        Stimulation of angiogenesis and survival of endothelial cells by human monoclonal Tie2 receptor antibody

        <P><B>Abstract</B></P> <P>Angiopoietin-1 (Ang1) and its endothelium-specific receptor, tyrosine kinase with Ig and epidermal growth factor homology domain 2 (Tie2), play critical roles in vascular development. Although the Ang1/Tie2 system has been considered a promising target for therapeutic neovascularization, several imitations of large-scale production have hampered the development of recombinant Ang1 for therapeutics. In this study, we produced a fully human agonistic antibody against Tie2, designated 1–4h, and tested the applicability of 1–4h as an alternative to native Ang1 in therapeutic angiogenesis. 1–4h significantly enhanced the phosphorylation of Tie2 in a dose- and time-dependent manner in human Tie2-expressing HEK293 cells and human umbilical vein endothelial cells. Moreover, 1–4h induced the activation of Tie2-mediated intracellular signaling such as AKT, eNOS, MAPK, and Focal Adhesion Kinase p125<SUP>FAK</SUP>. In addition, 1–4h increased the chemotactic motility and capillary-like tube formation of endothelial cells <I>in vitro</I> and enhanced the survival of serum-deprived endothelial cells. Taken together, our data clearly suggest that a human Tie2 agonistic antibody is a potentially useful therapeutic approach for the treatment of several ischemic diseases including delayed-wound healing and ischemic heart and limb diseases.</P>

        『스페인의 대지』와 『누구를 위하여 종은 울리나』에 나타난 헤밍웨이의 인본주의적 개인주의 연구

        This study examined Hemingway’s personal letters, articles, and narrations of documentaries filmed in Spain, focusing on selected works written during the Spanish Civil War. Hemingway had begun formulating his ideology about man’s responsibility to others, which asserted his unequivocal belief in basic inherent human rights, an ideology which may be termed his humanitarian individualism. Although elements of his humanitarian individualism may be identified in the majority of his works, it was during this bloody civil war, in which thousands of civilians were massacred by the Fascists, that he solidly articulated and demonstrated his humanitarian ideals. Written just after To Have and Have Not, Hemingway’s For Whom the Bell Tolls appears to have changed his focus from the individual to the community. This study explored Hemingway’s emotional journey into the heart of man and carefully observed his beliefs of humanitarian individualism, which advocated freedom, liberty, and justice for all people everywhere.

      • IT 버블과 녹색산업 성공전략

        The Korean government has pushed the very ambitious “green growth” strategy which has generates many government initiatives. There has been speculation that these government drives may trigger a new “green” bubble economy like the recent dot.com bubble. This research examines whether this concern may be realized or not. In order to investigate, first we characterize roles and impact of bubble. Only negative impacts of bubble economy have been frequently cited as we see in the global finance crisis which is believed to have been triggered by bubble in real estate market. The bubble burst of dotcom fever is another example. However, there are many positive roles that bubble economy brings. History shows that there are radical industrial structural changes with new technologies after bubbles. For instance, IT, especially, Web technology triggered dotcom bubble, which resulted in e-business era and broad impacts in business and our lives. Of course there have been bubbles without such innovative technologies such as the well-known tulip bubble in Netherlands. When bubble precedes industrial revolution, it supplies capital to technological innovation and gathers consumers’ attention. That will make market grow faster. This paper summarize characteristics of the so-called “momentum technology” that triggers bubble but eventually industrial revolution. They are (ⅰ) accelerating speed of production and diffusion of technology, (ⅱ) economies of scale, and (ⅲ) continuous innovation. Through these characteristics, consumers can enjoy lowering cost of technology. However, emerging green technologies do not exhibit such characteristics. Hence, there will be no “natural” momentum technology. On the other hand, to create a bubble economy, there must be a market feedback mechanism in capital market that provides market signal of price inflation so that more investors enter the market. However, currently there is no such market for green technology. While new “green technologies” do not have economical advantage or expectation to achieve that in near future, new regulatory compliance or government policies to curb CO₂ may reduce gap in cost or even reverse the price disadvantage against the incumbent technology. However, the economic gap seems too big for such possibilities. Another possibility of bubble creation is massive investment by government or incentives toward that direction. In late 90’s Korean government policy to promote new tech startups is a good example. We review political and socio-economic factors and environment that enabled such aggressive government policies. We show that there is no analogy to green technology. Korean government could pursue IT initiatives with already widely accepted belief on IT as the infrastructure of new economy. However, there is no such consensus on green technology yet. In addition, Korean government could promote new startups (dotcom’s) as a solution to the foreign currency crisis Korean economy had in the mid-90’s. It was considered as an alternative to Jaebeol oriented economy which was attributed as one of causes of the crisis. However, there is no such enabling factors to government massive drives toward green technology investment. We conclude that green technology and the investment for that will be very slow evolution rather than revolution.

