      • 증가 분산을 보이는 오차하에서 Michaelis-Menten Kinetic 상수 추정치의 분석을 위한 5가지 방법들의 비교

        Lineweaver-Burk 방법은 등분산 오차와는 달리 증가분산 오차인 경우 K?을 계사낳는 좋은 방법임을 앞의 결과들로 부터 알 수 있었다. 즉 기질농도 범위가 다른 방법에 비해 넓다. 그러나 정확도와 정도 모두 다른 방법에 비해 우수하였으며 오차의 크기에도 영향을 적게 받는다. Hanes-Woolf 방법은 추정치 K?의 C.V.가 기질 농도 범위 증가에 따라 급격히 증가하였고 이에 따른 정확도로 나빠졌다. 정확한 기질농도 범위를 선택한 경우 정확한 K?을 추정할 수 있으나(Table 2)그렇치 않은 경우는 매우 위험한 방법이었다. Woolf-Augustinsson-Hofstee 방법은 다른 방법들에 비해 K? 추정치의 C.V.가 가장 작은 방법이었으나 K?-K?의 분포가 각 실험에서 다른 나머지 방법에 비하여 크게 나타나 정확도가 좋지 않은 방법이었다. 이와 같은 결과가 실험 I과 실험 III에서 추정치들의 C.V.는 가장 작으나 K?-K? 분포가 가장 크게 나타났었다. 또한 Fig.5에서 보면 측정 오차가 커지면 K? 추정치는 실제값보다 작게 측정되는 경향이 있음을 볼 수 있었다. Wilkinson-Cleland 방법은 정확도에서 좋은 방법이었다.그러나 K? 추정치의 C.V.가 크며 (Table2)정확한 기질 농도 범위를 사용하지 않으면 실제값보다 작은 추정치를 가지는 경향이 있음을 알 수 있었다(Fig.2). 정확한 기질농도 범위에서 실험을 할 경우 정확도는 Hanes-Woolf 방법, Wilkinson-Cleland 방법, Lineweaver-Burk 방법,Direct linear 방법, Direct linear 방법, Woolf Augustinsson-Hofstee 방법의 순서로 양호하였다. 같은 효소에서 저해제나 활성제의 영향을 서로 비교할 때는 추정치들의 분산이 작은 Woolf-Augustinsson-Hofstee 방법이 좋다. 적정 기질농도 범위는 약 2 K?에 서 6K?사이로 각 방법에 따라 조금씩 차이가 있었으며 최소 필요 기질농도 수는 10개 내외가 적당하였다. The Michaelis-Menten equation was fitted to simulated data showing the increasing variance error by using three linear transformations(Lineweaver-Burk, Hanes-Woolf, Woolf-Augustinsson-Hofstee), and Cornsih-Bowen's direct linear plot and the method of Wikinson Cleland. The best method under the well definded conditions was those of Hanes-Wkklf's. The Woolf-Augustinsson-Hofstee plot appeared the worst method. The proper fanges of substrate concentration were different from each other between even-spaced within the defined range, were adequate.

      • Comparison of Monthly Ibandronate Versus Weekly Risedronate in Preference, Convenience, and Bone Turnover Markers in Korean Postmenopausal Osteoporotic Women

        <P>Patient preferences, convenience, and bone turnover markers were evaluated for the monthly ibandronate over the weekly risedronate regimen in Korean postmenopausal osteoporotic women. This was a 6-month, prospective, randomized, open-label, multicenter study with a two-period and two-sequence crossover treatment design. After a 30-day screening period, eligible participants with postmenopausal osteoporosis were randomized to receive either monthly oral ibandronate 150 mg for 3 months followed by weekly oral risedronate 35 mg for 12 weeks (sequence A) or the same regimen in reverse order (sequence B). Patient preference and convenience were evaluated by questionnaire. The changes in serum C-telopeptide after 3 months of treatment were analyzed. A total of 365 patients were enrolled in this study (sequence A 182, sequence B 183). Of patients expressing a preference (83.4%), 74.8% preferred the monthly ibandronate regimen over the weekly regimen (25.2%). More women stated that the monthly ibandronate regimen was more convenient (84.2%) than the weekly regimen (15.8%). There was no significant difference in the change in bone turnover marker between the two treatments. The two regimens were similarly tolerable. There were fewer adverse events in the monthly ibandronate group compared to the weekly risedronate group in terms of gastrointestinal side effects (nausea and abdominal distension). This study revealed a strong preference and convenience for monthly ibandronate over weekly risedronate in Korean postmenopausal osteoporotic women. There was no significant difference in change of bone turnover marker and safety profile between the two regimens.</P>

