        Molecular cloning and characterization of novel human JNK2 (MAPK9) transcript variants that show different stimulation activities on AP-1

        The c-Jun NH2-terminal kinase (JNK) signaling pathway participates in many physiological functions. In the current study we reported the cloning and characterization of five novel JNK2 transcript variants, which were designated as JNK2 α3, JNK2 α4, JNK2 β3, JNK2 γ1 and JNK2 γ2, respectively. Among them, JNK2 α4 and JNK2 γ2 are potential non-coding RNA because they contain pre-mature stop codons. Both JNK2α3 and JNK2β3 contain an intact kinase domain, and both encode a protein product of 46 kDa, the same as those of JNK2α1 and JNK2β1. JNK2γ1 contains a disrupted kinase domain and it showed a disable function. When over-expressed in mammalian cells, JNK2α3 showed higher activity on AP-1 than that of JNK2β3 and JNK2γ1. Furthermore, JNK2α3 and JNK2β3 showed different levels of substrate phosphorylation, although they both could promote the proliferation of 293T cells. Our results further demonstrate that JNK2 isoforms preferentially target different substrates and may regulate the expression of various target genes. [BMB reports 2010; 43(11): 738-743]

      • CD3<sup>+</sup> CD4<sup>+</sup> and CD3<sup>+</sup> CD8<sup>+</sup> Lymphocyte Subgroups and their Surface Receptors NKG2D and NKG2A in Patients with Non-small Cell Lung Cancer

        Background: To explore the prevalence of lymphocyte subgroups $CD3^+$ $CD4^+$ and $CD3^+$ $CD8^+$ and their surface receptors NKG2D and NKG2A in patients with non-small cell lung cancer (NSCLC). Materials and Methods: A total of 40 patients with NSCLC were divided into different groups according to different clinical factors (TNM staging, pathological patterns and genders) for assessment of relations with $CD3^+$ $CD4^+$ and $CD3^+$ $CD8^+$ and the surface receptors NKG2D and NKG2A of T lymphocytes in peripheral blood by flow cytometry. Results: Patients in the advanced group had evidently lower levels of $CD3^+$ $CD4^+$ but markedly higher levels of $CD3^+$ $CD8^+$ in peripheral blood than those with early lesions (p<0.05). In addition, NSCLC patients in the advanced group had obviously higher $CD3^+$ $CD4^+$ NKG2D and $CD3^+$ $CD8^+$ NKG2A expression rates but lower $CD3^+$ $CD4^+$ NKG2A and $CD3^+$ $CD8^+$ NKG2D expression rates (p<0.05). However, there were no significant differences between NSCLC patients with different genders and pathological patterns in expression levels of lymphocyte subgroups $CD3^+$ $CD4^+$ and $CD3^+$ $CD8^+$ and their surface receptors NKG2D and NKG2A. Conclusions: Unbalanced expression of surface receptors NKG2D and NKG2A in $CD3^+$ $CD4^+$ and $CD3^+$ $CD8^+$ lymphocytes may be associated with a poor prognosis, greater malignancy and immunological evasion by advanced cancers, related to progression of lung cancer.

      • Pulmonary Lobectomy Combined with Pulmonary Arterioplasty by Complete Video-assisted Thoracic Surgery in Patients with Lung Cancer

        Objective: To explore the feasibility of pulmonary lobectomy combined with pulmonary arterioplasty by complete video-assisted thoracic surgery (VATS) in patients with lung cancer, and summarize its surgical methods. Materials and Methods: Twenty-one patients with lung cancer in Beijing Chest Hospital Affiliated to Capital Medical University from Feb., 2010 to Jun., 2013 were selected, males and females accounting for 15 and 6 cases, respectively. Ten underwent right upper lobectomy, 5 right lower lobectomy, 4 left upper lobectomy (in which left upper sleeve lobectomy was conducted for 2) and 2 left lower lobectomy. At the same time, local resection of pulmonary arterioplasty was performed for 12 patients, and sleeve resection of pulmonary arterioplasty for 9. Results: Twenty-one patients recovered well after surgery. Thoracic drainage tube was maintained for 3-8 days, with an average of 4.9 days, and hospital stays were 8-15 days, with an average of 11 days. There were no deaths in the perioperative period, and the complications like pulmonary embolism, bronchopleural fistula, chest infection and pulmonary atelectasis did not occur after surgery. Conclusions: Performance of pulmonary lobectomy and pulmonary arterioplasty together by complete VATS is a safe and effective surgical method, which can expand the indications of patients with lung cancer undergoing thoracoscopic pulmonary lobectomy, and make more patients profit from such minimally invasive treatment.

