<i>Tpl2</i> Kinase Impacts Tumor Growth and Metastasis of Clear Cell Renal Cell Carcinoma

<P>Due to the innate high metastatic ability of renal cell carcinoma (RCC), many patients with RCC experience local or systemic relapses after surgical resection. A deeper understanding of the molecular pathogenesis underlying advanced RCC is essential for novel innovative therapeutics. <I>Tumor progression locus 2</I> (<I>Tpl2</I>), upregulated in various tumor types, has been reported to be associated with oncogenesis and metastatic progression via activation of the MAPK signaling pathway. Herein, the relevance of <I>Tpl2</I> in tumor growth and metastasis of RCC is explored. Inspection of The Cancer Genome Atlas (TCGA) indicated that Tpl2 overexpression was significantly related to the presence of metastases and poor outcome in clear cell RCC (ccRCC), which is the most aggressive subtype of RCC. Moreover, expression of Tpl2 and CXCR4 showed a positive correlation in ccRCC patients. Depletion of Tpl2 by RNAi or activity by a <I>Tpl2</I> kinase inhibitor in human ccRCC cells remarkably suppressed MAPK pathways and impaired <I>in vitro</I> cell proliferation, clonogenicity, anoikis resistance, migration, and invasion capabilities. Similarly, orthotopic xenograft growth and lung metastasis were significantly inhibited by <I>Tpl2</I> silencing. Furthermore, Tpl2 knockdown reduced CXCL12-directed chemotaxis and chemoinvasion accompanied with impaired downstream signaling, indicating potential involvement of Tpl2 in CXCR4-mediated metastasis. Taken together, these data indicate that Tpl2 kinase is associated with and contributes to disease progression of ccRCC.</P><P><B>Implications:</B> <I>Tpl2</I> kinase activity has prognostic and therapeutic targeting potential in aggressive clear cell renal cell carcinoma. <I>Mol Cancer Res; 11(11); 1375–86. ©2013 AACR</I>.</P>

Contents of phenolic compound including isoflavones and phenolic acids as well as antioxidant effects in high-protein soybean cultivar "Saedanbaek" were evaluated under different roasting conditions. The roasted soybean exhibited significantly higher antioxidant activity than unroasted soybean in the three antioxidant methods including 2,2-diphenyl-1-picrylhydrazyl, 2,2-azino-bis(3-ethylbenzthiazoline-6-sulphonic acid), and Ferric reducing antioxidant power. In particular, the roasted soybean at $200^{\circ}C$ for 15 min showed the highest antioxidant activity in comparison with other conditions. The contents of phenolic compounds, isoflavone aglycones (genistein, daidzein, and glycitein), isoflavone ${\beta}$-glucosides (genistin, daidzin, and glycitin), and phenolic acids increased, whereas isoflavone malonyl-${\beta}$-glucosides decreased during roasting process. Moreover, total phenolic and flavonoid contents as well as those of isoflavone aglycones, isoflavone-${\beta}$-glucosides, and phenolic acids increased, leading to a general increase in antioxidant activity after roasting. These results suggest that the roasting soybean extracts could contribute to obtaining natural antioxidants in certain food applications.
Analysis of Aroma Compound Profiles in Doenjang (Soy Paste) Made from Twelve Different Soybeans.

Fermented soybean food is one of most economical and health food due to its valuable nutritional and medicinal attributes and have been consumed for centuries as flavoring ingredient in Korea. On fermented soybean food such as doenjang, aroma compounds are important property because they determine taste and grade of fermentation. This study investigated variety of aroma compounds of doenjang made from different soybean genotypes. Aroma compounds in twelve different doenjang made from two cultivar (Daewon and Taekwang) and ten elite lines were extracted by steam distillation extraction (SDE) method and analyzed by gas chromatography/mass spectrometry (GC/MS). Aroma compound were detected over 80 kinds in eight samples (Daewon, Taekwang, MY177, MY187, MY189, MY192, MY204 and MY205) and under 70 kinds in four samples (yeonchun1, MY188, MY203 and MY206). Among the detected aroma components, 47 compounds were assigned as aromatic compounds (21), long chain fatty acid (13), short chain fatty acid (5) and others (8) by the computer library (Wiley 7n program). The major aroma compounds in twelve different doenjang were benzaldehyde, benzeneacetaldehyde, 2-methoxy-4-vinylphenol, pyrazine type compounds, cysteaminesulfonic acid, palmitic acid, oleic acid and linoleic acid. Each doenjang made by same condition (microbe, temperature, aging time and etc) had a difference in ratio and variety of aroma compounds due to different components having soybean genotype.

