        유산균 농도가 돌산갓김치의 항산화효과 및 ACE 저해활성에 미치는 효과

        Starter로 사용하기 위하여 $20^{\circ}C$에서 4일간 발효된 돌산 갓김치에서 미생물을 분리하였다. 이를 starter로 접종하여 $4^{\circ}C$와 $10^{\circ}C$에서 50일간 발효시키면서 발효특성, 항산화활성, ACE 저해활성을 조사하였다. 1 X $10^{10}$ CFU/mL의 접종농도에서 적숙기(pH 4.5)에 도달하는 시간이 $4^{\circ}C$에서는 최고 5.6배 이상, $10^{\circ}C$에서는 5배 단축되었다. 총균수는 starter 접종농도가 높을수록 발효초부터 높은 균수를 유지하였다. 항산화활성도 starter 접종농도가 높을수록 전반적으로 골게 나타났고, 1 X $10^{10}$ CFU/mL의 접종농도에서 $4^{\circ}C$와 $10^{\circ}C$ 각각 최대 67%와 75%를 나타내었다. 따라서 단위 시간당 세포의 생성율($lnX_{max}$/$t_{max}$)과 항산화활성($lnX_{max}$/$t_{max}$)은 비례관계를 가지며, 초기 접종농도가 높을수록 단위 시간 당 항산화활성이 높음을 알 수 있었다. ACE 저해활성도starter의 농도가 높을수록 ACE 저해활성도 놀게 나타났다. 즉, 1 X $10^{10}$ CFU/mL의 접종농도에서 $4^{\circ}C$에서는 52%, $10^{\circ}C$에서는 최대 76%를 나타내었다. 파라서 일정시간당 체포의 생성율($lnX_{max}$/$t_{max}$)이 증가할수록 ACE 저해율 ($P_{max}$/$t_{max}$)도 증가하였으며, 초기 접종농도가 높을수록 단 위 시간당 ACE 저해율이 높았다. The bacterial strain was isolated from the 4th day's fermented Dolsan Leaf Mustard Kimchi(DLMK) at $20^{\circ}C$. It was used as Kimchi starter, and then its physiological activity was investigated for 50 days at $4^{\circ}C$ and $10^{\circ}C$ The physiological activity of DLMK was examined for both antioxidative and Angiotensine Converting Enzyme(ACE) inhibitory activity. In the starter-inoculated DLMK(1 X $10^{10}$ CFU/mL) at 4 and $10^{\circ}C$, the optimal ripening period was more shortend than that of control(without starter) up to about 5.6 and 5 times, respectively. The maximal antioxidative activity in the starter-inoculated DLMK(1 X $10^{10}$ 10 CFU/mL) at 4 and $10^{\circ}C$ were 67% and 75%, respectively. The yield of cell concentration per day($lnX_{max}$/$t_{max}$) and the yield of antioxidative activity per day($P_{max}$/t$t_{max}$) had a linear relationship. Also, the yield of antioxidative activity per day was increased with increasing the concentration of inoculated bacterium. By adding 1 X $10^{10}$ CFU/mL at 4 and $10^{\circ}C$, the ACE inhibitory activity of DLMK was maximal. The rates of inhibiting activities were 52% and 76%, respectively. Consequently, physiological activities were significantly affected by the inoculation concentrations of starter, but bacterium itself was not appeared the physiological activity. We assume that the bacterium metabolizes certain materials in DLMK and released compounds such as glucosinolates or its metabolized forms from DLMK show the antioxidative and ACE inhibitory activity.

        Cytotoxic, Antioxidative, and ACE Inhibiting Activities of Dolsan Leaf Mustard Juice (DLMJ) Treated with Lactic Acid Bacteria

        This study was performed to know whether there is any change of physiological activity in DLMJ which is inoculated by lactic acid bacteria. Lactic acid bacteria were isolated from Dolsan leaf mustard Kimchi (DLMK) at 20oC. In the optimum ripening period, the population of Leuconostoc and Lactobacilli in the DLMK were found to be high. The Leuconostoc, Lactobacilli and Lactococci strains were identified as Leuconostoc mesenteroides, Leuconostoc gelidum, Weissella confusa, Lactobacillus plantarum, Lactobacillus raffinolactis, Lactococcus lactis and Weissella confusa using the Biolog system. The most predominant strain which was isolated from DLMK was Weissella confusa. As the results of the phylogenetic analysis using 16s rDNA sequence, the Weissella confusa turned out to be Weissella kimchii, with 99.0% similarity. To investigated the change of physiological activity in DLMJ by lactic acid bacteria, 7 predominant strains inoculated to DLMJ (Dolsan Leaf Mustard Juice). The cytotoxicity was found to be under 19.55% all cases. Also, the antioxidative activity of the DLMJ treated with lactic acid bacteria was very low, which might have been due to the reduced antioxidative phytochemicals during the preparation of the sterile sample. The ACE inhibiting activity of DLMJ by inoculation with Weissella kimchii was shown to be the highest (94.0%). This could be that the degradation of sulfur containing materials in DLMJ by Weissella kimchii gave rise to ACE inhibiting activity.

