        Association and functional relevance of E237G, a polymorphism of the high-affinity immunoglobulin E-receptor β chain gene, to airway hyper-responsiveness

        <P>Summary</P><P>Background</P><P>The hyper-sensitivity reaction of IgE, with its high-affinity receptors (FcϵRI), is central to the phenomenon of atopic diseases.</P><P>Objective</P><P>To evaluate the genetic effects of non-synonymous single-nucleotide polymorphisms (SNPs) of FcϵRI on intermediate phenotypes of asthma, i.e. atopy and airway hyper-responsiveness (AHR), in the Korean general population.</P><P>Subjects and methods</P><P>Atopy and AHR were evaluated in a cohort of 2055 subjects, aged 10–18 years, using skin prick tests (SPTs) for common aeroallergens and total serum IgE and methacholine bronchial provocation tests. All FcϵRI-α, FcϵRI-β, and FcϵRI-γ gene exons of 24 healthy subjects were sequenced to locate informative non-synonymous SNPs (minor allele frequency >2%). Informative SNPs were then scored, using the high-throughput single base extension method. Relative risk (RR) was determined by multiple logistic regression analysis, after adjusting for confounding factors. The functional relevance of non-synonymous SNPs was analysed using the sorting intolerant from tolerant (SIFT) program.</P><P>Results</P><P>The SNP search found only one informative non-synonymous SNP in FcϵRI-β: E237G (minor allele frequency=0.21). The positive rate of AHR was lower among subjects with the 237<SUP>*</SUP>E allele than among those with 237<SUP>*</SUP>G [RR (95% confidence interval)=0.41 (0.19–0.89); <I>P</I>=0.01]. However, the E237G substitution was not associated with either a positive SPT response or total serum IgE levels. Sequence evolution analysis predicted that the E237G variation is an intolerant amino acid substitution, with functional importance.</P><P>Conclusion</P><P>In the Korean general population, AHR is significantly associated with the E237G polymorphism of FcϵRI-β, which results in an intolerant amino acid substitution.</P>

        소 모색관련 유전자 MC1R 의 RCR - RFLP Marker 를 이용한 한우육 판별

        The melanocortin 1 receptor(MClR) plays a central role in regulation of eumelanin(black/brown) and phaeomelanin(red/yellow) pigment synthesis within the mammalian melanocytes. Mutations within the gene encoding MC1R have been shown to explain coat colour variations within several species including cattle. This study was performed to develop the identification technique of Hanwoo meat using MC1R gene associated with the coat colors of cattle. Alleles of the MC1R locus were detected by PCR-RFLP analysis and genotype frequency and DNA sequences of MC1R gene were compared among cattle breeds. Genomic DNA was extracted from meat or blood samples of five breeds including Hanwoo(n=200), Holstein(n=100), Angus(n=20), Hereford(n=20) and Charolais(n=20). The MC1R gene was used to amplify 739bp and 173bp of the bovine E-locus corresponding to positions 228-966 and 318-490, respectively, using the two specific primers. The amplified products were digested with Bse118 I or Msp I and Aci I enzymes, and DNA fragments were separated by gel electrophoresis for RFLP genotype analysis. Six genotypes, E^D/E^D E^D/E^+, E^D/e, E^+/E^+,E^+/e and e/e, controlled by three alleles E^D, E^+ and e were observed in MC1 locus. When the amplified DNA product(739bp) was digested with Bse118 I enzyme, Hanwoo meat showed a single band of 739bp, whereas two fragments of 531bp and 208bp were detected in Holstein meat and Angus breed, respectively. Also, in the RFLP patterns using Msp I enzyme, Hanwoo meat produced two fragments of 535bp and 174bp, while three fragments of 328bp, 207bp and 174bp were observed in Holstein meat and Angus breeds, respectively. Therefore, breed-specific RFLP markers showing distinct differences between these breeds were found by PCR-RFLP analysis. When the amplified DNA product(173bp) was digested with Aci I enzyme to classify subtype of E allele, the E^D allele produced three fragments of 97, 68 and 8bp, while the E^+ and d alleles produced two fragments of 173 and 8bp according to the Aci I recognition sequence. Among the six genotypes, two genotypes of E^+/e and e/e were observed in Hanwoo and their frequencies were 0.07 and 0.93, respectively. However, the E^D/ED and E^D/e genotypes were present in Holstein and E^D/E^D, E^D/E^+ and E^D/e genotypes in Angus breeds. Therefore, the E^+/e and e/e genotypes observed in Hanwoo and E^D/E^D, E^D/E^+ and E^D/e genotypes detected only in Holstein and Angus breeds may be useful as breed-specific DNA markers for distinguishing between Hanwoo meat and Holstein and Angus meats. When comparing MC1R sequences among Hanwoo, Holstein and Angus, a Gly → Val amino acid change due to a single base(G) deletion at colon 104 was found in Hanwoo. Consequently, breed specific RFLP genotypes of MC1R gene related to bovine coat colors could be used as DNA markers for identification of Hanwoo meat from Holstein and Angus meats.

