      • PPAR-Gamma Pro12Ala Polymorphism and Gastric Cancer Risk in a Turkish Population

        Background: Peroxisome proliferator-activated receptor gamma ($PPAR{\gamma}$) is a ligand dependent transcription factor involved in various processes, including carcinogenesis. We aimed to investigate any possible association of the $PPAR{\gamma}$ Pro12Ala (rs1801282) polymorphism with risk of developing gastric cancer (GC). Patients and Methods: A hospital based case control study was designed covering 50 patients with GC and 120 healthy controls. The frequencies of $PPAR{\gamma}$ Pro12Ala (rs1801282) were determined using a polymerase chain reaction-restriction fragment length polymorphism (PCR-RFLP) assay. Results: The Ala12 allele of the $PPAR{\gamma}$ Pro12Ala G gene was associated with a 1.95 fold increased risk of GC development (p: 0.022; 95% CI: 1.58-2.40). Subgroup analyses showed that the same allele was also associated with metastasis (p: 0.000; OR:4.09; 95%CI:2.273-7.368) and differentiation (p: 0.004; OR:1.95; 95%CI:1.335-2.875) in patients with GC. Conclusion: This study suggests that the $PPAR{\gamma}$ Pro12Ala G (Ala12) allele might be associated with development, differentiation and metastatic process of GC in the Turkish population. Further studies conducted in larger study groups and in different ethnic populations will be needed to clarify the exact role of the $PPAR{\gamma}$ Pro12Ala polymorphism in GC.

      • Thermostructural shape memory effect observations of ductile Cu-Al-Mn smart alloy

        The Cu-Al-Mn shape memory alloy (SMA) with a new different composition was fabricated by arc melting method. The characteristic shape memory effect (SME) property of Cu-Al based SMA was revealed by performing thermostructural measurements. The differential scanning calorimetry (DSC) tests were taken to observe the reversible martensitic phase transformation peaks of the alloy as evidence of SME property of the alloy. To determine the basic thermodynamical parameters of the alloy, these endothermic and exothermic transformation peaks were analyzed by the tangent differentiation method that was performed automatically by the DSC analyzing program over a manually selected part on the DSC curve and by these analyses the characteristic martensitic transformation temperatures (working temperatures) that found below 100℃ and the enthalpy change values of the alloy were directly obtained. The other kinetic transformation parameters of the alloy - the entropy change, hysteresis, and equilibrium temperature - were also determined. The common high-temperature behavior of the Cu-Al based Heusler alloys was detected by differential thermal analysis (DTA) measurement. The XRD and metallography tests that were conducted at room temperature showed the presence of M18R and the dominant 2H martensite structures that formed in the alloy and this dual martensitic structure was also prescribed by determining the theoretical e/a ratio of the alloy. Furthermore, the microhardness tests on the alloy demonstrated the high ductility feature of the alloy. All results demonstrated that the CuAlMn alloy exhibiting a shape memory effect property can be useful in smart alloy applications.

        Comparison of code provisions on lap splices

        The code provisions on lap splices are critically assessed in the light of 203 beams without transverse reinforcement and 278 beams with transverse reinforcement. For comparison, the provisions given in the ACI 318, Eurocode 2, and TS 500 Codes are considered. The ACI Committee 408 recommended provision and a new proposal are also taken into account throughout the assessment. The comparison with real beam tests where the splice region was subjected to constant moment indicates that current provisions in the Codes do not agree acceptably with test results. The steel stress prediction graphs calculated by means of the Code provisions show high scatter and remain unsafe especially for test data without transverse reinforcement. Both the recent recommended provision by ACI Committee 408 and a new design expression proposed by the author have much less scatter with fewer unsafe predictions. The simplified design provision proposed by ACI Committee 408 does not yield similar results to that of the advanced design provision proposed by the same committee and therefore it could conveniently be replaced with the simpler equation proposed by the author.

      • GSTT1 is Deregulated in Left Colon Tumors

        Our aim was to determine GSTT1 expression levels in left colon tumors and paired normal tissue in order to identify specific alterations in GSTT1 mRNA levels. Alterations in GSTT1 expression in twenty-four left-sided colon tumors and paired cancer free tissue were determined by qRT-PCR. Significant fold changes were determined with t-test. When compared with cancer free tissue, left colon cancers showed a significant decrease in GSTT1 expression. However, GSTT1 mRNA levels among different grades increased gradually in correlation with tumor grade. Our results suggest that downregulation of GSTT1 in left-sided colon cancers is an early event and is reversed with cancer progression, probably due to cellular defense mechanisms as a response to changes in the microenvironment.