        홍어 숙성과 기능성

        홍어 발효에 따른 부위별 생리기능성을 측정하기 위하여 홍어 발효과정의 일반성분의 변화, pH, 암모니움 이온농도, 항균성, 항산화성, 항암성을 측정하였다. 발효 온도(4, 10, $20^{\circ}C$)에 따른 기간별 홍어의 일반성분 분석의 결과 큰 차이를 나타내지 않았다. 홍어 유출액의 pH 변화는 $4^{\circ}C$에서 발효한 경우는 pH의 상승폭이 10, $20^{\circ}C$에 비해 낮았고, 1$0^{\circ}C$에서는 발효 1일만에 급격하게 증가하다가 5일까지 일정하게 유지한 후 2차 증가를 나타내었다. $20^{\circ}C$에서는 발효 초기부저 급격하게 증가하기 시작하여 발효 4일에 8.9를 나타내었다. 홍어 유출액의 암모니움 이온 농도 변화에서도 pH의 경우와 비슷하였는데, $4^{\circ}C$발효 온도에서는 시간별 암모니움 이온 농도 함량에 큰 차이를 나타내지 않았고, $10^{\circ}C$에서 발효시킨 홍어의 경우 5일에, $20^{\circ}C$에서 발효시킨 홍어의 경우 4.5일에 암모니움 이온 농도가 약 $10.2\mug/mL$로 발효 진행 초기인 것으로 나타났다. 홍어 가식부 및 내장 열수 추출물의 발효기간별 생리기능성을 측정한 결과 가식부 열수 추출물의 항균성은 거의 없는 것으로 나타났고, 내장은 발효 8일째에 시료 2%첨가한 군에서 43.3%로 가장 높은 항균효과를 나타내었다. 항산화성은 가식부 열수 추출물 2%첨가한 군에서 발효 0일째에 61.2%로 가장 높았고, 내장 동일 농도에서도 발효 0일째에 54.4%로 비교적 높았다. 항암성은 시료 농도 $1,000 \mug/mL$에서 가식부는 발효 8일에 52.7%였고, 내장은 발효 10일에 58.3%로 가장 높게 나타났다. 따라서 항균성과 항암성은 발효가 진행될수록 비교적 높게 나타났고, 항산화성은 발효시키지 않은 원료 자체에서 높은 효과를 나타내었다. 홍어 뇌 및 연골 열수 추출물의 농도별 생리기능성은 항균성은 뇌 시료 2%첨가한 군에서 균 배양 4시간째에 41.0%였고, 동일 농도에서 연골은 균 배양 14시간째에 35.8%를 나타내었다. 항산화성은 뇌 열수 추출물에서는 효과가 없었고, 연골 시료 2%첨가한 군에서는 25.0%로 가식부나 내장에 비해 낮게 나타났다. This study was conducted to investigate the physiological properties of various parts of skate body after fermentation by measuring compositional properties including pH and $NH_4^+$. Other functional properties, such as antibacterial activities, antioxidative activities and anticancer activities were measured. Effects of fermentation temperature (4, 10, $20^{\circ}C$) did not affect compositional properties of fermented skate. The pH of fermented skate extract at 4$^{\circ}C$ did not increase as much as that at 10 and $20^{\circ}C$, Particularly at $10^{\circ}C$, the pH increased rapidly after Day 1 and remained unchanged until another increase at Day 5. At 2$0^{\circ}C$, the pH increased rather rapidly at early stage of fermentation and reached 8.9 at Day 4. The pattern of $NH_4^+$ concentration of fermented skate extract was similar to that of pH. Particularly at $4^{\circ}C$ fermentation, $NH_4^+$ concentration was not affected by fermentation time. The concentration of $NH_4^+$. reached approximately $10.2\mug/mL$at $10^{\circ}C$ for Day 5 and $20^{\circ}C$ for Day 4-5, indicating the early stage of fermentation. According to physiological activities of hot water extracts of skate fillet and viscera as affected by fermentation time, antibacterial activity of 2% viscera extract concentration was 43.3% at Day 8, while there was no antibacterial activity from fillet extract. As for the antioxidative activity, fillet extract and viscera extract both at 2% concentration at Day 0 showed 61.2% and 54.4%, respectively. Anticancer activities were highest (52.7% for fillet extract and 58.3% for viscera extract) at $1,000 \mug/mL$ concentration at Day 8. Antibacterial activities and anticancer effects were relatively high as fermentation was progressed, while antioxidative activities were highest before fermentation started. As for the physiological activities of hot water extract from brain and cartilage, antibacterial activities were observed at 41.0% when 2% brain extract was added at 4 hours of incubation, while 35.8% with 2% cartilage extract at 14 hours of incubation. Antioxidative activities were not found in brain extract, but cartilage extract at 2% showed 25.0% of antioxidative activity, which was lower than fillet and viscera extract.