        孫悟空大鬧天宮故事素材來源新說

        There are three diffrent opinions on the source material of the story about Sun Wukong(孫悟空)’s fight with Huangdi(黃帝)(which we called Danao Tiangong(大鬧天宮)). In my opinion,although there are some similiars between the story of Danao Tiangong(大鬧天宮) and the story which is from Baopuzi Neipian(≪抱樸子ㆍ內篇≫ mentioned in Wang Sisi(王四四)’s paper, and thestory which is named “Dingshengwang’s raising to the sky(頂生王昇天)”mentioned in Chen Yinque(陳寅恪)’s paper,but the relevances between the stories are not mentioned in their papers above. The viewpoint claimsed in this paper is that the source material of the story of Danao Tiangong(大鬧天宮) is the story of Chiyou(蚩尤)’s fight with Huangdi (黃帝).Evidences are as follows.Firtly,there are also many similiars between the story of Chiyou(蚩尤)’s fight with Huangdi(黃帝) annd the story of Danao Tiangong(大鬧天宮).In this paper meticulous compare and analysis are did to reply it.Secondly, and also the more importantly,the relevances between the two stories are prompted .That is the image of Sun Wukong(孫悟空) is fused with the image of Chiyou(蚩尤) . For example, Sun Wukong(孫悟空)’s external image comes from Chiyou(蚩尤). Additionally, the detail about Sun Wukong(孫 悟空)’s eating iron shot and drinking copper juice druing the period when he is suppressed under the wuxing mountain(五行山) , is from the records of the tribe of chiyou’s eating iron shot and drinking copper. Accoriding to the similiars between the story of Chiyou(蚩尤)’s fight with Huangdi(黃帝) and the story of Danao Tiangong(大鬧天宮), and the relevances between the two stories,we can make a conculion that the story of chiyou(蚩 尤)’s fight with Huangdi(黃帝)is the prototype of the story of Danao Tiangong(大鬧天宮). In another word,the conculion is that the source material of the story of Danao Tiangong(大鬧天宮) is the story of Chiyou(蚩尤)’s fight with Huangdi(黃帝)

        朴祥植≪東渡日史≫所載漢詩硏究

        In the year 1880, Park Sang-sik (朴祥植) followed the second Xiuxinshi (修信 使) Kim Hongjib (金弘集) as Xiangshuji (鄕書記) to go to Japan. From June 25th when they left from Donglaifu (東萊府) in Busan, till August 12th when they returned from Japan, during this period, Park Sang-sik (朴祥植) recorded what he saw and listened in Japan and edited it into a book named Dongdoilsa (≪東渡 日史≫). Dongdoilsa (≪東渡日史≫) is formed with three parts. The mainest and also the most literary part is his diary recorded in Dongdoilsa (≪東渡日史≫). What needs us to pay attention is his 17 poems in the dairies. It is the only way for us to understand Park Sang-sik (朴祥植)’s poems written in Chinese. But till now there are not so many research findings on Park Sang-sik (朴祥植)’s poems written in Chinese. The purpose of this paper is to do research on these poems.

        Rapid characterization of ginsenosides in the roots and rhizomes of Panax ginseng by UPLC-DAD-QTOF-MS/MS and simultaneous determination of 19 ginsenosides by HPLC-ESI-MS

