      • Glycine tomentella의 callus 에서 新梢再分和

        Callus cultures were established from immature cotyledon of Glycine tomentella. MS medium supplemented with 0.02mg/l NAA plus 2.0mg/l zeatin and MS medium with 2.0mg/l IBA plus 0.13mg/l BAP plus 2.0mg/l GA₃were found suitable for initiation and multiplication of callus cultures. Shoot buds were regenerated when the callus was transferred to MS medium supplement with 0.02mg/l NAA plus 0.2mg/l BAP.

      • Lipoxygenase-3 缺乏 大豆系統의 選拔과 特性

        Carotene bleaching test와 SDS-polyacrylamide 전기영동을 통하여 수집한 대두 유전자원 723계통으로 부터 lipoxygenase-3 동위효소가 결핍된 두 계통(N0. 67. No. 217)을 선발하였다. 비린냄새가 아주 적은 돌연변 선발계통은 생리적 이상이 발견되지 않았으며 지방산조성 및 아미노산조성도 정상계통과 큰 차이가 없었다. 연구결과는 경제적 이용성을 갖는 대두품종의 육성에 보탬이 될 뿐만 아니라 주요작물의 유전자원수집과 보존의 중요성을 말해준다. 723 soybean germplasm collections were screened for genotypes missing lipoxy-genase-3 isozyme(linoleate : oxygen oxidoreductase, E.C. 1.13.11.12). Two lines 'No. 67' and 'No. 217' were found to lack lipoxygenase-3 by both carotene bleaching test and improved SDS-polyacrylamide gel electrophoresis. The mutant seeds showing very low beany flavor germinated, grew, and matured normally. Their food composition was similar to that of the normal type of soybean. These results encourage us to breed soybean cultivars having a great economical value for the utilization of soybean and demonstrate the importance of collecting and maintaining extensive germplasm collections of our major crops.

      • 유도전동기를 제어하기 위한 인버터의 공간벡터 퍼지제어

        A system with fast torque response is very beneficial in applications where direct self control(DSC) is highly desirable. The response of DSC is slower during startup and during change in command torque. Fuzzy control is used for implementation of DSC to improve its slow response. Simulation implementation of the fuzzy logic controller was carried out to verity the behavior of the controller. The simulation results with fuzzy control are compared with those of the conventional DSC. The starting flux and torque response and the responses to the step changes in command torque with fuzzy implementation show a considerable improvement over the conventional control. The steady state responses in both the cases are the same.

        작물의 육종에서 야생 근연종은 다양한 유전자의 공급원으로서 중요하나 그들 유용유전자의 재배종으로의 도입 성패는 상호간에 존재하는 교잡불화합성의 극복에 달려있다. 본 연구에서는 대두와 그의 야생 근연종인 G. tomentella간의 아속간교잡에서 얻어진 발생초기단계의 배 또는 배주를 기내배양을 통하여 발아시키고자 하였다. 구형단계의 초기 잡종배는 0.1mg/1 IBA와 0.25mg/1 2-iP를 첨가한 B5배지에 배양함으로서 발아시킬 수 있었다. 또한 초기잡종배를 가진 배주를 0.1mg/1 IBA를 첨가한 B5배지에 배양하여 성숙시키고 0.1mg/1 IBA와 0.25mg/1 첨가한 B5배지에 계대배양함으로서도 배의 발아를 유도할 수 있었다. 그러나 이들 중 일부의 배양체는 칼루스화 되기도 하였다. Wild relatives of crop plants have been effectively employed in various breeding programs as additional sources of genetic diversity. This paper reports that the rescue of hybrid embryos between the soybean (Glycine max) and a wild perennial relative, G. tomentella, were accomplished through in vitro culture. The young embryos at the globular stage were matured and germinated by culturing on B5 medium supplemented with 0.1mg/1 IBA plus 0.25mg/1 2-iP. Embryos were also germinated by culturing ovules containing young embryos on B5 medium supplemented with 0.1mg/1 IBA for two month followed by culture on B5 medium supplemented with 0.1mg/1 IBA plus 0.25mg/1 2-iP. In some cases, however, the cultures proliferated to callus.

      • Deep Hole Drilling에서 절삭유가 가공성에 미치는 영향에 관한 연구

        This work deals with o investigation of the influence of various additives to a base stock cutting fluid in order to develop a better deep hole drilling. This investigation has been aimed at development of an oil which gives a maximum cutting efficiency and a minimum wear rate of the tool and the guiding pads. The purpose of this study is to analyze how guide pad of tools, workpiece and the change of contained quantity of the extremen pressure additive i cutting fluids have effects on the hole over size of cutting hole, surface foughness of workpiece, wear rates of guide pad and roundness during the deep hole machining of SM55C with solid BTA drill by using BTA drilling system through experiment. Conclusion reached is as follows. 1)It has been proved that contained quantity of sulphyr more affects machinability than that of extreme pressure additive of chlorine in BTA drilling during deep hole drilling. 2)The range of 1.5~2.0% of sulphur quantity is suitable to machinability for workpiece in BTA drilling. 3)The wear of guide pad is affeted by the quantity of extreme pressure additive of chlorine.

      • 동양란의 미세대량증식을 위한 효율적 배지조성

        Experiments are described which determine the optimal nutrient medium for in vitro micropropagation of oriental Cymbidium. Actively growing one year-old rhizomes were cultured for five months in 15 different formulated media, and their responses were investigated. The hyponex(N:P:K=6.5:6:19) basal medium supplemented with 4g tryptone, 1.5g activated charcoal, and growth regulators combinated with 1mg NAA+1mg kinetin or 2mg NAA+0.5∼1mg BA per liter was found to be suitable for rhizome growth and organogenesis. Several media factors affecting in vitro culture of rhizomes were also discussed.

