Objectives: To explore the correlation between multi-slice spiral CT (MSCT) perfusion parameters and the expression of vascular endothelial growth factor (VEGF) as well as matrix metalloproteinase-2 (MMP-2) in breast cancer. Methods: Forty five breast cancer patients and 16 patients with benign breast tumor, both confirmed by pathology examination, were enrolled. All underwent MSCT perfusion imaging to obtain perfusion maps and data for parameters including blood flow (BF), blood volume (BV) and permeability surface (PS). Cancer patients did not receive treatment prior to surgery. The expression of VEGF and MMP-2 were examined with both immunohistochemistry and Western blotting. Results: The levels of VEGF and MMP-2 by immunohistochemistry were significantly higher in the breast cancer group (P < 0.01) than the benign tumor group. Relative OD values from Western blotting were also higher in cancer cases (P < 0.05). Similarly, the mean MSCT perfusion parameters (BF, BV, PS) were significantly higher in the breast cancer group (P < 0.01), BF and BV positively correlating with VEGF expression (r = 0.878 and 0.809 respectively, P < 0.01); PS and VEGF and MMP-2 expression were also positively correlated (r= 0.860, 0.786 respectively, P < 0.01). Conclusion: There is a correlation between breast cancer MSCT perfusion parameters and VEGF andMMP-2 expression, which might be useful for detection of breast lesions, qualitative diagnosis of breast cancer, and evaluation of breast cancer treatment.

Nano-crystals embedded in glass always have interesting quantum effects, while nano-crystal patterns may have wide potential applications in preparing PIC, OEIC and other types of electronic and optoelectronic devices. A direct current electric field, accompanied by heat treatment, was applied on tellurite or silicate glass sheets. The silver ions in a diffused film were introduced into the glass matrix under a high-voltage electric field, instead of supplied during glass melting. The direct diffusion of metal ions, the nucleation and crystallization of nano-crystals in the glass were well controlled by the electric field accompanied by heat treatment. It was found that silver nano-crystal patterns can be transfer printed into tellurite and silicate glass sheets with both patterned diffusion and patterned crystallization. Twenty natural compounds isolated fromtraditional Chinese medicines were examined for theirantiviral activities against PCV2 in vitro. Antiviral activitywas analyzed by the assays of blocking of virus entry,inhibition of virus replication and inactivation of virusparticles in the virus suspension. 50% cytotoxic concentration(CC50) and 50% effective concentration (EC50) for eachcompound were determined by using the MTT method andimmunofluorescence assay (IFA), respectively. The resultsobtained from the inhibition of virus replication andinactivation of virus particles showed that the maximuminhibition rates for matrine and scutellarin were 57 and72.69% and their SI were 29.87 and 18.66, respectively. However, the results from the assays of inhibition of virusentry and inhibition of virus replication demonstrated thatboth aesculetin and arecoline hydrochloride stimulatedPCV2 replication, with the maximum stimulation rate of387.3 and 219.4%. In summary, both matrine and scutellarinhad antiviral activity against PCV2, while aesculetin andarecoline hydrochloride promoted PCV2 infection replicationin vitro.

MicroRNAs (MiRNAs) play important roles in coordinating a variety of cellular processes and abnormal expression has been linked to the occurrence of several cancers. The miRNA miR-451 is downregulated in colorectal carcinoma (CRC) cells, suggested by several research groups including our own. In this study, synthetic miR-451 mimics were transfected into the SW620 human CRC cell line using Lipofectamine 2000 and expression of miR-451 was analyzed by real time PCR, while expression of CAB39, LKB1, AMPK, AKT, PI3K and Bcl2 was analyzed by Western blot, and cell growth was detected by MTT assay. In comparison to the controls, a significant increase in the expression of miR-451 was associated with significantly decreased expression of CAB39, LKB1, AMPK, AKT, PI3K and Bcl2. The capacity of cell proliferation was significantly decreased by miR-451 expression, which also inhibited cell growth. Our study confirmed that miR-451 has a repressive role in CRC cells by inhibiting cell growth through down-regulating the P13K/AKT pathway.
Knockout of the EgriBLOS2 gene results in the transparent integuments of Ectropis grisescens larvae

The body colors of insects have evolved specialized roles in body protection, signaling and physiology. In some case, the larvae can camouflage their body colors to adapt the habitants and protect themselves. However, the genetic and molecular basis on larval body colors of the tea geometrid, Ectropis grisescens, remains poorly known. Here, we reported an effect of the lysosome-related organelles complex-1, subunit 2 (EgriBLOS2) gene knockout on larval integuments of E. grisescens, by using a clustered regularly interspaced short palindromic repeat (CRISPR)/CRISPR-associated protein 9 (Cas9) system. Combining transcriptomic data and PCR approaches, we first identified the EgriBLOS2 gene from E. grisescens, which encoded 137 amino acids and comprised 3 introns. In the phylogenetic analysis, EgriBLOS2 clustered in the Lepidoptera clade with high conservation to members in other lepidopteran species. Developmental expression profiles revealed that EgriBLOS2 was constantly tran scribed at each stage, in which its expression was significantly lower in 2nd instar larvae than that of other instars. After injecting EgriBLOS2-specific guide RNA and Cas9 messenger RNA into eggs, 70% of larvae showed the translucent integuments in G0 generation, with an emphasis on black splayed patterns in the 2nd and 8th segments of abdomens. However, some typical characteristics of larvae were not obviously changed in mutant instars, such as ocelli, mouthparts and other appendants. This study has unraveled the roles of EgriBLOS2 in the formation of larval integument colors, and provides an alternative strategy for pest management based on the colors in this species.