        Background: Ginsenosides are the characteristic and principal components which manifest a variety of the biological and pharmacological activities of the roots and rhizomes of Panax ginseng (GRR). This study was carried out to qualitatively and quantitatively determine the ginsenosides in the cultivated and forest GRR. Methods: A rapid and sensitive ultra-high-performance liquid chromatography coupled with diode-array detector and quadrupole/time of flight tandem mass spectrometry (UPLC-DAD-QTOF-MS/MS) was applied to the qualitative analysis of ginsenosides and a 4000 QTRAP triple quadrupole tandem mass spectrometer (HPLC-ESI-MS) was applied to quantitative analysis of 19 ginsenosides. Results: In the qualitative analysis, all ingredients were separated in 10 min. A total of 131 ginsenosides were detected in cultivated and forest GRR. The method for the quantitative determination was validated for linearity, precision, and limits of detection and quantification. 19 representative ginsenosides were quantitated. The total content of all 19 ginsenosides in the forest GRR were much higher than those in the cultivated GRR, and were increased with the growing ages. Conclusion: This newly developed analysis method could be applied to the quality assessment of GRR as well as the distinction between cultivated and forest GRR.

        『西游記』中的湖北方言與吳承恩的“荊府紀善”之任

        오승은이 만년에 荊府紀善이란 직책에 재임하였는가에 대한 문제는 80년대부터 오늘날까지 학계에서 계속해서 논의되어 왔다. 최근 蔡鐵鷹 등과 같은 학자들은 심도 있는 고증을 바탕으로 오승은이 荆府纪善이란 직책에 재임한 사실에 대해 보다 설득력 있는 논증을 펴내고 있다. 이는 『서유기』 연구자들의 주목을 받았으나 아직까지 이러한 관점은 학계에서 정론으로 인정받지 못하고 있다. 필자는 『서유기』에 대량의 호북의 방언 어휘와 문법을 활용하였다는 점에서 오승은이 호북성에서 긴 시간을 머물렀다고 보았다. 그렇지 않고서야 『서유기』에 호북 방언의 어휘와 문법이 많이 활용되었다는 점을 이해하기가 힘들것이다. 오승은의 생애를 살펴보면, 그가 호북성과 관계를 맺게 될 수 있었던 계기는 長興縣丞으로 벼슬을 하였을 때 억울한 누명을 벗긴 이후 받은 荊府紀善이란 관직의 补任과 유일하게 관련된다. 따라서 필자는 오승은이 명나라 때 호북에 위치한 荊王府의 荊府紀善이란 관직에 실제로 재임하였을 뿐만 아니라 그 재임기간도 또한 짧지 않았을 것이라고 본다. 비록 『서유기』에 호북 방언이 대량적으로 활용되는 것만으로 오승은이 荊府紀善이란 관직을 실제로 역임했다는 사실을 증명하기란 쉽지 않지만, 역으로 생각하면 『서유기』 내 호북 방언의 대량 사용을 통해 오승은이 荊府紀善이란 관직을 실제로 역임하였다는 사실에 대해서는 방증할 수 있을 것이다. 이것이 바로 본 연구의 의미와 가치이다. 만약 오승은이 荊府紀善이란 관직을 실제로 역임하였다면, 『서유기』는 오승은이 荊府紀善이란 관직에 재임하는 동안 창작된 작품이라고 볼 수 있으며, 이는 우리가 주목할 만한 결론이라고 할 수 있겠다. From 1980s till today, scholars spent so much time on discussing the question whether Wu Cheng’en(吳承恩) took up the position named Jingfu Jishan(荊府紀善). Recently, Some scholars such as Cai Tieying(蔡鐵鷹) proved that Wu Cheng’en did take up the position, which is proved by well-informed textual research.It attracts many scholars’ attention and support. Even so this viewpoint has not been a conclusion and consensus in the academic circle. According to my opinion,through the vocabularies and the grammars of Hubei province’s dialect which is been used of large number in the novle 『Journey to the West(西游記)〉, we can prove that the author of 『Journey to the West』 had stayed in Qizhou(蕲州)which belonged to Hubei Province for a long time. Or, it’s very difficult to explain why there are so many vocabularies and the grammars of the dialect of Hubei province used in the novel 『Journey to the West』. But according to the arguments that we have, during Wu Cheng’en’s whole life,there’s just one possibility that made him be associated with Hubei Province, where he worked as an official named Jingfujishan(荊府紀善)in Qizhou of Hubei province after he was wronged as an County Magistrate(縣丞) in Changxing(長興).Therefore,it can be concluded that Wu Cheng’en did work as Jingfujishan(荊府紀善) in Qizhou of Hubei province and he stayed there for a long time.Although the existence of so many vocabularies and the grammars of the dialect of Hubei province which are been used of large number in the novle 『Journey to the Wes t』 can not be considered as a irrefutable evidence,but it also can be used as a circumstantial evidence. This is the significance and value of this paper. If Wu Cheng’en did work as Jingfujishan(荊府紀善) in Qizhou of Hubei province and work for a long time, the viewpoint that the novle 『Journey to the West』 is written by Wu Cheng’en when he worked as Jingfujishan(荊府紀善) is a viewpoint that we should take seriously.

