Background: Progressive muscle weakness is aggravated not only in the skeletal muscles but also in the respiratory muscles in many patients with neuromuscular diseases (NMD). Inspiratory muscle training (IMT) has been reported as therapy for pulmonary rehabilitation to improve respiratory strength, endurance, exercise capacity, and quality of life, and to reduce dyspnea. Objects: The purpose of this study was to determine the effect of playing harmonica for 5 months on pulmonary function by assessing the force vital capacity (FVC), peak cough flow (PCF), maximal inspiratory pressure (MIP), maximal expiratory pressure (MEP), and maximal voluntary ventilation (MVV) in patients with NMD. Methods: Six subjects with NMD participated in this study. The subjects played harmonica once a week for 2 hours at a harmonica academy and twice a week for 1 hour at home. Thus, training was performed thrice a week for 23 weeks. The examiner assessed pulmonary function by measuring FVC in the sitting and supine positions and PCF, MIP, MEP, and MVV in the sitting position at the beginning of training and once a month for 5 months. Results: Both sitting and supine FVC significantly increased after playing harmonica (p=.042), as did MIP (p=.043) and MEP (p=.042). Conclusion: Playing harmonica can be used as an effective method to improve pulmonary function in patients with NMD.

Aujeszky's disease (AD), also called pseudorabies, is an infectious viral disease caused by an alpha herpes virus and has domestic and wild pigs, as well as a wide range of domestic and wild animals, as the natural host. Aujeszky's disease virus (ADV) virions contain several envelope glycoproteins. Among them, glycoproteins gB, gC and gD are regarded as the major immunogenicity proteins and the antibodies induced by them can neutralize virus in vitro or in vivo. In this study, we investigated expression of these glycoproteins using the bacterial and baculovirus expressionn system. Successful expression of ADV glycoproteins in E. coli was confirmed by SDS-PAGE and Western blot analysis and their optimal expression condition was determined. However, the recombinant proteins generated in the bacterial expression system which lacks glycosylation process frequently lose their biological activity. We tried to express the ADV glycoproteins using the baculovirus expression vector system. The recombinant gB, gC and gD were detected at approximately 100, 60 and 50 kDa on SDS-PAGE and Western blotting, respectively. The optimal expression conditions were determined for MOI(multiplicity of infection) and post-infection days. One MOI and 4 or 5 days post-infection were the best conditions for the expression of the ADV glycoproteins in Sf21 cells. We are currently investigating the antigenicity of recombinant proteins using experimental animals.

Objective Nephrin is one of the slit membrane proteins of podocytes in the kidney. It is known that the nephrin is shed in the urine in nephropathy accompanying proteinuira. So the aim of this study was to evaluate the difference in the serum nephrin expression between normal and preeclamptic pregnancies. Methods A total of 20 pregnant women from May to September 2008 who received prenatal care and underwent delivery at our institute participated in the study. The preeclamptic group includes 13 women diagnosed as preeclampsia and a normal group of 7. Their serum were collected before delivery and analyzed by Western blotting for comparing serum nephrin expression. Results There was no difference in age, body weight of pregnant women, blood urea nitrogen, serum creatinine, urine creatinine level and gestational age between two groups. However, preeclampsia group had significantly higher systolic and diastolic blood pressure ( P<0.001), serum soluble fms-like tyrosine kinase-1 level ( P= 0.002), and lower birth weight ( P=0.011). In serum Western blot analysis, serum nephrin was detected in 10 of 13 in preeclampsia women (76.9%) but only in 2 of 7 (28.6%) in normal pregnancy women showing statistically significant difference ( P=0.032). Conclusion A higher prevalence of nephrin expression in the maternal serum was found in the preeclampsia when compared to the normal pregnancy.

Expression of glycoproteins gC and gD were observed on SDS-PAGE or Western blot analysis, but gB was not. Optimal concentration of IPTG and inducing time were determined as 1.0 mM and 4 h, respectively, for the expression of both gC and gD in E. coli. A sodium dodecyl sulfate (SDS) was the most efficient detergent in solubilizing insoluble recombinant protein.

Objectives In the past, studies have been shown that Forsythiae Fructus (FF), Ulmi Cortex (UC) and JwaGwiEum (JGE) are effective in treating allergic reactions. However, no report shows the difference in effects of FF, UC and JGE in allergic rhinitis (AR). Therefore, the purpose of this study was to differentiate the effects of FF, UC and JGE in AR induced by ovalbumin (OVA) in mice. Methods From this experiment, the effects of FF, UC and JGE were several - changes in body weight, hematologic changes such as WBC, RBC, Hb and PLT counts, immunological changes such as levels of histamine, IgE, IL-4 and expressions of iNOS & COX-2 mRNA. Moreover, histological change of nasal mucosa was also investigated. Results FF administration group and JGE administration group significantly inhibited level of IgE compared to the control group. Also, FF administration group, UC administration group and JGE administration group inhibited IL-4 and expressions of iNOS & COX-2 mRNA. In histological assessment, FF administration group, UC administration group and JGE administration group showed reduced amount of subepithelial edema, desquamated mucosa and hyperplasia of basement membrane from OVA.

Epigenetic Regulation in the Brain after Spinal Cord Injury : A Comparative Study

Objective : After spinal cord injury (SCI), functional and structural reorganization occurs at multiple levels of brain including motor cortex. However, the underlying mechanism still remains unclear. The current study was performed to investigate the alterations in the expression of the main regulators of neuronal development, survival and death, in the brain following thoracic contusive SCI in a mouse model. Methods : Eight-week-old female imprinting control region mice (n=60; 30-35 g) were used in this study. We analyzed the expression levels of regulators such as brain-derived neurotrophic factor (BDNF), glial cell line-derived neurotrophic factor (GDNF), nerve growth factor (NGF) and histone deacetylase (HDAC) 1 in the brain following thoracic contusive SCI. Results : The expression of BDNF levels were elevated significantly compared with control group at 2 weeks after injury (p<0.05). The expression of NGF levels were elevated at 2, 4 weeks compared with control group, but these difference were not significant (p>0.05). The GDNF levels were elevated at 2 week compared with control group, but these differences were not significant (p>0.05). The difference of HDAC1 levels were not significant at 2, 4 and 8 weeks compared with control group (p>0.05). Conclusion : These results demonstrate that the upregulation of BDNF may play on important role in brain reorganization after SCI.
