        A non-muscle myosin heavy chain 9 genetic variant is associated with graft failure following kidney transplantation

        Background: Despite current matching efforts to identify optimal donor-recipient pairs for kidney transplantation, alloimmunity remains a major source of late transplant failure. Additional genetic parameters in donor-recipient matching could help improve long-term outcomes. Here, we studied the impact of a non-muscle myosin heavy chain 9 gene (MYH9) polymorphism on allograft failure. Methods: We conducted an observational cohort study, analyzing the DNA of 1,271 kidney donor-recipient transplant pairs from a single academic hospital for the MYH9 rs11089788 C>A polymorphism. The associations of the MYH9 genotype with risk of graft failure, biopsy-proven acute rejection (BPAR), and delayed graft function (DGF) were estimated. Results: A trend was seen in the association between the MYH9 polymorphism in the recipient and graft failure (recessive model, p = 0.056), but not for the MYH9 polymorphism in the donor. The AA-genotype MYH9 polymorphism in recipients was associated with higher risk of DGF (p = 0.03) and BPAR (p = 0.021), although significance was lost after adjusting for covariates (p = 0.15 and p = 0.10, respectively). The combined presence of the MYH9 polymorphism in donor-recipient pairs was associated with poor long-term kidney allograft survival (p = 0.04), in which recipients with an AA genotype receiving a graft with an AA genotype had the worst outcomes. After adjustment, this combined genotype remained significantly associated with 15-year death-censored kidney graft survival (hazard ratio, 1.68; 95% confidence interval, 1.05-2.70; p = 0.03). Conclusion: Our results reveal that recipients with an AA-genotype MYH9 polymorphism receiving a donor kidney with an AA genotype have significantly elevated risk of graft failure after kidney transplantation.

        <P></P> <P>The CCAUV.U-K3.1 is a follow-up of the comparison CCAUV.U-K3 which was the third key comparison in the ultrasound branch organised under the auspices of the Consultative Committee for Acoustics, Ultrasound and Vibration (CCAUV) of the BIPM. It was necessary to allow participants with discrepant values and additional laboratories to have another possibility to take part in an international comparison and to test their measurement facilities. The comparison dealt with power measurement in the frequency range 2 to 10.5 MHz and a power range between 10 mW and 15 W.         Nutrient requirements and evaluation of equations to predict chemical body composition of dairy crossbred steers

        Objective Objectives were to estimate energy and protein requirements of dairy crossbred steers, as well as to evaluate equations previously described in the literature (HH46 and CS16) to predict the carcass and empty body chemical composition of crossbred dairy cattle. Methods Thirty-three Holstein×Zebu steers, aged 19±1 months old, with an initial shrunk body weight (BW) of 324±7.7 kg, were randomly divided into three groups: reference group (n = 5), maintenance level (1.17% BW; n = 4), and the remaining 24 steers were randomly allocated to 1 of 4 treatments. Treatments were: intake restricted to 85% of ad libitum feed intake for either 0, 28, 42, or 84 d of an 84-d finishing period. Results The net energy and the metabolizable protein requirements for maintenance were 0.083 Mcal/EBW0.75/d and 4.40 g/EBW0.75, respectively. The net energy (NEG) and protein (NPG) requirements for growth can be estimated with the following equations: NEG (Mcal/kg EBG) = 0.2973(±0.1212) ×EBW0.4336(±0.1002) and NPG (g/d) = 183.6(±22.5333)×EBG−2.0693(±4.7254)×RE, where EBW, empty BW; EBG, empty body gain; and RE, retained energy. Crude protein (CP) and ether extract (EE) chemical contents in carcass, and all the chemical components in the empty body were precisely and accurately estimated by CS16 equations. However, water content in carcass was better predicted by HH46 equation. Conclusion The equations proposed in this study can be used for estimating the energy and protein requirements of crossbred dairy steers. The CS16 equations were the best estimator for CP and EE chemical contents in carcass, and all chemical components in the empty body of crossbred dairy steers, whereas water in carcass was better estimated using the HH46 equations.         Insect Resistance and Horticultural Trait Genetic Values of Potato Families

        This work aimed to evaluate 11 potato families for insect resistance horticultural traits. The families were derived from crosses between introduced insect resistant and adapted genotypes. A randomized complete block design, with three replications was used. The plot consisted of 25 genotypes of a family, summing up to 75 genotypes tested per family. Two susceptible cultivars were included in the experiment as checks. The genotypes were evaluated for insect resistance, tuber yield traits, tuber appearance and tuber skin smoothness. The genetic value for the intensity of insect attack in the leaves and in the tubers was lower(higher resistance) in the 11 families than in the checks. For yield traits, there was no predominant effect among the families regarding the origin of the resistant donor species(Solanum berthaultii and/or S. chacoense). However, there was a large contribution of the C-1485-16-87 recurrent parent in crosses with resistant genotypes for tuber yield and tuber number. In relation to tuber appearance, only the family derived form the C-1485-16-87/ ND140(S. berthaultii) cross did not differ from the checks. had genetic value tuber skin smoothness, it was identified that families derived from S. berthaultii resistant parents showed higher genetic values than the check cultivars. For tuber skin smoothness, the genetic values of the families neither differed significantly from each other nor from the checks.

        Genetic Distances Among Rice Mutant Genotypes Assessed by AFLP and Aluminum Tolerance-Related Traits

        Increasing genetic variability with mutagenic agents has been broadly employed in plant breeding because it has the potential to alter one or more desirable traits. In this study, a molecular analysis assessed by Amplified Fragment Length Polymorphisms(AFLPs) and a morphological analysis based on seedlings subjected to aluminum stress were compared. Also, an analysis of allelic frequencies was performed to observe unique alleles present in the pool. Genetic distances ranging from 0.448 to 0.953 were observed, suggesting that mutation inducing was effective in generating variability. The genetic distances based on morphological data ranged from 0(genotypes 22 and 23) to 30.38(genotypes 15 and 29). In the analysis of allelic frequency, 13 genotypes presented unique alleles, suggesting that mutation inducing was also targeting unique sites. Mutants with good performance under aluminum stress(9, 15, 18 and 27) did not form the same clusters when morphological and molecular analyses were compared, suggesting that different genomic regions may be responsible for their better performance.