        Face Stability Analysis of Slurry Shield Tunnels in Rock–Soil Interface Mixed Ground

        When shield tunneling in rock-soil interface (RSI) mixed ground, maintaining the stability of the tunnel face is very challenging. This study aims at the investigation of the face stability of slurry shield tunnels in RSI mixed ground. A new apparatus was developed to simulate pressurized slurry supporting the tunnel face in centrifuge tests. The progressive failure of the tunnel face was triggered with slow decrease of slurry pressure. With two equivalent model tunnels of circular shape and semicircular shape, the variations of soil (water) pressure and the displacement field in front of the tunnel can be obtained simultaneously. The face failure was found to be a local collapse in the upper soil layer. The limit support pressure was picked out at the sudden change of the measured soil or water pressures. Then the classical wedge-prism model was introduced and modified to calculate the limit support pressure for slurry shield tunnels in RSI mixed ground considering the partial collapse. Numerical analysis was applied to verify the theoretical analysis and sensitivity analysis was performed to estimate the influence of different parameters on the theoretical solutions.

        A Novel 7-O-Modified Genistein Derivative with Acetylcholinesterase Inhibitory Effect, Estrogenic Activity and Neuroprotective Effect

        To find the multi-target-directed compounds for the treatment of Alzheimer’s disease (AD), we synthesized 7-(4-(diethylamino)butoxy)-5-hydroxy-3-(4-hydroxyphenyl)-4H-chromen-4-one, a novel 7-O-modified genistein derivative (GS-14), and investigated its acetylcholinesterase (AChE) inhibitory effect, estrogenic activity and neuroprotective effect. GS-14 acted as a selective AChE inhibitor in vitro, with an IC50 value of 0.17 μM and showed no inhibition activity against butyrylcholinesterase (BuChE). The Lineweaver–Burk plot revealed that GS-14 was a non-competitive AChE inhibitor with a Ki value of 0.23 μM and the molecular docking model indicated that GS-14 interacted with the peripheral anionic site (PAS) of AChE. The MCF-7 proliferation assay demonstrated that GS-14 possessed estrogenic activity and GS-14 exhibited a high specificity for estrogen receptor β (ERβ) with a dissociation constant (Ki) of 2.86 nM compared with that of 1.01 μM for estrogen receptor α (ERα) in the molecular docking study. GS-14 also possessed a neuroprotective effect and showed the best protective effect against the β-amyloid protein-induced injury on SH-SY5Y cells at a concentration of 1 nM. Considering its AChEinhibition activity, estrogenic activity and neuroprotective effect, GS-14 may be a potential multi-target agent for the treatment of AD.

      • ON THE END EXTENDING IN THE HILBERT-HUANG TRANSFORM

        Hilbert-Huang transform is powerful method of nonlinear and non-stationary time series analysis. Two approaches, mirror periodic and extrema extending method, have been developed for handling the end effects of Hilbert-Huang transform. The mirror periodic method is based on the data distribution at ends. Two mirrors are placed at the extrema of both ends. The data within the mirrors are extended with mirror reflection to obtain a periodic sequence. With this method, the end effects of empirical mode decomposition and Hilbert transform are solved once forever. The mirror periodic method can get best results by placed the mirrors at the extrema, which have symmetric property of the data. Extrema extending method is implemented by adding two maxima and minima at the ends, which are defined by the two consecutive extrema of the data at ends. It is easy to perform and has equally well behalf as mirror periodic method. The extrema method is superior to the mirror periodic method when the data has strong asymmetric waveform.
