 적합도, 문항의 난이도 및 응답 범주의 수 역시 적절한 것으로 판단되었다. 마지막으로 본 연구의 결과를 토대로 핵심역량 진단도구가 초등 예비교사 양성과정에서 갖는 의의와 시사점에 대해 논의하였다. The purpose of this paper was to develop the core competency scale for primary pre-service teachers based on the concept of talent and key competencies constructed by A university of Education, and also to describe its psychometric evaluation. The survey was conducted with a total of 692 students attending to A university of education. Two complementary analytical approaches, i.e., factor analysis and Rasch analysis, were applied to investigate the psychometric properties of the scale. A Confirmatory factor analysis supported the factor models designed by their conceptual factor structure. A Rasch analysis provided further evidence of the psychometric quality of the scale in terms of item properties, the rating scale effectiveness, and item fit statistics. The final version of the scale contains 142 items in seven sub-scales assessing primary pre-service teachers’ core competencies. Finally, the information provided by this scale can contribute to a better understanding of primary pre-service teachers’ core competencies and, therefore, inform educators and researchers about how to enhance per-service teachers’ core competency.
Effect of KIOM-79 on Diabetes-Induced Myocardial Fibrosis in Zucker Diabetic Fatty Rats

<P>KIOM-79, a herbal mixture of parched Puerariae radix, gingered Magnoliae cortex, Glycyrrhizae radix, and Euphorbiae radix, has a strong inhibitory effect on advanced glycation end products (AGEs) formation. We investigated the beneficial effects of KIOM-79 on cardiac fibrosis in Zucker diabetic fatty (ZDF) rats. KIOM-79 (50 or 500 mg/kg/day) was orally administered for 13 weeks. AGEs formation and collagen expression in the myocardium were assessed by immunohistochemistry. The expression levels of the receptor for AGEs (RAGE), transforming growth factor-<I><I>β</I></I>1 (TGF-<I><I>β</I></I>1), collagen IV, fibronectin, urotensin II, and urotensin II receptor were examined in the myocardial tissue of ZDF rats. KIOM-79 treatment at 500 mg/kg inhibited the accumulation of AGEs, reduced RAGE mRNA and protein expression, and reduced the upregulation of cardiac fibrogenic factors, such as fibronectin and collagen IV, in heart of ZDF rats. Additionally, KIOM-79 ameliorated urotensin II/receptor gene expression in the cardiac tissue of ZDF rats. Our findings indicate that KIOM-79 diminishes cardiac fibrosis in ZDF rats by preventing AGEs accumulation and RAGE overexpression and by modulating the cardiac urotensin II/receptor pathway, which decreases the amount of profibrotic factors, such as TGF-<I><I>β</I></I>1, fibronectin, and collagen in cardiac tissue.</P>
지체부자유 아동의 거주성 향상을 위한 기초적 연구 : 주거내부의 부분개조에 대하여

The purpose of this study was to investigate sociodemographic characteristics related to the residential space of handicapped children, to investigate problems in their housing, and to propose solutions for planning their housing. Docummentary research and questionnaire survey methods were used. The questionnaire consisted of sociodemographic characteristics and residential space usage of handicapped children. Data were analyzed by frequency and percentage. The results of this study were that the types of handicaps could be classified as Quadriplegia, Hemiplegia, Monoplegia and Diplegia. The type with the highest frequency was Quadriplegia. The handicapped children use residential space with other family members. The space problems which were problematic for them included entrance doors, room size, toilet facilities, and floor materials. Solutions to these problems were presented in terms of residential space shared with other family members. The implementation of these solutions will reflect better housing for handicapped children.
The Role of High-Mobility Group Box-1 Protein in the Development of Diabetic Nephropathy

<P>Abstract</P><P><I>Background/Aims:</I> The purpose of the experiment reported here was to assess the involvement of high-mobility group box-1 (HMGB1), receptor for advanced glycation end products (RAGE) and nuclear factor (NF)-κB signaling pathway in the development of rat diabetic nephropathy. <I>Methods:</I> Diabetes was induced by intraperitoneal streptozotocin injection in 7-week-old male rats. At 20 weeks of age, renal expression of HMGB1 was detected by immunohistochemistry. The expression of RAGE and NF-κB activity was studied by Western blot and electrophoretic mobility shift assay in renal tissues of normoglycemic and diabetic rats, respectively. <I>Results:</I> HMGB1 was highly expressed in both the cytoplasmic and nuclear patterns in diabetic renal glomerular cells and tubular epithelial cells, although in normal rats, HMGB1 was expressed only in the cell nuclei. The expression of RAGE, a potential receptor for HMGB1, and NF-κB activity were also greater in diabetic than in normal rats. Moreover, diabetes increased the binding of NF-κB to the RAGE promoter. <I>Conclusion:</I> These findings suggest that hyperglycemia-induced HMGB1 release may induce the renal injury in diabetic rats, and that the pathogenic role of HMGB1 might be dependent on RAGE and through activation of NF-κB.</P><P>Copyright © 2011 S.
