Immunomodulatory and Therapeutic Potential of Enrofloxacin in Bovine Sub Clinical Mastitis

Mukherjee, Reena,Dash, P.K. Asian Australasian Association of Animal Productio 2003 Animal Bioscience Vol.16 No.6

Immunomodulatory and therapeutic potential of Enrofloxacin was studied in bovine sub clinical mastitis (SCM). The therapeutic efficacy was adjudged by Somatic cell count and Total bacterial count of the milk, whereas, the immuno modulatory potential of the drug was assessed by measuring myeloperoxidase (MPO) and acid phosphates (ACP) enzyme level in the milk leukocytes. Forty-five cows were divided into three equal groups. Gr I consisting 15 cows served as healthy control, whereas, 30 cows (SCM), Gr II and Gr III, selected on the basis of California Mastitis Test (CMT) positive reaction. Gr II cows received 150 mg of Enrofloxacin, once a day for three days and Gr.III received sterile 5 ml PBS (pH 7.4) for 7days, both the treatment were given by intramammary route. The observation was made up to 30 days post-treatment (PT). The CMT of the healthy milk was negative (0), whereas, it ranged between 1 point score and 2 point score in SCM. The Somatic cell count (SCC) and Total bacterial count (TBC) decreased significantly (p<0.05) on day 3 PT in GrII cows in Enrofloxacin treated group, however, such changes were insignificant in PBS treated group. Traces of MPO and ACP enzyme were found in the healthy milk. The mean ACP level enhanced by 70% on day 3 PT in GrII and only 18.7% in Gr. III cows. The mean MPO level enhanced to 32% in Gr. II and 18 % in Gr. III cows on day 3 PT. Concomitant use of Enrofloxacin in SCM at sub optimal dose was found to reduce the bacterial load by increasing the bactericidal enzyme level in the milk polymorphonuclear cells (PMNs) in bovine SCM, which indicates its immunomodulatory potential in mastitis.

Inhibition of AP-1 by SARI negatively regulates transformation progression mediated by CCN1

        Dash, R,Su, Z-Z,Lee, S-G,Azab, B,Boukerche, H,Sarkar, D,Fisher, P B Macmillan Publishers Limited 2010 Oncogene Vol.29 No.31

Enhanced expression of the CCN family of secretory integrin-binding proteins correlates with many essential components of the cancerous state, including tumor cell adhesion, proliferation, invasion and migration. Consequently, CCN1 expression is elevated in various cancers, including breast cancer, and its expression directly correlates with poor patient prognosis. Using subtraction–hybridization, combined with induction of cancer cell terminal differentiation, we cloned SARI (suppressor of activator protein (AP)-1, regulated by interferon (IFN)), an IFN-β-inducible, potent tumor suppressor gene that exerts cancer-selective growth inhibitory effects. Forced expression of SARI using an adenovirus (Ad.SARI) inhibits AP-1 function and downregulates CCN1 expression in multiple cancer lineages, resulting in a profound inhibition in anchorage-independent cell growth and tumor cell invasion. Overexpression of SARI reduces CCN1-promoter activity through inhibition of AP-1 binding. Accordingly, SARI selectively blocks expression of the transformed state in rat embryo fibroblast cells that stably overexpress c-Jun. These results illustrate that SARI inhibits AP-1 transactivating factor binding to the cis-element of the CCN1 promoter, possibly through its interaction with c-Jun. Overall, SARI can directly inhibit CCN1-induced transformation by inhibiting the transcription of CCN1, as well as indirectly by inhibiting the expression of c-Jun (and hence blocking AP-1 activity). In these contexts, transformed cells ‘addicted’ to AP-1 activity are rendered susceptible to SARI-mediated inhibition of expression of the transformed phenotype.

Recent advancements in development and application of microbial cellulose in food and non-food systems

        O. P. Shemil Shahaban,Bhosale Yuvraj Khasherao,Rafeeya Shams,Aamir Hussain Dar,Kshirod Kumar Dash 한국식품과학회 2024 Food Science and Biotechnology Vol.33 No.7

Microbial cellulose is a fermented form of very pure cellulose with a fibrous structure. The media rich in glucose or other carbon sources are fermented by bacteria to produce microbial cellulose. The bacteria use the carbon to produce cellulose, which grows as a dense, gel-like mat on the surface of the medium. The product was then collected, cleaned, and reused in various ways. The properties of microbial cellulose, such as water holding capacity, gas permeability, and ability to form a flexible, transparent film make it intriguing for food applications. Non-digestible microbial cellulose has been shown to improve digestive health and may have further advantages. It is also very absorbent, making it a great option for use in wound dressings. The review discusses the generation of microbial cellulose and several potential applications of microbial cellulose in fields including pharmacy, biology, materials research, and the food industry.