        서비스조직의 조직문화와 인적자원요소가 조직유효성에 미치는 영향

        The main purpose of this study is to testify the relationships among organizational culture, human factor and organizational effectiveness in service organizations by focusing on mediating effect of hotel employee`s followership as follows; First, the structure of more effective hotel organization culture will be identified. Second, the style of more effective human resource factor-entrepreneurship and leadership-will be identified. Third, the relationship among the hotel organization culture and entrepreneurship, leadership, organizational effectiveness will be statistically tested, and the relative importance levels will be identified. Finally, the mediating effect of followership of hotel employee between the hotel organization culture and human resource factor will be identified. The result of this study were as follows; First, Hypothesis H1; Both organization culture of mutual agreement and organization culture of development will have a meaningful effect on the hotel organizational effectiveness. Second, Entrepreneurship has positive relationship with Organizational Effectiveness. In other words, both innovation and risk-taking appeared to improve organizational effectiveness through mediating effect of followership. Third, the leadership type having the greatest effect on organizational effectiveness was shown to transformational leadership. It is necessary to conduct a study on a new paradigm leadership found to have a high effect on organizational effectiveness in the process of analysis. And it is necessary to consider the method of its application. Fourth, it is approved that followership is an important factor to increasing organizational effectiveness. As a result, followership had a meaningful mediating effect on organizational effectiveness in order to improve hotel management performance.

        개에서 Protamine Sulfate 에 의한 혈압 하강 기전

        Introduction : Protamine reversal of heparin anticoagulation often produces profound hypotension. However, the precise mechanisms of its hypotensive effect have not been fully elucidated. Using a canine model, we explored the effects of cyclo-oxygenase inhibitor, indomethacin (INDO), and nitric oxide synthetase inhibitor, NG-nitro-L-arginine methyl ester (L-NAME) either alone or both on the cardiopulmonary responses to protamine. Methods : Fifty-four mongrel dogs in five groups were studied during 1.5% halothane anesthesia. GroupI(n=17) received heparin (300 IU/kg iv) followed by protamine (3 mg/kg iv over 30 s) 5 min after the heparin. The same protocol were used in groups II(n=11), III (n=12), and IV (n=7), except that L-NAME (20 mg/kg), INDO (10 mg/kg), and INDO (10 mg/kg) plus L-NAME (10 mg/kg) were infused over 10 min beginning 30 min before the protamine injection, respectively. Animals in group V (n=7) were given protamine (3 mg/kg) alone. Mean arterial pressure (MAP), mean pulmonary arterial pressure (MPAP), left ventricular end-diastolic pressure (LVEDP), LVdP/dt and cardiac output and left circumflex coronary flow (LCX flow) via Doppler flowmeter and heart rate were continuously recorded in baseline conditions and up to 15 min. Plasma NOx (NO2-, NO3-) levels were also measured before (baseline) and 3, 5, 10, and 15 min after protamine injection. Results: In group I, protamine caused immediate but transient decreases of MAP (41%), cardiac index (CI, 58%), dP/dt (28%), and LVEDP (62%) and increases of MPAP (38%) and systemic and pulmonary vascular resistance indices (SVRI, 30%; PVRI, 316%). INDO significantly attenuated the hemodynamic responses to protamine, whereas L-NAME did not affect them at all. INDO plus L-NAME prevented protamine-induced hypotension, but CI ( 24%) and LVEDP ( 30%) showed similar changes as those in group II. Protamine increased MPAP but inconsistently, meanwhile no correlation was found between the magnitude of increase of MPAP and decrease of MAP at peak responses in groups I-IV. LCX flow increased significantly (124∼188%) immediately after protamine infusion without any changes in plasma NOx levels in groups I-IV. Neither significant hemodynamic effects nor NOx release was found in animals given protamine alone. Conclusion : Protamine in the presence of heparin induces profound hypotension which may be mediated by a prostanoid and other potent vasodilators. In addition, increase of PAP and NO release may not play a significant role in the protamine-induced hypotension. (Korean J Anesthesiol 1998; 34: 27∼38)

        전기방사로 제작된 재이용 가능한 PAN/제올라이트 섬유의 암모니아 흡착 특성

        BACKGROUND: Generally, ammonia occurs from agricultural waste disposal. Ammonia is known as a harmful substance to the human body and has a bad influence such as eutrophication on the ecosystem. It is possible to remove the ammonia by ammonia adsorption method using natural zeolite, without external influence. However, due to the natural zeolite shape, it is hard to reuse. METHODS AND RESULTS: Electrospinning method can produce fiber with constant diameter. Moreover, electrospinning method has no limitation for selecting the material to make the fiber, and thus, it is valuable to reform the surface of adsorbent. In this study, reusable membrane was made by electrospinning method. The highest removal efficiency was shown from the membrane with 20% of zeolite included, and it has been verified that it is possible to reuse the membrane through chemical treatment. The highest ammonia removal efficiency was about 92.4%. CONCLUSION: In this study, ammonia adsorption characteristics of zeolite fibers were studied. Electrospinning method can produce zeolite fiber with even distribution. Ammonia can be removed efficiently from ion exchange ability of the natural zeolite. The result of adsorption isotherm indicated that both Freundlich model and Langmuir model provided the best fit for equilibrium data. And study on desorption has demonstrated that the ion exchange from zeolite was reversible when 0.01 M NaCl and KCl solution were used.