      • 大豆未熟子葉祖織의 營養培地條件에 따른 器官分化

        大豆의 末熟子葉組織을 사용하여 여러가니 營養培地에서 칼루스와 胚狀體로부터 器官을 分化시키고자 하였던 바 얻어진 결과를 要約하면 다음과 같다. 1. 칼루스誘起는 MS 基本培地에 1.0mg/12,4-D,와 2.0mg/1kinetin을 첨가하거나 (M5培地) 하였을 때 가장 효과적이었다. 2. M4培地에 培養한 칼루스로부터 生長點모양의 완전한 구조를 갖춘 細胞群을 組織學的으로 관찰할 수 있었다. 3.칼루스를 器官分化를 위한 培地에 ?代培養하였을때 根分化, 칼루스로부터胚狀體의 形成, 胚狀體로부터 싹이나 잎의 分化는 供試한 大豆系統이나 使用한 營養培地에 따라 약간의 差異는 있으나 비교적 용이할 것으로 여겨졌다. 4. 供試한大豆系統인 뱀콩의 末熟子葉組織을 M4培地에서 칼루스를 誘起시킨 후 MS基本培地에 0.005mg/1 picloram 과 2.0 mg/1 BAP를 첨가한 培地(OM2培地)에 ?代培養하였을 때 칼루스로부터 胚狀體의 形成이 잘 되었고 胚狀體들로 부터 잎이 分化되었다. The experiments were conducted to investigate organ differentatin rom calls and embryoid under various media using immature ctyledon of soybeam. The results obtained were as followings: 1. 1. Omg 2,4-D plus 0.2mg kinetin or 0.2 mg BAP perr litre on MS basal mediu were efective growth regulators for callus induction. 2. Meristematic structure was observed from the callus induced by 1.0mg 2,4-D plus 0.2mg kinetin per litre in MS basal medium in which callus growth showed best response. 3. Root and embryoid from callus and shoot or leaflet from embryoid were differentiated on the medium subcultured for organ different on the soybean lines and media applied. 4. Embryoid from callus and leaflet from the embryoid were differentiated on the nutritional condition subscultured with 0.005mg picloram plus 2.0mg kinetin per litre on MS in immature cotyledon of spybean line Baemkong.

      • 잎파래 (Enteromorpha linza)의 葉體無菌化와 元形質體 培養

        잎파래(Enteromorpha linza) 葉體의 無菌化와 葉體에서 分離한 原形質體의 培養에 關한 몇가지 條件을 決定코자 하였다. 洗劑로 葉體의 附着微生物을 除去하기 爲해서는 0.1%液으로 15∼20分間 洗 하는 것이 效果的이었으며, 組織內部의 微生物은 1% KI-I2液으로 2.5∼5分間 處理하여 抗生劑 2%가 添加된 培地함으로서 除去가 可能하였다. 葉體의 原形質體는 2% AAP가 添加된 各種酵素液으로 容易하게 분리할 수 있었다. 原形質體의 無菌培養을 爲한 抗生物質의 添加濃度는 1.5%程道가 適當하였으며 照度는 2000lux以上일 境遇培養에 큰 影響을 미치지는 않는 傾向이었다. Basic studies for obtaining the axenic tissue, isolation and culture of protoplasts of Enteromorpha linza have been attempted. Treating with 0.1% detergent for 15-20mins were effective for cleaning the collected tissue. Exposure the tissue to a 1% KI-I2 solution for 2.5-5.0 mins subsequently culture them in the medium containing 2.0% antibiotic materials were good for inhibition of bacterial growth as well as enzyme solutions composed of abalone acetone powder, cellulase R-10, and driselase. The 2.0% antibiotic medium was good for the suppression of bacterial growth in protoplasts culture. The development of protoplasts was not largely influenced by the light strength and photoperiods.

        The purpose of this investigation was to develop a new carrier using chitin and glucan as the model for Aspergillus oryzae cell wall. In case of the ratio of chitin and glucan(3/1), the activity of the immobilized protease had higher than that of model(chitin/glucan=1/3) and carriers were formed gel. In case of soluble chitin and glucan treated with calcium carbonate, there were a 1.8-fold increase in activity of immobilized protease compared with untreated chitin. The scanning electron microscope image showed that a pore size of this carrier had a homogenized pore size and a smooth surface.

        Development of Tobacco Mosaic Virus (TMV) resistant Flue-cured $\textrm{F}_1$ Hybrid, KF118

        Tobacco mosaic virus (TMV), TMV-common strain, causes severe damage at growth, yield and leaf quality on flue-cured tobacco in Korea. Controlling TMV outbreak with cultivating practices and chemicals are limited, thus, the best way would be developing TMV resistant variety. A new flue-cured hybrid, KF118, was developed from a cross of maternal parent MSTC175 and TC853 at the Agro-tech. Research Group, KT&G Central Research Institute. KF118 exhibited more vigorous growth characteristics in early stage than that of NC82, standard variety in Korea, while other characters were favorable to those of NC82, It is highly resistant to TMV and bacterial wilt(Ral-stonia solanacerum), and black shank(phytophthora parasitica var. nicotianae) is comparable to that of NC82. It should adapt well to the flue-cured production area, and can reduce premature flowering under unfavorable weather conditions. Yield of KF118 is 1 to 2% higher, and leaf quality is 4% higher than NC82. Fl hybrid, KF118, met acceptable standards for chemical and physical characteristics of cured leaf, and ranked high in good smoking taste by panel members in KT&G.