        Proposed mechanism in the change of cellular composition in the outer medullary collecting duct during potassium homeostasis.

        <P>Potassium depletion (K?-D) induces hypertrophy and hyperplasia of collecting duct cells, and potassium repletion (K?-R) induces regression of these changes. The purpose of this study was to examine the time courses of the changes in cellular composition, the origin of intercalated cells (ICs) and the mechanism responsible for these changes. SD rats received K?-depleted diets for 1, 7, or 14 days. After K?-D for 14 days some of the rats received normal diets for 1, 3, 5, or 7 days. In the inner stripe of the outer medulla, K?-D increased significantly the number and proportion of H?-ATPase-positive ICs, but decreased the proportion of H?-ATPase-negative principal cells (PCs). However, proliferation was limited to H?-ATPase-negative PCs. During K?-R, the cellular composition was recovered to control level. Apoptosis increased during K?-R and exclusively limited in H?-ATPase-negative PCs. Double immunolabeling with antibodies to PC and IC markers identified both cells negative or positive for all markers during both K?-D and K?-R. Electron microscopic observation showed that ultrastructure of AE1-positive some cells were similar to AE1-negative some cells during K?-R. LC3 protein expression increased significantly and autophagic vacuoles appeared particularly in PCs on days 14 of K?-D and in ICs on days 3 of K?-R. These results suggest that PCs and ICs may interconvert in response to changes in dietary K+ availability and that autophagic pathways may be involved in the interconversion.</P>

      • Aerosol delivery of eukaryotic translation initiation factor 4E-binding protein 1 effectively suppresses lung tumorigenesis in K-ras<sup>LA1</sup> mice

        Conventional radiotherapy or chemotherapy for the long-term survival of patients with lung cancer is still difficult for treatment in metastatic and advanced tumors. Therefore, the safe and effective approaches to the treatment of lung cancer are needed. In this study, the effect of delivered eukaryotic translation initiation factor 4E (eIF4E)-binding protein 1 (4E-BP1) on lung cancer progression was evaluated. Recombinant adeno-associated virus (rAAV)-M3/4E-BP1 was delivered into 6-week-old K-ras<SUP>LA1</SUP> lung cancer model mice through a nose-only inhalation system twice a week for 4 weeks. Long-term repeated delivery of 4E-BP1 effectively reduced tumor progression in the lungs of K-ras<SUP>LA1</SUP> mice. Reduction of eIF4E by overexpression of 4E-BP1 resulted in suppression of cap-dependent protein expression of basic fibroblast growth factor (bFGF or FGF-2) and vascular endothelial growth factor (VEGF). In addition, delivered 4E-BP1 inhibited the proliferation of lung cancer cells in K-ras<SUP>LA1</SUP> mice model. Our results suggest that long-term repeated viral delivery of 4E-BP1 may provide a useful tool for designing lung cancer treatment.

        The main purpose of this study was to characterize the air pollutants emission factors in electric power plant (EPP) using fossil fuels. The electric power plant is a major air pollution source, thus knowing the emission characteristics of electric power plant is very important to develop a control strategy. The major air pollutants of concern from EPP stacks are particulate matter (PM), sulfur oxides (SOx), nitrogen oxides (NOx), carbon monoxide (CO) and heavy metals. Throughout the study. the following results arc estimated: - PM : 8.671E-05∼8.724E+01 PM emission (kg) per fuel burned (ton) - SOx: 4.749E-04∼7.877E+01 SOx emission (kg) per fuel burned (ton) - NOx : 1.578E-02∼9.857E+00 NOx emission (kg) per fuel burned (ton) - CO : 3.800E-04∼1.291E+00 CO emission (kg) per fuel burned (ton) - Hg : 1.220E+01∼3.108E+02 Hg emission (mg) per fuel burned (ton) From the statistical analysis by Wilcoxon signed ranks test between the emission factors of ours and U.S. EPA's. we can yielded that: p > 0.05.

