Hollow Spherical Curcumin Nanomicelles CUR@PLA@PF127: A New Trial on Breast Cancer Cells

        Fatma S. El-Banna,Nemany A. N. Hanafy,Magdy E. Mahfouz,Maged El Kemary 한국생물공학회 2023 Biotechnology and Bioprocess Engineering Vol.28 No.5

Curcumin (CUR) nanomicelles were developed by solvent-evaporation method using Poly (lactic acid) (PLA) for the first time with Pluronic F127 (PF127) to produce CUR@PLA@PF127 nanomicelles. The X-ray diffraction (XRD) spectra confirmed the encapsulation of CUR into the produced nanomicelles (CUR@PLA@PF127). Fourier transform infrared spectroscopy (FTIR) analysis showed the interactions of CUR with PLA and PF127. Scanning electron microscope (SEM) and transmission electron microscope (TEM) analysis showed the formation of spherical shape of CUR@PLA@PF127 with a diameter around 130-160 nm. To investigate the anticancer effect of the encapsulated CUR, the activity of CUR@PLA@PF127 nanomicelles, free CUR and blank nanomicelles (PLA@PF127) were assayed against two cell lines in-vitro, human breast adenocarcinoma cells (MCF-7) and human breast skin fibroblast cells. The cytotoxicity assay of CUR@PLA@PF127 nanomicelles, revealed an IC50 value of 159.7 ± 1.36 μg/mL for MCF-7 cells which is much lower than that observed for free CUR. This result suggests that CUR@PLA@PF127 nanomicelles increased CUR potency due to improving its water solubility, resulting in longer release profile, and increasing antiproliferative effect on breast cancer cells (MCF-7). We anticipate that CUR@PLA@PF127 in drug delivery, improving drugs design and that the developed nanomicelles will be regarded one of the most promising nanocarriers for the treatment of breast cancer, and opening the door to future applications in cancer therapies.

        A Self-Consistent Semi-Analytical Model for AlGaAs/InGaAs PMHEMTs

Abdel Aziz, M.,El-Banna, M.,El-Sayed, M. The Institute of Electronics and Information Engin 2002 Journal of semiconductor technology and science Vol.2 No.1

A semi-analytical model based on exact numerical analysis of the 2DEG channel in pseudo-morphic HEMT (PMHEMT) is presented. The exactness of the model stems from solving both Schrodinger's wave equation and Poisson's equation simultaneously and self-consistently. The analytical modeling of the device terminal characteristics in relation to the charge control model has allowed a best fit with the geometrical and structural parameters of the device. The numerically obtained data for the charge control of the channel are best fitted to analytical expressions which render the problem analytical. The obtained good agreement between experimental and modeled current/voltage characteristics and small signal parameters has confirmed the validity of the model over a wide range of biasing voltages. The model has been used to compare both the performance and characteristics of a PMHEMT with a competetive HEMT. The comparison between the two devices has been made in terms of 2DEG density, transfer characteristics, transconductance, gate capacitance and unity current gain cut-off frequency. The results show that PMHEMT outperforms the conventional HEMT in all considered parameters.

      • Bithionol residue analysis in animal-derived food products by an effective and rugged extraction method coupled with liquid chromatography–tandem mass spectrometry

        Zheng, Weijia,Park, Jin-A,Abd El-Aty, A.M.,Kim, Seong-Kwan,Cho, Sang-Hyun,Choi, Jeong-Min,Yi, Hee,Cho, Soo-Min,El-Banna, H.A.,Shim, Jae-Han,Chang, Byung-Joon,Wang, Jing,Kim, Jin-Suk,Shin, Ho-Chul Elsevier 2017 Journal of chromatography. B, Analytical technolog Vol.1064 No.-

<P><B>Abstract</B></P> <P>Herein, we developed a simple analytical procedure for the quantitation of bithionol residues in animal-derived food products such as porcine muscle, eggs, milk, eel, flatfish, and shrimp using a modified quick, easy, cheap, effective, rugged, and safe (QuEChERS) extraction method coupled with liquid chromatography–electrospray ionization tandem mass spectrometry (LC-ESI+/MS-MS). Samples were extracted with 0.1% solution of formic acid in acetonitrile and the extract was purified using a C18 sorbent. Separation was performed on a Waters XBridge™ C18 reversed-phase analytical column using 0.1% solution of formic acid/acetonitrile as the mobile phase. Six-point matrix-matched calibration indicated good linearity, with the calculated coefficients of determination (<I>R</I> 2) being≥0.9813. Intra- and inter-day recoveries (determined at spiking levels equivalent to 1×and 2×the limit of quantitation (0.25μg/kg)) ranged between 80.0 and 94.0%, with the corresponding relative standard deviations (RSDs) being≤8.2%. The developed experimental protocol was applied to different samples purchased from local markets in Seoul, which were tested negative for bithionol residues. In conclusion, the proposed method proved to be versatile and precise, being ideally suited for the routine detection of bithionol residues in animal-derived food products with various protein and fat contents.</P> <P><B>Highlights</B></P> <P> <UL> <LI> Bithionol residues were quantified in porcine muscle, whole milk, eggs, eel, flatfish, and shrimp. </LI> <LI> Samples were extracted using a modified EN-QuEChERS method followed by LC–MS/MS analysis. </LI> <LI> Satisfactory extraction efficiency was obtained using 0.1% formic acid in acetonitrile as an extractant. </LI> <LI> None of the market samples tested positive for the target analyte. </LI> <LI> The developed method can help establish the MRL of bithionol, which is yet to be set by regulatory authorities. </LI> </UL> </P>

Comparison of preservation enrichment media for long storage duration of Campylobacter jejuni

        AL-Fawares O'la,Al-Khresieh Rozan O.,Aburayyan Walid,Seder Nesrin,Al-Daghistani Hala I,El-Banna Nasser,Abu-Taleb Emran M. 한국미생물학회 2023 미생물학회지 Vol.59 No.3

Research on Campylobacter jejuni has been greatly impaired due to poor culturing and storage techniques. This study aims to evaluate different preservation methods for C. jejuni for long-term periods of preservation. Three different preservation methods (i, ii, and iii) were tested for their potential for long-term storage of C. jejuni which could be recommended as a simple and reliable method. In the first preservation method (i), 15% glycerol was added to 85% of the bacterial suspension of 4 McFarland standards (McF) and yeast extract. In the second method (ii) the bacterial suspension was supplemented with defibrinated lacked horse blood along with skim milk. Skim milk and 50% glycerol were added to the bacterial suspension in method (iii). Results indicated that method (ii) maintained the highest preservation rate of C. jejuni and the bacteria was capable to grow even after 12 months of storage at -80°C. Method (i) was less efficient than method (ii) where isolates eventually lost their ability to grow after 8 months. Method (iii) showed the least successful preservation rate as many isolates started to die after six months of preservation. This study emphasizes the importance of using defibrinated lacked horse blood along with skim milk during the preservation of bacterial isolates of C. jejuni.