      • 수분노화를 통한 BKNO₃의 열역학적 및 속도론적 노화 분석

        화약 산업에서 화약은 높은 폭발 압력을 가지고 있는 것뿐만 아니라 장기간 보관하였을 때 같은 폭발력을 유지하여야 한다. 하지만 화약이 노화되어 폭발력에 변화가 생기기 때문에 노화 매커니즘을 알아야 한다. BKNO₃화약을 이용하여 자발적인 노화를 확인하기 위해 DSC 분석 및 AKTS 시뮬레이션을 하였다. BKNO₃의 바인더가 가장 먼저 자발적 발열반응을 하지만, AKTS 시뮬레이션 결과 고온 조건에서 일어 나지 않는 결과를 얻을 수 있었다. 외부 요인인 수분에 의해 만들어진 수분노화된 화약을 통하여 분석해 본 결과 열량과 반응속도가 감소함을 확인하였다. XPS로 분석한 결과 B-O 산화물 peak이 발견되었고 TEM과 EELS로 Boron 표면에 Boron oxide가 생긴다는 것을 확인 할 수 있었다. 생성된 Boron oxide는 화약의 에너지 포텐셜을 낮추고 연소가 시작될 때 금속과 산화물의 표면적을 감소시켜 반응속도를 낮춰 화약의 낮은 peak 압력을 야기 한다. Prediction of mechanism and long-term aging about BKNO₃, which is a core initiator in pyrotechnic mechanical device(PMD) has been estimated using Thermogravimetric analysis(TGA), differential scanning calorimetry(DSC) and a commercial software (Advanced Kinetics and Technology Solutions(AKTS)). BKNO₃ that is different with ZPP and THPP includes Laminac used in a polymer binder. In a result of DSC, We found that exothermic reaction of decomposition about Laminac has happened prior to that of oxidation associated with explosion. the exothermic reaction has been delayed even at the high temperature (~120 ℃). To confirm the effect of external facors such as humidity, The accelerated aging BKNO₃ at 71 ℃ in 50% relative humidity was characterized. The relative heat of combustion and reaction rate decrease gradually by aging time. The XPS analysis characterizes the oxidization peak at 191 eV regarding oxidization of boron, and the TEM-EDS analysis prove the formation of oxide shells. The thicknesses of oxide shells increase by 39 nm in sixteen weeks, which reduce the potential energy and reaction rate on BKNO₃..

      • 열역학적 및 속도론적 분석을 통한 THPP의 노화 안정성 확인

        화약 기반 소자 (PMD)에서 장기간 보관을 하게 되면 소자 안에 있는 화약이 노화되어 폭발력에 변화가 생기기 때문에, 일정 기간이 지나면 폐기 처분을 하게 된다. 그렇기 때문에 PMD 안에 사용하는 화약은 자발적 혹은 외부적 요인에 대하여 화학적 및 물리적으로 안정해야 한다. 기존에 사용되는 화약으로서 BKNO3과 THPP를 대표적으로 이용하기 때문에, 이 화약들을 기반으로 하여 열역학적 및 속도론적 분석을 실시하였다. Differential scanning calorimeter (DSC)를 이용하여 발열량과 반응속도를 분석하였는데, 그 결과 THPP에서는 열량 차이 및 반응속도에 큰 변화가 보이지 않았다. 추가적으로, 노화에 직접적으로 연관되는 산화막 형성을 확인하기 위하여 XPS 및 TEM-EDS 분석하였는데, 열적 분석 결과와 상응하는 결과로서 산화막이 관측 되지 않았다. 이는 THPP가 장기 안정성 측면에서 가장 유명한 화약이라고 판단 할 수 있다. When stored for long periods in a powder-based device (PMD), the explosive power in the device is aged and the explosive power is changed. Thus, The gunpowder used in the PMD must be chemically and physically stable for both internal and external factors. Since BKNO3 and THPP are used as representative gunpowder, thermodynamic and kinetic analyzes were performed based on these gunpowders. Differential scanning calorimeter (DSC) was used to analyze the calorific value and reaction rate. As a result, there was no significant change in caloric value and reaction rate in THPP. In addition, XPS and TEM-EDS analyzes were performed to confirm the formation of oxide films directly related to aging, and no oxide films were observed as a result of thermal analysis. It can be concluded that THPP is the most famous gunpowder in terms of long-term stability.

        KLHL3 deficiency in mice ameliorates obesity, insulin resistance, and nonalcoholic fatty liver disease by regulating energy expenditure

        Obesity is a growing global epidemic that can cause serious adverse health consequences, including insulin resistance (IR) and nonalcoholic fatty liver disease (NAFLD). Obesity development can be attributed to energy imbalance and metabolic inflexibility. Here, we demonstrated that lack of Kelch-like protein 3 (KLHL3) mitigated the development of obesity, IR, and NAFLD by increasing energy expenditure. KLHL3 mutations in humans cause Gordon’s hypertension syndrome; however, the role of KLHL3 in obesity was previously unknown. We examined differences in obesity-related parameters between control and Klhl3−/− mice. A significant decrease in body weight concomitant with fat mass loss and improved IR and NAFLD were observed in Klhl3−/− mice fed a high-fat (HF) diet and aged. KLHL3 deficiency inhibited obesity, IR, and NAFLD by increasing energy expenditure with augmentation of O2 consumption and CO2 production. Delivering dominant-negative (DN) Klhl3 using adeno-associated virus into mice, thereby dominantly expressing DN-KLHL3 in the liver, ameliorated diet-induced obesity, IR, and NAFLD. Finally, adenoviral overexpression of DN-KLHL3, but not wild-type KLHL3, in hepatocytes revealed an energetic phenotype with an increase in the oxygen consumption rate. The present findings demonstrate a novel function of KLHL3 mutation in extrarenal tissues, such as the liver, and may provide a therapeutic target against obesity and obesity-related diseases.
