        A Novel Accessory Molecule Trim59 Involved in Cytotoxicity of BCG-Activated Macrophages

        BCG-activated macrophages (BAM) could kill the tumor cells through cell-cell contact. In this process membrane proteins play an important role. However, up to date, few membrane proteins were revealed. In this study, we se-lected a surface molecule named Trim59, which was spe-cifically expressed on BAM membrane (compared with the negative control). We cloned and prokaryoticly expressed the extracellular domain of Trim59, purified the recombinant protein and generated polyclonal antibodies. Immunohistochemistry showed that Trim59 abundantly expressed in spleen, stomach and ovary; intermediately expressed in brain, lung, kidney, muscle and intestine; but not in thymus, liver, heart, uterus. Using the antibodies to block Trim59 on BAM significantly reduced BAM cyto-toxicity against MCA207 cells. This demonstrated that Trim59 serves as an indispensable molecule in maintaining BAM activity. Overexpression of Trim59 in Raw264.7 cell line failed to lyse target MCA207 cells, which potentiated Trim59 per se could not enhance macrophage cytotoxicity; on another hand, overexpression of Trim59 enhance the pinocytosis and Phagocytosis activity of Raw-264.7, which imply Trim59 might mediate the cell-molecule interaction. Our results indicate Trim59 might be an essential accessory molecule in mediating BAM tumoricidal functions; and Trim59 is a phagocytosis-correlated molecule.

        Carbon Nanotubes Modified by Ionic Liquid and Its Effect on Properties of Silicon Rubber Composites

        In this paper, multi-walled carbon nanotubes (MWCNTs) were modified using ionic liquids (ILs), and silicone rubber/carbon nanotube composites were prepared. The surface of carbon nanotubes is coated with a layer of ionic liquid by non-covalent bond modification, and the van der Waals force between the carbon nanotubes is reduced to achieve the purpose of improving their dispersibility. The electrical conductivity of silicone rubber/carbon nanotube composites prepared by modified carbon nanotubes is improved. The dispersion of modified carbon nanotubes in silicone rubber matrix and the interface interaction between carbon nanotubes and silicone rubber are analyzed by rubber process analyzer and scanning electron microscopy (SEM) tests. The effects of modified carbon nanotubes on electrical conductivity, mechanical properties and electromagnetic shielding properties of silicone rubber composites were studied. The results show that the dispersion of carbon nanotubes in silicone rubber matrix is improved effectively through the modification of ionic liquid, the conductive property of the composite is improved, the electromagnetic wave reflection of the composite is increased, and the electromagnetic shielding efficiency of the composite is improved, and the shielding efficiency is up to 26 dB.

      • Surface-Enhanced Raman Scattering (SERS) Active Gold Nanoparticles Decorated on a Porous Polymer Filter

        <P>In this work, we designed a process to assemble gold nanoparticles onto a three-dimensional (3D) polymer surface, which can then be monitored using surface-enhanced Raman scattering (SERS). This work is the first demonstration of the assembly of gold nanoparticles on a filter film and in situ measurement with Raman spectroscopy. Herein, a polyhexamethylene adipamide (Nylon66) film embedded in the organic filter film was used as a template to fabricate a tunable SERS-active substrate. A 'hotspot''-rich gold-nanoparticle-decorated polymer substrate for SERS was prepared; this substrate exhibited high sensitivity in trace detection of targets. The study was conducted using 4-mercaptobenzoic acid as a probe molecule with the aim of comparing the scattering efficiency and the homogeneity of the Raman signal on selected substrates. In addition, we used the gold-decorated polymer film to detect a biotin-avidin complex. The most powerful advantage of the proposed microanalytical device is the in situ SERS application. The 3D nanoporous structures described in this work hold strong potential for use in various applications such as environmental monitoring and biomolecule detection.</P>