      • 5-Aminolevulinic Acid Fluorescence in Detection of Peritoneal Metastases

        Background: The value of 5-aminolevulinic acid (ALA) in fluorescence detection of peritoneal metastases and the underlying mechanisms were evaluated in patients with peritoneal surface malignancies. Materials and Methods: Oral 5-ALA was administered at a concentration of 20 mg/kg body weight with 50 ml of water 2 hours prior to surgery (n=115). The diagnostic value of 5-ALA based fluorescence production was evaluated following white light inspection during prior to cytoreductive surgery and hyperthermic intraperitoneal chemotherapy. Then, peptide transporter PEPT1 (ALA influx transporter) and ATP-binding cassette transporter ABCG2 (porphyrin efflux transporter) gene expression was determined with quantitative real time (qRT)-PCR and pathological diagnoses confirmed for all tissue samples. Results: The 5-ALA based photodynamic detection rate was 17% for appendiceal mucinous neoplasms, 54% for colorectal cancers, 33% for gastric cancers, 67% for diffuse malign peritoneal mesotheliomas, and 89% for epithelial ovarian cancer of peritoneal metastases. 5-ALA was detected in all cases of peritoneal metastases originating from cholangiocarcinomas whereas it was not able to detect any in granulosa cell and gastrointestinal stromal tumor cases. Furthermore, PEPT1 was overexpressed whereas ABCG2 expression was downregulated in tumors detected with fluorescence. Conclusions: 5-ALA provided 100% specificity and high sensitivity to detect peritoneal metastases in subgroups of patients with peritoneal surface mailgnancies. ALA influx transporter PEPT1 and porphyrin efflux transporter ABCG2 genes are important in tumor specific 5-ALA induced fluorescence in vivo. Further studies should clarify diagnostic utility of 5-ALA in peritoneal surface malignancies.

        Biotransformation of propolis phenols by L. plantarum as a strategy for reduction of allergens

        Propolis is a natural product produced by honeybees. It has antioxidant effects as well as antimicrobial, antiseptic, antibacterial, anti-inflammatory and antimutagenic properties. Except these important healthy properties some cytotoxic effects causing allergies also have been reported. In this study have been evaluated changes of phenolic compounds including allergens molecules found in propolis. Before biotransformation, propolis samples were treated with different solvent (ethanol and polyethylene glycol) to facilitate solvation of solid samples. Biotransformation was done by three different strains of Lactobacillus plantarum (10, 8014, ATCC). Results demonstrated the importance of used solvent/ treatment for extraction procedure and strains of L. plantrum. The lowest values of main allergens were determined as 321 ng/mL for BCAFE, 320 ng/mL for 1.1 DMAECAFE and 8.02 ng/mL for CAPE. The study is the first work deal with evaluation of bioconversion of propolis by different L. plantarum strains and their effects on phenolic profile.

        Investigation of shape memory characteristics and production of HfZrTiFeMnSi high entropy alloy by mechanical alloying method

        High entropy alloy (HEA) with shape memory effect (SME) has been the subject of great interest for the past few decades. However, with the increased demands for new materials for high thermal applications, the research activities on the multi elemental high entropy shape memory alloys (HESMA) have been increased by many folds recently. The nano crystalline HEA powder with shape memory effect developed in this study, HfZrTiFeMnSi, was produced by mechanical alloying (MA) for the first time. In this method equiatomic ratio of Hf, Zr, Ti, Fe, Mn, and Si were mixed together and milled by MA process for 100 h. The powder formed was of amorphous in nature. Elemental mapping of the powder from SEM-EDS revealed homogeneity of the alloying elements confirming successful fabrication of HfZrTiFeMnSi HEA powder. The DSC studies from ambient to 500 ◦C of the annealed alloy powders showed reversible austenitic to martensitic (A↔M) transformations. The A↔M transformation hysteresis seemed to vary with the milling time and annealing temperature. The enthalpy values, ΔH, for the transformation were calculated from the DSC plots using tangent method for peak area calculation. Regardless of the annealing temperature, the thermal analysis revealed that the ΔH, equilibrium temperature (T0), and crystallization temperature values decreased with the increasing milling time.