      • Aspergillus awamori의 효율적인 protoplast 생성을 위한 homogenization효과

        The effect of homogenization for efficient protoplasts formation from Aspergillus awamori was studied. After homogenization for 5 min, a high yield of protoplasts was obtained. Prolonged treatment beyond 15 min decreased the yield of protoplasts. Protoplasts prepared from enzyme treatment after homogenization showed an increase in protoplast yields as compared with the protoplast yields prepared from the only lytic enzyme treatment.The present study shows that high yields of protoplasts can be prepared from Asp. awamori using homogenization before enzyme treatment.

      • Aspergillus oryzae 酸性 proteinase의 再固定化

        The present study is an attempt to investigate the immobilized carrier of Aspergilius oryzae cell wall. Optimum conditions for immobilization of Triton X-100 soluble acid proteinase on fractionated cell wall were pH 3.5 and 35℃. The activity of the immobilized enzyme(IME) had similar to or more than that of NATIVE cell wall fraction-bound acid proteinase(CWE). Crosslinking with glutaraldehyde on a adsorbed layer increased the immobilization of acid proteinase. Optimum pH of IME was 2.3. Optimum temperature, the stability of pH and temperature in IME were similar to those of native CWE. Considering bioreactor, the characterization of IME may prevent microbial contamination under acid pH condition during processing.

        방사형 집수정 취수시 대수층 특성에 따른 지하수위의 변화

        The groundwater drawdown at each monitoring well near a radial collector well along Anseong Stream, Korea, was measured and compared with the calculated drawdown using the mirror well concept. The drawdown calculation is performed by treating the collector well as a large vertical well in a homogeneous isotropic aquifer. The measured drawdown at each monitoring well is slightly different from the calculated value due to anisotropy in the hydraulic conductivity and aquifer thickness. The difference between the measured and calculated values at Well OW-7 is large, reaching approximately 48 cm, because a horizontal well is not installed along this direction. Sensitivity analysis of the hydraulic conductivity and aquifer thickness indicates that the hydraulic conductivity is more sensitive to groundwater level changes. Groundwater level changes become a concern when a radial collector well with a large pumping rate capacity is installed, which highlights the need to thoroughly investigate the aquifer characteristics in the surrounding area. 본 연구는 안성천에 설치된 방사형집수정에서 약 4,000 m3/d의 취수시 주변지역에서의 지하수위 강하량을 측정하였으며, mirror well 개념을 적용하여 계산된 지하수위 강하량과 비교 분석하였다. 지하수위강하량의 계산은 방사형집수정을 균질 등방성 매질내의 대형 수직정으로 고려하여 실시하였다. 양수시각 관측정에서 측정된 지하수위 강하량은 계산된 값과 약간의 오차가 발생하는데, 이는 수리전도도 및대수층 두께 등의 이방성 때문으로 보인다. 또한, OW-7 지점은 실측 및 계산 지하수위 강하량의 차이가약 48 cm로 크게 나타났는데, 이는 이 방향으로의 수평집수관이 설치되지 않았기 때문이다. 연구지역내에서 출현 가능한 수리전도도와 대수층 두께를 고려한 지하수위 변화의 민감도를 분석한 결과, 대수층의 두께보다는 수리전도도가 지하수위 변화에 민감하게 작용하는 것으로 나타났다. 대용량 취수 시설인방사형 집수정의 개발시에는 배후지 수위 강하에 대한 우려가 존재하게 되므로 주변지역에 대한 대수층특성을 정밀하게 파악하여 지하수위 변화를 예측, 평가할 필요가 있다.
