        Wastewater Utilization: A Place for Managed Wetlands - Review -

        Constructed wetlands are being used for the removal of nutrients from livestock wastewater. However, natural vegetation typically used in constructed wetlands does not have marketable value. As an alternative, agronomic plants grown under flooded or saturated soil conditions that promote denitrification can be used. Studies on constructed wetlands for swine wastewater were conducted in wetland cells that contained either natural wetland plants or a combination of soybeans and rice for two years with the objective of maximum nitrogen reduction to minimize the amount of land required for terminal treatment. Three systems, of two 3.6 by 33.5 m wetland cells connected in series were used; two systems each contained a different combination of emergent wetland vegetation: rush/bulrush (system 1) and bur-reed/cattail (system 2). The third system contained soybean (Glycine max) in saturated-soil-culture (SSC) in the first cell, and flooded rice (Oryza sativa) in the second cell. Nitrogen (N) loading rates of 3 and $10kg\;ha^{-1}\;day^{-1}$ were used in the first and second years, respectively. These loading rates were obtained by mixing swine lagoon liquid with fresh water before it was applied to the wetland. The nutrient removal efficiency was similar in the rush/bulrush, bur-reed/cattails and agronomic plant systems. Mean mass removal of N was 94 % at the loading rate of $3kg\;N\;ha^{-1}\;day^{-1}$ and decreased to 71% at the higher rate of $10kg\;N\;ha^{-1}\;day^{-1}$. The two years means for above-ground dry matter production for rush/bulrushes and bur-reed/cattails was l2 and $33Mg\;ha^{-1}$, respectively. Flooded rice yield was $4.5Mg\;ha^{-1}$ and soybean grown in saturation culture yielded $2.8Mg\;ha^{-1}$. Additionally, the performance of seven soybean cultivars using SSC in constructed wetlands with swine wastewater as the water source was evaluated for two years, The cultivar Young had the highest yield with 4.0 and $2.8Mg\;ha^{-1}$ in each year, This indicated that production of acceptable soybean yields in constructed wetlands seems feasible with SSC using swine lagoon liquid. Two microcosms studies were established to further investigate the management of constructed wetlands. In the first microcosm experiment, the effects of swine lagoon liquid on the growth of wetland plants at half (about 175 mg/l ammonia) and full strength (about 350 mg/l ammonia) was investigated. It was concluded that wetland plants can grow well in at least half strength lagoon liquid. In the second microcosm experiment, sequencing nitrification-wetland treatments was studied. When nitrified lagoon liquid was added in batch applications ($48kg\;N\;ha^{-1}\;day^{-1}$) to wetland microcosms the nitrogen removal rate was four to five times higher than when non-nitrified lagoon liquid was added. Wetland microcosms with plants were more effective than those with bare soil. These results suggest that vegetated wetlands with nitrification pretreatment are viable treatment systems for removal of large quantities of nitrogen from swine lagoon liquid.

        Neutralization of Human Papillomavirus by Specific Nanobodies Against Major Capsid Protein L1

        The human papillomavirus (HPV) is the main cause of cervical cancer in developing countries. Rapid diagnosis and initiation of treatment of the HPV infection are critical. Various methods have been employed to reduce the immunogenicity of antibodies targeting HPV serotypes. Nanobodies are the smallest fragments of naturally occurring single-domain antibodies with their antigenbinding site compromised into a single domain. Nanobodies have remarkable properties such as high stability, solubility, and high homology to the human VH3 domain. In this study, a phagemid library was employed to enrich for nanobodies against the L1 protein of the human papilloma virus. Binding reactivity of the selected clones was evaluated using phage enzyme-linked immunosorbent assay (phage-ELISA). Finally, two nanobodies (sm5 and sm8) with the best reactivity against the Gardasil vaccine and the purified HPV-16 L1 protein were expressed and purified using a Ni+-NTA column. The accuracy of expression and purification of the nanobodies was confirmed by sodium dodecyl sulfate polyacrylamide gel electrophoresis (SDS-PAGE) and immunoblotting assays. In vitro studies demonstrated that neutralization was achieved by the selected nanobodies. The ease of generation and unique features of these molecules make nanobodies promising molecules for the new generation of HPV diagnosis and therapy.

        Animal Welfare in Different Human Cultures, Traditions and Religious Faiths

        Animal welfare has become a growing concern affecting acceptability of agricultural systems in many countries around the world. An earlier Judeo-Christian interpretation of the Bible (1982) that dominion over animals meant that any degree of exploitation was acceptable has changed for most people to mean that each person has responsibility for animal welfare. This view was evident in some ancient Greek writings and has parallels in Islamic teaching. A minority view of Christians, which is a widespread view of Jains, Buddhists and many Hindus, is that animals should not be used by humans as food or for other purposes. The commonest philosophical positions now, concerning how animals should be treated, are a blend of deontological and utilitarian approaches. Most people think that extremes of poor welfare in animals are unacceptable and that those who keep animals should strive for good welfare. Hence animal welfare science, which allows the evaluation of welfare, has developed rapidly.

        Biology of Porphyra pulchella sp. nov. from Australia and New Zealand

        Porphyra pulchella sp. nov. Ackland, West, Scott and Zuccarello was obtained at Mimosa Rock National Park, New South Wales; Westgate Bridge, Victoria, Australia; and Waihau Bay, North Island, New Zealand. It occurs mainly in mangrove habitats and is very small (± 1 mm) in field collections. In laboratory culture at 21 ± 2°C tiny blades (0.5-3.0 mm) reproduced exclusively by archeospores liberated from vegetative cells of the upper sector of the blades. The archeospores displayed amoeboid and gliding motility once discharged. At 14 ± 2°C the blades grew to 25 mm and produced longitudinal spermatangial streaks mixed with ‘phyllosporangial’ streaks. The discharged ‘phyllospores’ showed amoeboid motility and germinated forming asexual blades. A conchocelis phase with typical bangiophycidean pit connections was observed in blade cultures after 8-10 weeks at 14 ± 2°C. Conchocelis filaments produced conchosporangia and these released amoeboid conchospores that developed into archeosporangiate blades. Molecular data indicate that all 3 isolates are genetically identical.
