In order to compare the metabolic activities of methanol utilizing bacteria, Pseudomonas sp. grown in different carbon sources, changes in respiratory activities, prinicipal enzyme activities for the energy metabolism, and the macromolecular compositions of the cells grown on methanol or glucose were measured. 1. The respiratory activity of cells grown on methanol was higher than that of cells grown on glucose, while glucose exhibited the highest $O_2-consumption$ rate among the different respiratory substrates. 2. TRhe activity of hydroxy pyruvate reductase which participates in serine pathway was high in the cells grown on methanol. However, activities of NAD-linked alcohol dehydrogenase, formaldehyde dehydrogenase and formate dehydrogenase were slightly lower in the cells grown on glucose thant on methanol. 4. For succinic dehydrogenase and malic dehydrogenase which take part in TCA cycle, the specific activities were higher in the cells grown on methanol than in those grown on glucose. No activity of glucose-6-phosphate dehydrogenase, which participates in pentose monophosphate shunt, was detectable in the cells grown on either carbon sources. 5. Protein contents of the cells grown on methanol increased relatively compared with those of the cells grown on glucose. However, there are no changes in the contents of carbohydrate and nucleic acid.
잎파래(Enteromorpha linza)의 原形質體에 對한 紫外線照射의 影響

잎파래(Enteromorpha linza)의 엽체를 材科로 原形質體를 分離하였다. 2%의 abalone acetone powder와 5%의 driselase로 調製된 효소액으로 엽체 1㎠ 당 1.07 X ?? 개의 原形質體를 獲得할 수 있었다. 原形質體는 紫外線을 照射한 후 培養에 따른 反應을 調査하였다. UV로서 이들 原形質體에 對하여 效果的으로 突然變異를 誘起하기 爲해서는 약 5~10분간 照射하는 것이 좋을 것으로 推定된다. Protoplasts were isolated from the thalli of Enteromorpha linza. An enzyme mixture of 2% abalone acetone powder and 5% driselase yielded 1.07 X ?? protoplasts from 1㎠ fresh thalli. Protoplasts were irradiated with UV, and then their responses in culture were investigated. The irradiation time for effective mutagenesis on the protoplasts was estimated about 5~10 mins.

진주시를 중심으로 한 서부경남 일원의 토양, 공장폐수 및 하수등에서 메탄올을 탄소원으로 이용하는 세균 210균주를 분리하여 이중 가장 우수한 메탄올자화능을 가진 균주를 선정하여 배양적 특성에 의거하여 균체생산을 최대로 높일 수 있는 최적 배양조건을 추적하여 다음과 같은 결과를 얻었기에 그 결과를 보고한다. 1. strain No.30-1의 최고균체생산을 위한 최적 pH 및 최적 온도는 각각 pH 8.0 및 30℃이었다. 2. 메탄올 및 (NH_4)_2SO_4의 최적 농도는 각각 2∼3% 및 0.3%이었다. 3. 생장소로서 가해준 Thiamine-HCI의 농도는 100㎍/ℓ이며, 무기이온으로서 Fe^++ 이온 및 Mn^++ 이온의 농도는 각각 2㎎/ℓ이었다. 이와같은 최적배양조건에서 strain No.30-1의 최대균체수율을 위한 배양시간은 72시간 이후에 최대정지기에 도달하였다. 210 strains were isolated from soil, waste water and sewage samples collected mainly in Chinju city and western area of Gyeongsangnam-do by enrichment culture on methanol as the sole source of carbon. Among these microorganisms, strain No.30-1 was exhibited most prominent methanol utilizing activity. Experiments were carried out to determine the optimum growth conditions for the highest biomass yield. The results were as follows; 1. The optimum pH and temperature were seen at pH 8.0 and 30℃. 2. The optimum initial concentration of methanol and that of (NH_4)_2SO_4 as nitrogen source were 2.0 to 3.0% and 0.3%, respectively. 3. The optimum concentration of vitamin and inorganic ions such as Fe^++ ion and Mn^++ ion were 100㎍ Thiamine-HCI, 2㎎ Fe^++ ion and 2㎎ Mn^++ ion, per liter medium. From the data obtained, it was shown that culture time for the highest biomass yield of the strain No.30-1 take place after 72 hrs, and then arrived at the maximum stationary phase.