        S. Srikrishnan,T. Shaafi,P.K. Dash 한양대학교 청정에너지연구소 2023 Journal of Ceramic Processing Research Vol.24 No.5

Industrial revolution and economic growth in any country is aided and abetted by effective defense research and developmenthand in hand with medical supremacy across the globe. Advent avionics system and advanced defense technology have sprungup from various nations further driving the need for hypersonic speed propulsion systems in transport or military applications. Pulse detonation systems may be an effective cure for such insolvency and may be used as an advanced highspeed propulsionsystem for various applications. The research work discusses the effect of cross-sections like cylinder, square and rectangle inachieving controlled detonation with supersonic velocity at exit and effective pressure for further expansion. The analysisresulted that the cylindrical cross-section with an orifice type blockage at three locations evenly spread throughout the lengthof the tube with 60 percent blockage ratio to be the most effective with maximum exit velocity of 2995.6 m/s and effectivepressure of 10.5 bar for an injection pressure of 100 bar and velocity 0.15 m/s. An experimental model using cold flow wasconducted to ratify the results from the numerical simulation and found to be similar with a differentiation of 14% due to thevariation of the working fluid.

Are Truelove and Witts criteria for diagnosing acute severe colitis relevant for the Indian population?A prospective study

        Saransh Jain,Saurabh Kedia,Sawan Bopanna,Dawesh P Yadav,Sandeep Goyal,Peush Sahni,Sujoy Pal,Nihar Ranjan Dash,Govind Makharia,Simon P. L. Travis,Vineet Ahuja 대한장연구학회 2018 Intestinal Research Vol.16 No.1

Background/Aims: Truelove and Witts criteria have been used to define acute severe colitis since the 1950s. However, hemoglobin(an additional criterion of the definition) levels in the general population in developing countries are lower than in the population of developed countries. We aimed to determine the relevance of Truelove and Witts criteria in the Indianpopulation. Methods: Consecutive patients with acute severe colitis satisfying the Truelove and Witts criteria, hospitalized ata single center between April 2015 and December 2016 were included. All patients received intravenous corticosteroids and 16 required colectomy. The hemoglobin levels at admission were subsequently excluded from the classification criteria, and the effect this had on the criteria for diagnosis was determined. Results: Out of 61 patients of acute severe colitis diagnosed according to the original Truelove and Witts criteria, 12 patients (20%) had 1 additional criterion, 33 (54%) had 2 additionalcriteria and 16 (26%) had 3 or more additional criteria in addition to 6 or more blood stained stools on admission. On excludinghemoglobin as an additional criterion from the Truelove and Witts definition, all patients still met the criteria for acute severecolitis. Conclusions: Truelove and Witts criteria can be used to define acute severe colitis in India, despite lower mean hemoglobinin the native population.

Stochastic free vibration analysis of smart random composite plates

        Singh, B.N.,Vyas, N.,Dash, P. Techno-Press 2009 Structural Engineering and Mechanics, An Int'l Jou Vol.31 No.5

The present study is concerned with the stochastic linear free vibration study of laminated composite plate embedded with piezoelectric layers with random material properties. The system equations are derived using higher order shear deformation theory. The lamina material properties of the laminate are modeled as basic random variables for accurate prediction of the system behavior. A $C^0$ finite element is used for spatial descretization of the laminate. First order Taylor series based mean centered perturbation technique in conjunction with finite element method is outlined for the problem. The outlined probabilistic approach is used to obtain typical numerical results, i.e., the mean and standard deviation of natural frequency. Different combinations of simply supported, clamped and free boundary conditions are considered. The effect of side to thickness ratio, aspect ratio, lamination scheme on scattering of natural frequency is studied. The results are compared with those available in literature and an independent Monte Carlo simulation.

      • Oscillatory spin-polarized tunnelling from silicon quantum wells controlled by electric field

        Jansen, Ron,Min, Byoung-Chul,Dash, Saroj P. Nature Publishing Group 2010 Nature materials Vol.9 No.2

Spin-dependent electronic transport is widely used to probe and manipulate magnetic materials and develop spin-based devices. Spin-polarized tunnelling, successful in ferromagnetic metal junctions, was recently used to inject and detect electron spins in organics and bulk GaAs or Si. Electric field control of spin precession was studied in III–V semiconductors relying on spin–orbit interaction, which makes this approach inefficient for Si, the mainstream semiconductor. Methods to control spin other than through precession are thus desired. Here we demonstrate electrostatic modification of the magnitude of spin polarization in a silicon quantum well, and detection thereof by means of tunnelling to a ferromagnet, producing prominent oscillations of tunnel magnetoresistance of up to 8%. The electric modification of the spin polarization relies on discrete states in the Si with a Zeeman spin splitting, an approach that is also applicable to organic, carbon-based and other materials with weak spin–orbit interaction.