      • Deep Level Transient Spectroscopy법에 의한 InAs/InP계 양자점구조의 에너지준위 연구

        MOCVD (metal organic chemical vapor deposition)장치로 자발형성시킨 InAs/Inp계 양자점구조의 에너지 준위를 DLTS(deep level transient spectroscopy) 측정법을 이용하여 연구하였다. InP 기판의 온도 530℃에서 InAs 양자점을 형성한 다음 InP 덮개층 성장시 성장온도에 따른 InAs 양자점의 물성변화를 조사하기 위해 성장온도를 530℃에 고정시킨 경우와 먼저 530℃ 성장하고 다시 620℃의 온도에서 성장시키는 2단계 성장기법으로 성장시킨 두 종류의 시료를 준비하였다. 이들 시료에 대한 DLTS 측정결과 활성화에너지와 운반자방출단면적은 덮개층 성장시 기판온도를 530℃에 고정시킨 시료에서는 E_c-0.56 eV 와 4×10^(-15)㎠으로 각각 나타났으나, 덮개층 성장온도를 530℃ 와 620℃에서 성장시킨 시료에서는 E_c-0.74eV와 2.46×10^(-12)㎠로 더 크게 나타났다. 이는 고온에서 덮개층 성장시 InAs 양자점과 InP 덮개층사이의 inter-diffusion효과로 고려되었다. 한편, E_c-0.56 eV에너지 준위의 운반자 포획장벽높이는 0.18 eV로 나타났다. We studied on the energy levels of self-assembled InAs/InP quantum dot structures which was grown by metal organic chemical vapor deposition by using deep level transient spectroscopy(DLTS) method. The InAs quantum dots were grown on InP substrate at 530℃. During the growth of InP capping layer, the temperature of the first sample was fixed at 530℃ and in the other case, two step growth at temperatures of 530℃ and 620℃ were used. In the first sample, we could get one electron trap level with the activation energy of E_c-0.56 eV and the emission cross section of 4×10^(-15)㎠. This electron trap level was considered as the quantum dot level. On the other hand, in the second sample, the activation energy and the emission cross section were obtained E_c-0.74 eV and 2.46×10^(-12)㎠, respectively. This result considered as the inter-diffusion effect of InAs quantum dot during the growth at high temperature. In addition, the carrier capture barrier height of E_c-0.56 eV electron trap level is 0.18 eV.

      • Evaluation of indentation tensile properties of Ti alloys by considering plastic constraint effect

        The uniaxial tensile properties of metallic materials can be determined by representing the indentation stress and strain by the load/depth parameters from instrumented indentation tests using a spherical indenter. Here we propose a modified algorithm for evaluating the tensile properties of Ti alloys with low E/σ<SUB>y</SUB> values by taking into account the plastic constraint effect in low-strain regions. The ratio of elastic modulus to yield strength, E/σ<SUB>y</SUB>, can be interpreted as a measure of the amount of deformation plastically accommodated during indentation. For low E/σ<SUB>y</SUB> materials, deformation is elastic-dominated, so less constraint occurs than in high E/σ<SUB>y</SUB> materials. Finite element methods and spherical indentation tests were used to explore the constraint effect for a variety of materials characterized by different E/σ<SUB>y</SUB>. The constraint factors were plotted as (E/σ<SUB>y</SUB>)(a/R) based on expanding cavity model. The modified approach was experimentally verified by comparing tensile properties from uniaxial tensile test and instrumented indentation tests.

        Pulse Shape Discrimination of Nuclear Recoil and Electron Recoil Events With a NaI(Tl) Crystal for Dark Matter Search

        <P>In order to investigate discrimination between nuclear recoil and electron recoil events for the KIMS-NaI dark matter search experiment, we measured the pulse shapes produced by neutrons and gamma rays in a NaI(Tl) crystal. Relatively good pulse shape discrimination (PSD) power due to high light output of recently developed crystals makes it possible to test whether the annual modulation signal observed by the DAMA/LIBRA experiment is caused by nuclear recoil events. We applied the PSD to underground data taken with a 9.15 kg low-background and high-light-output NaI(Tl) crystal for 134 days. Good agreement between underground data and electron recoil events was observed.</P>

      • Optical subthreshold current method for extracting the interface states in MOS systems

        Optical subthreshold current method (OSCM) is proposed for characterizing the interface states in MOS systems using the current-voltage characteristics under a photonic excitation. An optical source with a subbandgap (E<SUB>ph</SUB>>E<SUB>g</SUB>) photonic energy (E<SUB>ph</SUB>=0.943 eV, P<SUB>opt</SUB>=+5 dBm), which is less than the silicon bandgap (E<SUB>g</SUB>=1.12;eV), is employed for the optical subthreshold current characterization of interface states in the photoresponsive energy band. We applied the OSCM method under a subbandgap photonic excitation to MOS systems with a poly-Si gate and verified a U-shaped distribution of interface trap density D<SUB>it</SUB>=10<SUP>10</SUP>∼10<SUP>12</SUP> eV<SUP>-1</SUP>cm<SUP>-2</SUP> for n- and p-type MOSFETs with W/L=30 μm/1.2 μm.

        Comparison of bioaccumulation and elimination of Escherichia coli and male-specific bacteriophages by ascidians and bivalves

        <P>Levels of Escherichia coli and male-specific bacteriophages (MSBs) were determined in the filter feeders obtained from retail markets, commercial farms, and wild beds in Korea. The accumulation and elimination of E. coli and MSBs were compared between ascidians and bivalves (oysters and mussels) during relaying and depuration. E. coli concentrations in ascidians from retail markets ranged between < 20 and 460 most probable number/100 g while MSBs were not detected. E. coli levels in bivalves from commercial farms and wild beds were not significantly different but bacterial levels in ascidians were consistently lower. Ascidians exhibited much lower ability than bivalves to accumulate E. coli and MSBs during relaying in a polluted coastal area. This study also shows that an equilibrium was developed between levels of microbes in water and ascidians and shellfish during relaying. E. coli and MSBs in ascidians decreased quickly during depuration in a clean seawater tank. However, after 1 day, E. coli in bivalves decreased by only 1.1-1.6 logs, and the elimination of MSBs was negligible. Therefore, depuration is an effective means to reduce the health risk of contaminated ascidians.</P>