Eight triterpenoids, six lanostanes 1-6, one lupenane 7, and one oleanane 8, were isolated by bioactivity-guided fractionation of the ethylacetate extract from roots of Glycine max (L.) Merr. All isolated compounds were examined for their inhibitory activities against human ACAT-1 (hACAT-1) and human ACAT-2 (hACAT-2). Among them, three triterpenoids showed potent hACAT inhibitory activities, (24R)-ethylcholest-5-ene-3,7-diol (1) and 3b -hydroxylup-20(29)-en-28-oic acid (7) exhibited more potent inhibitory activity against hACAT-1 (1: IC50 = 25.0 1.2 and 7: IC50 = 11.5 0.4 m M) than hACAT-2 (1: IC50 = 102.0 5.4 and 7: IC50 = 33.9 3.7 m M), respectively. Interestingly, 5a ,8a -epidioxy-24(R)-methylcholesta-6,22-diene-3b -ol (4) has proven to be a specific inhibitor against hACAT-1 (IC50 = 38.7 0.8 m M) compared to hACAT-2 (IC50 >200). In conclusion, this is the first study to demonstrate that triterpenoids of G. max have potent inhibitory activities against hACAT-1 and hACAT-2.

<P>Landfill gas (LFG), which is primarily composed of CH<SUB>4</SUB>, CO<SUB>2</SUB>, and N<SUB>2</SUB>, is produced from theanaerobicdigestion of organic materials. To investigate the feasibility ofthe storage and transportation of LFG via the formation of hydrate,we observed the phase equilibrium behavior of CO<SUB>2</SUB>–CH<SUB>4</SUB>–N<SUB>2</SUB> mixture hydrates. When the specificmolar ratio of CO<SUB>2</SUB>/CH<SUB>4</SUB> was 40/55, the equilibriumdissociation pressures were gradually shifted to higher pressuresand lower temperatures as the mole fraction of N<SUB>2</SUB> increased.X-ray diffraction revealed that the CO<SUB>2</SUB>–CH<SUB>4</SUB>–N<SUB>2</SUB> mixture hydrate prepared from the CO<SUB>2</SUB>/CH<SUB>4</SUB>/N<SUB>2</SUB> (40/55/5) gas mixture formed a structureI clathrate hydrate. A combination of Raman and solid-state <SUP>13</SUP>C NMR measurements provided detailed information regarding the cageoccupancy of gas molecules trapped in the hydrate frameworks. Low-mass-ion discriminant equation: A new concept for colorectal cancer screening

<P>Blood metabolites can be detected as low-mass ions (LMIs) by mass spectrometry (MS). These LMIs may reflect the pathological changes in metabolism that occur as part of a disease state, such as cancer. We constructed a LMI discriminant equation (LOME) to investigate whether systematic LMI profiling might be applied to cancer screening. LMI information including <I>m</I>/<I>z</I> and mass peak intensity was obtained by five independent MALDI-MS analyses, using 1,127 sera collected from healthy individuals and cancer patients with colorectal cancer (CRC), breast cancer (BRC), gastric cancer (GC) and other types of cancer. Using a two-stage principal component analysis to determine weighting factors for individual LMIs and a two-stage LMI selection procedure, we selected a total of 104 and 23 major LMIs by the LOME algorithms for separating CRC from control and rest of cancer samples, respectively. CRC LOME demonstrated excellent discriminating power in a validation set (sensitivity/specificity: 93.21%/96.47%). Furthermore, in a fecal occult blood test (FOBT) of available validation samples, the discriminating power of CRC LOME was much stronger (sensitivity/specificity: 94.79%/97.96%) than that of the FOBT (sensitivity/specificity: 50.00%/100.0%), which is the standard CRC screening tool. The robust discriminating power of the LOME scheme was reconfirmed in screens for BRC (sensitivity/specificity: 92.45%/96.57%) and GC (sensitivity/specificity: 93.18%/98.85%). Our study demonstrates that LOMEs might be powerful noninvasive diagnostic tools with high sensitivity/specificity in cancer screening. The use of LOMEs could potentially enable screening for multiple diseases (including different types of cancer) from a single sampling of LMI information.</P>

국문초록본 연구에서는 2011년 우면산 토석류 피해지역을 대상으로 지상 LiDAR (Light Detection And Ranging) 측량을 통해 토석류 발생 후의 지형의 변화를 관측하였다. LiDAR 측량을 위해서 고해상도 디지털 카메라와 GPS가 장착된 RIEGLE LMS-Z390i모델의 지상스캔닝시스템이 이용되었다. 최상류지점에서의 하상경사는 15-25°의 범위를 보이며 발생지점에서 500m정도까지의 거리에서는 경사도가 10°이상으로서 비교적 큰 경사를 보였다. 500m이후에는 경사도가 감소하여 0-10°의 범위를 보이며 특히 1,000m이후에는 5°보다 낮게 유지되었다. 침식과 퇴적을 고려하는 수치해석 모의를 실시하였다. 수치모의는 Takahashi의 침식/퇴적속도식을 적용한 유한차분법을 이용하였다. 계산결과에서 500m이내 대부분의 지역에서는 1.5m깊이로 침식이 발생하며 500-1,000m구간에서 침식과 퇴적이 반복적으로 나타나고 있으며 1000m이후 구역에서는 퇴적이 우세하게 나타나는 것을 보였다. 수치모의 결과는 현장에서의 침식과 퇴적의 경향을 잘 표현하고 있다고 판단되며 이후 재해위험지도 등에 활용가능하다고 생각된다.
